A study of tin dioxide and antimony tetroxide supported vanadium oxide catalysts by solid-state 51V and 1H NMR techniques.

A series of vanadia catalysts with various V2O5 loadings supported on SnO2 and alpha-Sb2O4 are investigated by the application of X-ray diffraction and solid-state 51V and 1H NMR techniques. XRD results show no evidence for the formation of a crystalline vanadia phase on both supports. However, the 51V NMR spectra of the catalysts reveal the existence of two types of vanadia species on the surface of the support: one due to a dispersed vanadia phase at lower vanadia loadings and the other due to a crystalline vanadia phase at higher vanadium content. The quantity of the dispersed vanadia phase

Generation of human induced pluripotent stem cell (hiPSC) lines derived from three patients carrying the pathogenic CRYAB (A527G) mutation and one non-carrier family member.

A newly identified pathogenic variant (A527G) in alpha B-crystallin (αB-crystallin) has been linked to congenital cataract and young-onset dilated cardiomyopathy (DCM) within a Dutch family, although the disease mechanism remains unclear. Four human induced pluripotent stem cell (hiPSC) clones were generated from three symptomatic patients carrying the A527G variant, and one healthy proband. Peripheral blood mononuclear cells (PBMCs) were reprogrammed using integration-free Sendai viral pluripotency vectors. The established hiPSCs clones exhibited regular ESC-like morphology, expression of plu

Lens-specific expression of a major histocompatibility complex class I molecule disrupts normal lens development and induces cataracts in transgenic mice.

PURPOSE: Lens epithelial tissue does not normally express major histocompatibility complex (MHC) class I molecules. In addition, the mechanism of self-tolerance to intraocular antigens is unknown. To study the effect of class I expression in the lens, transgenic mice were produced that express an allo-MHC class I molecule under the alpha A-crystallin proximal promoter. METHODS: p alpha Dd was generated by fusion of the H-2Dd structural gene to the alpha A-crystallin proximal promoter. Transgenic mice were produced, and founder lines were identified by Southern blot hybridization. Eyes from tr

[Fibrogenic effect of anthracite dust compared with the effects of hard and bituminous coal dust on the lungs of albino rats].

In experiments on white rats the fibrogenic effects of the dust of anthracite and other types of coal, from the U.S.A. and Poland, were tested. What was found basing on hydroxyproline measurements in lungs was that the extent of fibrogenic effects of anthracite equals fibrogenic effects of bituminous coal dust containing 30% alpha-quartz. Non-anthracite coal dusts containing SiO2 crystalline phases (alpha-quartz) as an admixture--exhibit fibrogenic effects on white rats' lungs--proportional to free silica content. It is probable that fibrogenic effects of anthracite on the pulmonary tissue res

Unique contribution of heat shock transcription factor 4 in ocular lens development and fiber cell differentiation.

Mammalian ocular lens development results via a differentiation program that is highly regulated by tissue-specific transcription factors. Central to this is the terminal differentiation of fiber cells, which develop from epithelial cells on the anterior surface of the lens, accompanied by a change in cell shape and expression of structural proteins (such as membrane proteins MP19, MIP26, connexin 43, 46, and 50, cytoskeletal proteins CP49, CP115, and alpha, beta, and gamma crystallins), creating a transparent, refractive index gradient in the lens. Mutations in genes controlling eye developme

Transgenic studies on the role of optineurin in the mouse eye.

Mutations in the OPTN gene encoding for optineurin have been associated with primary open-angle glaucoma. The functional role(s) of optineurin in the normal and glaucomatous eye are unclear. As optineurin interferes with TNF-alpha mediated cell death in vitro, an involvement of optineurin in the regulatory pathways leading to apoptosis of retinal ganglion cells has been suggested. The goal of the present study was to study the molecular properties of optineurin and its capabilities to prevent apoptosis in vivo in the eyes of transgenic mice. The chicken betaB1-crystallin promoter was used to o

Identification of a novel oligomerization disrupting mutation in CRYΑA associated with congenital cataract in a South Australian family.

Congenital cataract is a heterogeneous disorder causing severe visual impairment in affected children. We screened four South Australian families with autosomal dominant congenital cataract for mutations in 10 crystallin genes known to cause congenital cataract. We identified a novel segregating heterozygous mutation, c.62G>A (p.R21Q), in the CRYΑA gene in one family. Western blotting of proteins freshly extracted from cataractous lens material of the proband demonstrated a marked reduction in the amount of the high-molecular-weight oligomers seen in the lens material of an unaffected individu

Detection and prevention of ocular phototoxicity of ciprofloxacin and other fluoroquinolone antibiotics.

Fluoroquinolone (FLQ) drugs are a potent family of antibiotics used to treat infections including ocular infections. To determine if these antibiotics may be phototoxic to the eye, we exposed human lens epithelial cells to 0.125-1 mm FLQs (ciprofloxacin [Cipro], lomefloxacin [Lome], norfloxacin [Nor] and ofloxacin [Ofl]), the precursor quinolone nalidixic acid (Nalid) and UVA radiation (2.5 J cm(-2)). Based on fluorescence confocal microscopy, FLQs are diffused throughout the cytoplasm and preferentially located in the lysosomes of lens epithelial cells. Neither FLQ exposure alone nor UVA expo

Kinetics and energetics of oligomer desorption from surfaces.

The dynamics of oligomer desorption from surfaces has been studied by measuring the desorption kinetics of a set of straight chain alkanes [ H(CH2)(n)H, with n = 5 to 60] from the surface of single crystalline graphite. Desorption is observed to be a first-order process and the preexponent of the desorption rate constant has a value nu = 10(19.6+/-0.5) sec(-1) and is independent of the oligomer chain length. More interestingly, we find that the barrier to desorption has a nonlinear dependence on chain length and takes the form DeltaE(double dagger)(des) = A+Bn(alpha), with the exponent alpha =

Age-Related Lens Membrane Composition and N101D Deamidation Modulate αA-Crystallin Membrane Interactions Relevant to Cataract.

