Clostridium difficile virulence factors: Insights into an anaerobic spore-forming pathogen.

The worldwide emergence of epidemic strains of Clostridium difficile linked to increased disease severity and mortality has resulted in greater research efforts toward determining the virulence factors and pathogenesis mechanisms used by this organism to cause disease. C. difficile is an opportunist pathogen that employs many factors to infect and damage the host, often with devastating consequences. This review will focus on the role of the 2 major virulence factors, toxin A (TcdA) and toxin B (TcdB), as well as the role of other putative virulence factors, such as binary toxin, in C. diffici…

[Investigation of the virulence factors of multidrug-resistant Acinetobacter baumannii isolates].

Acinetobacter baumannii is an opportunistic human pathogen which causes life-threatening nosocomial infections such as ventilator-associated pneumonia, bacteremia, meningitis, urinary tract and wound infections. Treatment options are very limited for infections caused by multi-drug resistant (MDR) A.baumannii strains. Until recently, the majority of studies related to A.baumannii have focused on antibiotic resistance, treatment protocols and epidemiological data, however, there have been few studies addressing the virulence factors of this organism. The features such as biofilm formation, seru…

Variable virulence factors in Burkholderia pseudomallei (melioidosis) associated with human disease.

Burkholderia pseudomallei is a Gram-negative environmental bacterium that causes melioidosis, a potentially life-threatening infectious dise … ated with localized skin lesions without sepsis. We show, for the first time, that variably present virulence factors play an important role in the pathogenesis of melioidosis. Collectively, our study provides a framework for assessing other non-ubiquitous bacterial virulence factors and their association with disease, such as candidate loci identified from large-s …

[Analysis of molecular fingerprint and virulence factors of Helicobacter pylori strains].

BACKGROUND: Helicobacter pylori is now accepted as the most important agent of gastritis in humans, as well as a risk factor for peptic ulcer disease and gastric carcinoma. The outcome of the infection is related to several factors, among them bacterial ones such as cagA and vacA s1m1 genotype. Random amplified polymorphic DNA (RAPD)-PCR, has been used to generate DNA fingerprints to evaluate similarity among strains within a bacterial species. AIM: To assess the association between RAPD fingerprinting, virulence factors and the disease. METHODS: H. pylori was isolated from 112 patients (41 …

Virulence factors and resistance mechanisms of Serratia marcescens. A short review.

Serratia marcescens, a Gram-negative bacillus that belongs to the family Enterobacteriaceae, is a human opportunistic pathogen bacterium that causes many diseases, such as urinary tract infections, respiratory tract infections, bacteremia, conjunctivitis, endocarditis, meningitis and wound infections. Many plasmides that confers multi-drug resistance were discovered, such as virulence factors, like cytotoxins that damage epithelial cells. The main topic of this paper presents a review about the molecular traits evolved in the pathogenic processes mediated by Serratia and its mechanism of resis…

Virulence factors, pathogenesis and vaccine protection in cholera and ETEC diarrhea.

Recent work has provided new insights into the pathogenesis of the potentially life-threatening diarrheas caused by Vibrio cholerae and enterotoxigenic Escherichia coli (ETEC): a new mechanism (post-translational degradation), which is involved in the control of cholera toxin expression, has been discovered. Recent evidence also suggests that vibrios upregulate cholera toxin expression in response to intestinal fluid components, and enterotoxin-carrying bacterial outer membrane vesicles might have a function in ETEC pathogenesis. An important role of the environment is supported by the correla

Possible virulence factors of Staphylococcus aureus in a mouse septic model.

Twenty clinical isolates of Staphylococcus aureus were examined to elucidate the virulence factors which are directly related to lethality in a mouse septic model. Heat or formalin treatment of the organism abolished the lethal activity of the live organism during challenge intravenously administered via the tail vein. Nevertheless, injection of ten times concentrated culture supernatant fluid (SUP) showed lethal activity in the mouse. However, there was no lethality when SUP was heated at 60 degrees C for 15 min. To examine variations of SUP lethality among strains, we collected 20 strains of…

Dermonecrotic toxin and tracheal cytotoxin, putative virulence factors of Bordetella avium.

We examined Bordetella avium for virulence factors common to Bordetella pertussis, including pertussis toxin, filamentous hemagglutinin, adenylate cyclase, dermonecrotic toxin, and tracheal cytotoxin. B. avium produced a dermonecrotic toxin and a tracheal cytotoxin. The dermonecrotic toxin of B. avium is a 155,000-molecular-weight, heat-labile protein which was lethal for mice, guinea pigs, young chickens, and turkey poults and produced dermonecrosis when injected intradermally into guinea pigs, chickens, and turkey poults. High-pressure liquid chromatography of B. avium culture supernatant fl…

Role of cell-associated and secretion-independent virulence factors of Pseudomonas aeruginosa in microbial keratitis and anti-virulence therapeutic strategies.

