The specification and growth factor inducibility of the pronephric glomus in Xenopus laevis.

We report a study on the specification of the glomus, the filtration device of the amphibian pronephric kidney, using an explant culturing strategy in Xenopus laevis. Explants of presumptive pronephric mesoderm were dissected from embryos of mid-gastrula to swimming tadpole stages. These explants were cultured within ectodermal wraps and analysed by RT-PCR for the presence of the Wilm's Tumour-1 gene, xWT1, a marker specific for the glomus at the stages analysed, together with other mesodermal markers. We show that the glomus is specified at stage 12.5, the same stage at which pronephric tubul…

Duplication and MHC linkage of the CTX family of genes in Xenopus and in mammals.

The effects of whole genome duplications that characterize the evolution of vertebrates have been studied on the gene of the Xenopus thymocyte molecule CTX and its mammalian relatives. CTX, with an extracellular part consisting of one V and one C2 external domain, defines a new subset of the immunoglobulin superfamily and is conserved from amphibians to mammals. The number of CTX loci, their polymorphism, and their genetic linkages have been studied in several Xenopus species and in humans. In the genetically simplest species, X. tropicalis (2n = 20), the unique CTX locus is linked to the MHC.…

LiCl-induced malformations of the eyes and the rostral CNS in Xenopus laevis.

Ocular malformations such as synophthalmia and cyclopia occur in all vertebrates including humans. We induced fused eye anlagen by exposure of Xenopus laevis embryos to dissolved LiCl. Three basic forms of malformations were observed in the rostral CNS; these represent major steps in a continuous sequence of ocular abnormalities: (i) "hour-glass eyes", (ii) synophthalmia, and (iii) cyclopia vera. The type of abnormity induced seemed to depend on the time and/or the dosage of LiCl exposure. In all cases studied, the histology of the retinae was normal. We reconstructed olfactory organs, eyes, a…

Ontogeny of the alloimmune response against a transplanted tumor in Xenopus laevis.

Xenopus laevis lymphoid tumor cells of the ff genotype grow after transplantation in inbred ff tadpoles or young post-metamorphic animals, but do not grow in fully grown ff adults. The ability to grow is lost progressively after metamorphosis and is apparently due to an immune response of the adult host against minor histocompatibility antigens (non-MHC encoded) expressed by the tumor cells. The difference in alloimmune responses between the larval and the adult immune system of the amphibian Xenopus has been subsequently investigated with this new in vivo model. The resistance of the host aga…

Effects of alcohols on responses evoked by inositol trisphosphate in Xenopus oocytes.

1. The effects of ethanol and other alcohols on inositol 1,4,5-trisphosphate (InsP3) signalling were studied in Xenopus oocytes by the use of flash photolysis of caged InsP3. Calcium liberation induced by InsP3 was monitored by voltage-clamp recording of Ca(2+)-activated membrane currents, and by fluorescence of the Ca2+ indicator Fluo-3. 2. Membrane current and fluorescence Ca2+ signals evoked by light flashes giving small responses were initially potentiated by bath application of ethanol (80-400 mM). However, the responses subsequently declined while ethanol was present and were strongly re…

Biosynthesis and secretion of rat salivary proteins by Xenopus laevis oocytes.

Xenopus laevis oocytes injected with poly (A+) RNA isolated from rat parotid and submandibular glands synthesize and secrete salivary proteins. Amylase was identified in the media of cultured oocytes injected with rat parotid mRNA by size and immunoprecipitation with anti-human amylase serum. Secretion of the salivary proteins was detectable in the medium eight h following the parotid mRNA injection and continued in a time-dependent fashion for up to 96 h. In contrast to rat parotid slices in culture, which demonstrate a regulated pathway of secretion highly responsive to the secretagogue isop…

The synthesis and expression of HLA-A and -B antigens in Xenopus laevis oocytes.

Purified mRNA's encoding the HLA-A and -B antigen heavy chains or beta 2-microglobulin were prepared from human B lymphoid cells by positive hybridization selection procedures. The role of chain association in the biosynthesis and intracellular transport of HLA-A and -B antigens was investigated by injecting these mRNA species into Xenopus laevis oocytes and following the fates of the translated products by immunoprecipitation. When mRNA encoding beta 2-microglobulin from the B lymphoblastoid cell line MST was coinjected with mRNA encoding the HLA-A and -B antigen heavy chains from the Burkitt…

Further studies on the melanophores of periodic albino mutant of Xenopus laevis.

It is still unknown why dermal melanophores disappear during larval development, and why no or very few epidermal melanophores appear during and after metamorphosis, in Xenopus laevis showing periodic albinism (ap). To elucidate these points, we investigated the occurrence of depigmentation in mutant (ap/ap) melanophores during in vitro proliferation and the incidence of melanophore differentiation from mutant melanoblasts in the skin in vitro. During in vitro proliferation of mutant melanophores, ap-type melanosomes decreased in number gradually and instead the number of premelanosomes increa…

An analysis of pigment cell development in the periodic albino mutant of Xenopus.

The periodic al,bino mutant (apap) of Xenopus in which the development of melanophores is impaired, is further reported here to possess an aberrant pattern of iridophore differentiation. The development of mutant and wild-type neural crest explants isolated in vesicles derived from tissues from identical and different genotypes was examined to determine if the mutant effect resides in the pigment cells or is mediated by the environmental tissues. Mutant melanophores and iridophores cultured in either mutant or wild-type tissues exhibited mutant patterns of differentiation. Wild-type pigment ce…

In Vivo Wide-Field Fluorescence Ca2+ Imaging of Xenopus Tadpole Brains.