PURPOSE: αA-crystallin (αAc), the predominant subunit comprising α-crystallin (αABc), becomes increasingly deamidated and membrane bound with cataract formation. Furthermore, the lens lipids and cholesterol (Chol) composition significantly change with age and cataract formation. Despite this, the relationship between αAc deamidation and lens membrane composition in cataractogenesis remains unclear. To address this, we analyzed the role of N101 deamidation and membrane composition in the membrane interactions of αAc. METHODS: Experimentation involved using electron paramagnetic resonance (EPR)

Unveiling the structural and functional consequences of the p.D109G pathogenic mutation in human αB-Crystallin responsible for restrictive cardiomyopathy and skeletal myopathy.

αB-Crystallin (αB-Cry) is expressed in many tissues, and mutations in this protein are linked to various diseases, including cataracts, Alzheimer's disease, Parkinson's disease, and several types of myopathies and cardiomyopathies. The p.D109G mutation, which substitutes a conserved aspartate residue involved in the interchain salt bridges, with glycine leads to the development of both restrictive cardiomyopathy (RCM) and skeletal myopathy. In this study, we generated this mutation in the α-Cry domain (ACD) which is crucial for forming the active chaperone dimeric state, using site-directed mu

Glycation and insolubility of human lens protein.

To learn whether glycation plays a role in insolubilization or in senile cataractogenesis, the reactivity of lens protein from normal and senile cataractous lenses and individual crystallin prepared from human lens with various sugars [glucose, glucose-1-phosphate (G-1-P), glucose-6-phosphate (G-6-P) and fructose], and the insolubility of those proteins were determined. The reactivity of human lens protein to glucose was increased in a dose-dependent manner, and it was demonstrated that 17.9, 18.5 and 24 kDa proteins were susceptible to glycation with sugars. The study also showed that alpha-,

Alteration of protein-protein interactions of congenital cataract crystallin mutants.

PURPOSE: A recent study demonstrated the presence of protein-protein interactions among lens crystallins in a mammalian cell two-hybrid system assay and speculated about the significance of these interactions for protein solubility and lens transparency. The current study extends those findings to the following crystallin genes involved in some congenital cataracts: CRYAA (R116C), CRYAB (R120G), and CRYGC (T5P). METHODS: A mammalian two-hybrid system was used to assay the protein-protein interactions. Congenital cataract crystallin genes were cloned and fused into the two-hybrid system vector

Inactivation of Smad4 leads to impaired ocular development and cataract formation.

PURPOSE: Signaling by members of the TGFβ superfamily of molecules is essential for embryonic development and homeostasis. Smad4, a key intracellular mediator in TGFβ signaling, forms transcriptional activator complexes with Activin-, BMP-, and TGFβ-restricted Smad proteins. However, the functional role of Smad4 in controlling different visual system compartments has not been fully investigated. METHODS: Using the Pax6 promoter-driven Cre transgenic, smad4 was conditionally inactivated in the lens, cornea and ectoderm of the eyelids. Lateral diffusion of small solutes and partition of amphipaths in defect structures of lipid bilayers.

The excimer formation technique has been applied to investigate the mechanism of the lateral diffusion in the crystalline P beta' phase of dipalmitoylphosphatidylcholine vesicles. This became possible at low pyrene concentrations. From the shape of phase-transition curves, from the effect of pressure up to 150 bar and from the perturbation induced by cholesterol, we conclude that the free volume model could also be applied to the P beta' phase. However, the diffusion is thought to occur in defect structures that are considered to form fluid pathways between domains of crystalline lipid. Partit

Sugar acetates as CO2-philes: molecular interactions and structure aspects from absorption measurement using quartz crystal microbalance.

Sugar acetates, recognized as attractive CO(2)-philic compounds, have potential uses as pharmaceutical excipients, controlled release agents, and surfactants for microemulsion systems in CO(2)-based processes. This study focuses on the quantitative examination of absorption of high pressure CO(2) into these sugar derivatives using quartz crystal microbalance (QCM) as a detector. In addition to the absorption measurement, the QCM is initially found to be able to detect the CO(2)-induced deliquescence of sugar acetates, and the CO(2) pressure at which the deliquescence happens depends on several

Cataract-causing defect of a mutant γ-crystallin proceeds through an aggregation pathway which bypasses recognition by the α-crystallin chaperone.

BACKGROUND: The transparency of the eye lens depends upon maintenance of the native state of the γ- and β-crystallins, which is aided by the abundant chaperones αA- and αB-crystallin. Mature onset cataract, the leading cause of blindness worldwide, involves the polymerization of covalently damaged or partially unfolded crystallins into light-scattering aggregates. A number of single amino acid substitutions and truncations of γ-crystallins result in congenital cataract in both humans and mice, though in many cases the coupling between the protein alterations and the accumulation of aggregates

Follicular Fluid Meiosis-Activating Sterol (FF-MAS) Enhances Lens Transparency By Increasing αA-Crystallin Solubility In Cataracts.

Insolubility and aggregation of crystallins are common features in cataracts. CYP51 has been identified as a novel candidate gene for cataracts; however, its associated phenotypes remain unconfirmed. We investigated the role of FF-MAS, synthesized by CYP51, in αA-crystallin solubility and lens transparency. FF-MAS was administered to HLE-B3 cells expressing αA-crystallin (R116C) and to rats with selenite-induced cataracts. Lens transparency was assessed using slit-lamp photography. Congo red staining and transmission electron microscopy were used to analyze lens uniformity. Western blotting an