PURPOSE: Microbial keratitis (MK) caused by Pseudomonas aeruginosa is a rapidly progressive and vision-threatening corneal infection worldwide. Even short delays in diagnosis or inappropriate therapy can lead to irreversible corneal scarring and blindness. This opportunistic pathogen colonizes, invades, and damages ocular tissue by taking advantage of disruptions in the corneal epithelial barrier and using a variety of virulence factors. The aim of this study was to summarize the current understanding of P. aeruginosa’s cell-associated, and secretion system-independent virulence factors involv…

A fadD mutant of Vibrio cholerae is impaired in the production of virulence factors and membrane localization of the virulence regulatory protein TcpP.

In the enteric pathogen Vibrio cholerae, expression of the major virulence factors is controlled by the hierarchical expression of several regulatory proteins comprising the ToxR regulon. In this study, we demonstrate that disruption of the fadD gene encoding a long-chain fatty acyl coenzyme A ligase has marked effects on expression of the ToxR virulence regulon, motility, and in vivo lethality of V. cholerae. In the V. cholerae fadD mutant, expression of the major virulence genes ctxAB and tcpA, encoding cholera toxin (CT), and the major subunit of the toxin-coregulated pilus (TCP) was drasti…

Assessment of hypermucoviscosity as a virulence factor for experimental Klebsiella pneumoniae infections: comparative virulence analysis with hypermucoviscosity-negative strain.

BACKGROUND: Klebsiella pneumoniae displaying the hypermucoviscosity (HV) phenotype are considered more virulent than HV-negative strains. Nevertheless, the emergence of tissue-abscesses-associated HV-negative isolates motivated us to re-evaluate the role of HV-phenotype. RESULTS: Instead of genetically manipulating the HV-phenotype of K. pneumoniae, we selected two clinically isolated K1 strains, 1112 (HV-positive) and 1084 (HV-negative), to avoid possible interference from defects in the capsule. These well-encapsulated strains with similar genetic backgrounds were used for comparative analy

Factors Governing the Cross-Species Virulence of Shiga Toxin-Producing Escherichia coli.

Phage-encoded Shiga toxin (Stx) released by Shiga toxin-producing E. coli (STEC) can kill multiple eukaryotic bacterial predators, including Acanthamoeba castellanii, Tetrahymena thermophila and Caenorhabditis elegans. However, the impact of Stx type, Stx amount, and the serogroup of the E. coli on the effectiveness of this exotoxin are poorly understood. These factors impact the severity of Stx-mediated disease in humans and therefore, by studying their role in modulating predator-prey interactions, we may gain insight into how these virulence factors evolved to contribute to human pathogenic…

Cellular and Humoral Immune Responses to Noncanonical Enterotoxigenic Escherichia coli Virulence Factors.

A broadly protective licensed vaccine against enterotoxigenic Escherichia coli (ETEC) remains unavailable. To inform rational ETEC vaccine design, we characterized immune responses to the noncanonical antigens EatA mucinase, EtpA adhesin, and YghJ metalloprotease following experimental ETEC infection. We analyzed longitudinal cellular and humoral responses to EatA, EtpA, and YghJ in 30 volunteers infected with ETEC. Antigen-specific CD4+ T cell responses were assessed using an activation-induced marker (AIM) assay detecting CD69, CD134, CD137, and CD154 expression after antigen stimulation. An

Changes in nomenclature, virulence factors, and antifungal resistance of the genus Candida.

Some Candida species of clinical interest have undergone recent nomenclature changes. These yeasts have a high capacity to adhere to and infect host tissues, driven by their virulence factors, as well as by the incidence of antifungal resistance. This review aimed to analyze the taxonomic changes of the main species of clinical interest within the Candida genus, considering the clinical implications of their virulence factors and the main mechanisms of antifungal resistance. The research results allowed us to understand that the updated nomenclature of Candida species is essential to maintain …

Virulence factors of Salmonella Typhi: interplay between the bacteria and host macrophages.

Salmonella Typhi (S. Typhi) is a Gram-negative bacterium that exclusively infects humans and causes typhoid fever- a major global public health concern responsible for approximately 9 million infections and 110,000 deaths annually. Macrophages, a key component of the innate immune system, play essential roles in pathogen clearance, antigen presentation, immune regulation, and tissue repair. As one of the primary targets of S. Typhi infection, macrophages significantly influence disease onset and progression. S. Typhi expresses a range of virulence factors, including the virulence-associated (V…

Entamoeba histolytica: Plasma membrane components and virulence factors in the invasive process.

Entamoeba histolytica, the etiological agent of amoebiasis, is a parasitic protozoan responsible for severe gastrointestinal complications, including colitis and hepatic abscesses. The parasite primarily colonizes the human large intestine, where the active stages confront a hostile environment. Specialized adaptations in the plasma membrane of E. histolytica are critical for survive in this niche and enable the parasite to persist harmlessly in asymptomatic infection or initiate pathogenic interactions that lead to tissue invasion. Early events in pathogenesis are the breaching of the intesti
