Melatonin enhances adult rat hippocampal progenitor cell proliferation via ERK signaling pathway through melatonin receptor.

Melatonin, a neurohormone secreted mainly by the pineal gland, has a variety of physiological functions and neuroprotective effects. Previous studies have shown that melatonin could stimulate the proliferation of neural stem/progenitor cells (NS/PCs). Recent studies reported that the activities of mitogen-activated protein kinase (MAPK) of neural stem cells (NSCs) changed in response to the proliferative effect of melatonin. Therefore, the aim of the present study was to explore the proliferative mechanism mediated by melatonin on the adult rat hippocampal NS/PCs. Treatment with melatonin sign

Dermatologic Toxicities Associated With Novel Pan-RAS/RAF Inhibitors.

BACKGROUND: The mitogen-activated protein kinase (MAPK) pathway plays a key role in cell-cycle regulation and tumor progression in cancer. Dermatologic toxicities (DTs) to newer pan-RAS (RMC-6236) and pan-RAF (LXH254/naporafenib) inhibitors are emerging. Therefore, knowledge of the morphologic patterns of DTs from pan-RAS/RAF inhibitors will enable quick diagnosis and appropriate management. METHODS: The clinical and histologic features of patients treated with pan-RAS or pan-RAF inhibitors for advanced stage RAS mutated malignancies were retrospectively reviewed. RESULTS: Associated DTs wer

Ganglioglioma with MAP2K1 Mutation and CDKN2A/B Homozygous Deletion: A Case Report.

Aims/Background Gangliogliomas are grade 1 glioneuronal tumors occurring predominantly in the temporal lobe, as per the World Health Organization (WHO) classification. Gangliogliomas often harbor BRAF (v-Raf murine sarcoma viral oncogene homolog B1) p.V600E hotspot mutation or other alterations leading to activation of RAS/RAF/MAPK (rat sarcoma virus oncogene/rapidly accelerated fibrosarcoma/mitogen-activated protein kinase) signaling pathway, which is the driver factor of this tumor. This study aims to investigate a case of ganglioglioma patient with distinctive molecular features, and to pre

Modulation of KSR activity in Caenorhabditis elegans by Zn ions, PAR-1 kinase and PP2A phosphatase.

Vulval differentiation in Caenorhabditis elegans is controlled by a conserved signal transduction pathway mediated by Ras and a kinase cascade that includes Raf, Mek and MAPK. Activation of this cascade is positively regulated by a number of proteins such as KSR (kinase suppressor of Ras), SUR-8/SOC-2, SUR-6/PP2A-B and CDF-1. We describe the functional characterization of sur-7 and several genes that regulate signaling downstream of ras. We identified sur-7 by isolating a mutation that suppresses an activated ras allele, and showed that SUR-7 is a divergent member of the cation diffusion facil

Raf-MEK-Erk cascade in anoikis is controlled by Rac1 and Cdc42 via Akt.

Signals from the extracellular matrix are essential for the survival of many cell types. Dominant-negative mutants of two members of Rho family GTPases, Rac1 and Cdc42, mimic the loss of anchorage in primary mouse fibroblasts and are potent inducers of apoptosis. This pathway of cell death requires the activation of both the p53 tumor suppressor and the extracellular signal-regulated mitogen-activated protein kinases (Erks). Here we characterize the proapoptotic Erk signal and show that it differs from the classically observed survival-promoting one by the intensity of the kinase activation. T

GDP-Loaded K-Ras Transiently Binds to Effector B-Raf RBD, Mirroring the Structure of the Active GTP-Loaded Complex.

The Ras-Raf protein association initiates the downstream mitogen-activated protein kinase (MAPK) signaling cascade. This interaction depends on the Ras-nucleotide ligand, whereby Raf preferentially binds to the active guanosine triphosphate (GTP)-loaded state over the guanosine diphosphate (GDP)-loaded state of Ras. Whether GDP-bound Ras can also form specific protein-protein interactions with Raf, however, remains unclear. Here, we characterize the interaction between human K-Ras in both nucleotide states and the Ras-binding domain (RBD) of B-Raf by solution NMR. Active K-Ras·GTP forms a tigh

BCR-ABL- and Ras-independent activation of Raf as a novel mechanism of Imatinib resistance in CML.