In vivo wide-field calcium ion (Ca2+) imaging enables visualization of neuronal activity across large brain regions in living Xenopus tadpoles. This approach provides a cost-effective and accessible alternative to two-photon microscopy for capturing population-level Ca2+ dynamics. The protocol outlines preparation of tadpoles expressing the genetically encoded Ca2+ indicator GCaMP6s, followed by cranial-window dissection, immobilization in low-melting-point agarose, and wide-field fluorescence imaging under low-intensity 480 nm excitation. Recordings are acquired at two frames per second for 3…

Foxj3 represses TGF-β signals-responsive transcriptions in Xenopus early development.

BACKGROUND: Forkhead box (Fox) proteins belong to a large family of transcription factors characterized by a highly conserved DNA binding domain termed the 'forkhead box'. They are involved in critical functions during early development and in the adult. OBJECTIVE: In this work, Foxj3 was found to regulate negatively the transcription of target genes induced by Activin/Nodal or BMP signal in Xenopus embryos. Thus, we sought to investigate the molecular mechanisms underlying this function of Foxj3 as a transcriptional repressor. METHODS: Microinjection of mRNA and anti-sense morpholino oligo …

Protocadherin 19 regulates axon guidance in the developing Xenopus retinotectal pathway.

Protocadherin 19 (Pcdh19) is a homophilic cell adhesion molecule and is involved in a variety of neuronal functions. Here, we tested whether Pcdh19 has a regulatory role in axon guidance using the developing Xenopus retinotectal system. We performed targeted microinjections of a translation blocking antisense morpholino oligonucleotide to knock down the expression of Pcdh19 selectively in the central nervous system. Knocking down Pcdh19 expression resulted in navigational errors of retinal ganglion cell (RGC) axons specifically at the optic chiasm. Instead of projecting to the contralateral op…

TLR5-Mediated Reactivation of Quiescent Ranavirus FV3 in Xenopus Peritoneal Macrophages.

Ranaviruses such as frog virus 3 (FV3) are large double-stranded DNA (dsDNA) viruses causing emerging infectious diseases leading to extensive morbidity and mortality of amphibians and other ectothermic vertebrates worldwide. Among the hosts of FV3, some are highly susceptible, whereas others are resistant and asymptomatic carriers that can take part in disseminating the infectious virus. To date, the mechanisms involved in the processes of FV3 viral persistence associated with subclinical infection transitioning to lethal outbreaks remain unknown. Investigation in Xenopus laevis has revealed …

Lysine demethylase 3a in craniofacial and neural development during Xenopus embryogenesis.

Epigenetic modifier lysine demethylase 3a (Kdm3a) specifically demethylates mono‑ and di‑methylated ninth lysine of histone 3 and belongs to the Jumonji domain‑containing group of demethylases. Kdm3a serves roles during various biological and pathophysiological processes, including spermatogenesis and metabolism, determination of sex, androgen receptor‑mediated transcription and embryonic carcinoma cell differentiation. In the present study, physiological functions of Kdm3a were evaluated during embryogenesis of Xenopus laevis. Spatiotemporal expression pattern indicated that kdm3a exhibited i…

Highly efficient bi-allelic mutation rates using TALENs in Xenopus tropicalis.

In the past decade, Xenopus tropicalis has emerged as a powerful new amphibian genetic model system, which offers all of the experimental advantages of its larger cousin, Xenopus laevis. Here we investigated the efficiency of transcription activator-like effector nucleases (TALENs) for generating targeted mutations in endogenous genes in X. tropicalis. For our analysis we targeted the tyrosinase (oculocutaneous albinism IA) (tyr) gene, which is required for the production of skin pigments, such as melanin. We injected mRNA encoding TALENs targeting the first exon of the tyr gene into two-cell-…

Aminoglycoside block of P2X2 receptors heterologously expressed in Xenopus laevis oocytes.

Aminoglycosides are polycationic antibiotics that have been shown to block a variety of cation channels. The inhibitory effect of externally applied aminoglycosides on P2X2 receptor currents was examined after heterologous expression in Xenopus laevis oocytes using the two-electrode voltage-clamp technique. All of the aminoglycosides tested inhibited the ATP-evoked responses with potencies ranging from 71 μM to 2 mM (IC(50) values). The ranked order of potency was streptomycin > gentamicin > neomycin > paromomycin > kanamycin. The inhibition of P2X receptor currents was independent of the ATP …

Psf2 plays important roles in normal eye development in Xenopus laevis.

PURPOSE: Psf2 (partner of Sld5 2) represents a member of the GINS (go, ichi, ni, san) heterotetramer [1] and functions in DNA replication as a "sliding clamp." Previous in situ hybridization analyses revealed that Psf2 is expressed during embryonic development in a tissue-specific manner, including the optic cup (retina) and the lens [2]. This article provides an analysis of Psf2 function during eye development in Xenopus laevis. METHODS: A morpholino targeted to Psf2 mRNA was designed to knockdown Psf2 translation and was injected into specific embryonic cells during early cleavage stages in…

Apoptosis-specific protein (ASP) identified in apoptotic Xenopus thymus tumor cells.

A novel apoptosis-specific protein (ASP) has recently been identified in the cytoplasm of apoptotic mammalian cells. This paper investigates whether ASP is found in Xenopus thymus tumor-derived lymphoid cell lines undergoing apoptosis and also in apoptotic, nontransformed splenocytes. Cultured Xenopus tumor lymphoid cells induced to undergo apoptosis by serum deprivation or treatment with the calcium ionophore, ionomycin, displayed altered morphology typical of apoptotic cells, as judged by flow cytometric light-scatter characteristics and by fluorescence microscopy of acridine-orange-stained …
