The opioid peptide beta h-endorphin 1-31 inhibited the plaque-forming cell (PFC) response to sheep red blood cells when rat splenocytes, immunized in vivo, were cultured in vitro with the peptide. The opioid antagonist naloxone failed to reverse the beta h-endorphin 1-31 suppression of the PFC response. Peptide fragments of beta h-endorphin 1-31, other opioid peptides, and alkaloids had no effect on the response. These data indicate that beta h-endorphin 1-31 suppresses antibody production or secretion by a specific, non-opioid receptor on the rat splenocytes.

beta-Endorphin- and ACTH-like immunoreactive peptides were measured in plasma and certain tissues of a patient suffering from an oat-cell-carcinoma of the lungs and Cushing's syndrome. Using sensitive radioimmunoassays in combination with gel-filtration, highly elevated levels of beta-endorphin, beta-lipotropin (beta-LPH), alpha-melanotropin (alpha-MSH) and ACTH were found.

Rats were taught to self-administer beta-endorphin (0.1 microgram or 0.5 microgram/microliter) or NaCl 9% by pressing a lever that activated a pump connected with a cannula implanted in the lateral cerebral ventricle (icv). A reversed-phase high-performance liquid chromatography procedure combined with radioimmunoassay (rp-HPLC-RIA) is presented. It makes possible the quantitative determination of the neuropeptides, beta h-endorphin and substance P, in small volumes of cerebrospinal fluid (CSF). The concentration-dependence of the recovery of the method is demonstrated and the effect of free silanol groups of the stationary phase on the recovery is discussed. CSF was shown to contain 9.6 +/- 1.1 fmol beta h-endorphin and 12.0 +/- 0.9 fmol substance P per ml.