Although the BCR-ABL tyrosine kinase inhibitor Imatinib has undoubtedly revolutionized the therapy of chronic myeloid leukaemia (CML), acquired drug resistance remains a common problem in CML therapy. Resistance often arises from second-line mutations in BCR-ABL or overexpression of the BCR-ABL protein but in ~20% of CML cases resistance mechanisms do not involve altered BCR-ABL function. Imatinib-resistant CML cell lines have been widely used for comparative proteome/genome-wide expression screens in order to decipher resistance mechanisms but a clearcut molecular mechanism or molecular playe

Langerhans cell histiocytosis: molecular mechanisms underlying pathogenesis and emerging targeted therapeutics.

Langerhans cell histiocytosis (LCH) is a nonhereditary myeloid neoplasm driven by somatic alterations in the mitogen-activated protein kinase (MAPK) signaling pathway, most commonly involving BRAF and MAP2K1 (MEK1). Oncogenic BRAF alterations activate MAPK signaling through diverse structural mechanisms. The prominent BRAF(V600E) mutation traps BRAF in a constitutively active status by forming the K507-E600 salt bridge independent of upstream RAS signaling. In contrast, non-V600E BRAF mutants exhibit variable kinase activity, among which kinase-dead variants rely on BRAF/CRAF heterodimerizatio

GILZ mediates the antiproliferative activity of glucocorticoids by negative regulation of Ras signaling.

Tsc22d3 coding for glucocorticoid-induced leucine zipper (GILZ) was initially identified as a dexamethasone-responsive gene involved in the control of T lymphocyte activation and apoptosis. However, the physiological role of this molecule and its function in the biological activity of glucocorticoids (GCs) has not been clarified. Here, we demonstrate that GILZ interacts directly with Ras in vitro and in vivo as shown by GILZ and Ras coimmunoprecipitation and colocalization upon PMA activation in primary mouse spleen T lymphocytes and thymus cells. The analysis of GILZ mutants showed that they

BH4 Oxidation-Derived H2O2 Activates ERK1/2 Signaling via B-Raf in Rat Dorsal Root Ganglion Neurons.

Elevated Tetrahydrobiopterin (BH4) levels are linked to various pain conditions. Pharmacological or genetic reduction of BH4 levels has analgesic effects in rodent models of neuropathic and inflammatory pain, but also affects neurological and cardiovascular functions. Little is known about the downstream mechanisms of BH4 that could be targeted to attenuate BH4-induced pain hypersensitivity without lowering BH4 levels. In this study, we exposed ex vivo-cultured rat dorsal root ganglion (DRG) neurons to BH4 and analyzed the activity of the sensory neuron-sensitizing kinase ERK1/2 via high-conte

Identification of a novel Raf-1 pathway activator that inhibits gastrointestinal carcinoid cell growth.

Carcinoids are neuroendocrine tumors (NET) that secrete hormones, including serotonin, resulting in the malignant carcinoid syndrome. In addition to the significant morbidity associated with the syndrome, carcinoids are frequently metastatic at diagnosis, and untreated mortality at 5 years exceeds 70%. Surgery is the only curative option, and the need for other therapies is clear. We have previously shown that activation of Raf-1 inhibits carcinoid cell proliferation. We investigated the ability of leflunomide (LFN), a Food and Drug Administration-approved medication for the treatment of rheum

Intrathecal delivery of farnesyl thiosalicylic acid and GW 5074 attenuates hyperalgesia and allodynia in chronic constriction injury-induced neuropathic pain in rats.

The role of mitogen-activated protein kinase (MAPK) family has been well defined in neuropathic pain. Ras and c-Raf constitute an important part of MAP kinase family as Ras/Raf/MEK/ERK2 signaling cascade. The present study was designed to investigate the analgesic potential of farnesyl thiosalicylic acid, a novel Ras inhibitor, and GW 5074, a selective c-Raf1 inhibitor, in chronic constriction-induced injury (CCI)-induced peripheral neuropathic pain. Neuropathic pain was induced by placing four loose ligatures around the sciatic nerve. The development of pain was assessed on 14th day in terms

RALGEF inhibitors suppress RAS-driven pancreatic cancer and metastasis.

RAS oncoproteins are the most frequently activated oncoproteins in cancer. Development of direct RAS inhibitors has proved technically challenging and has had limited success in the clinic. Those RAS inhibitors that have been approved tend to suffer from resistance development. Consequently, many attempts have focused on inhibiting RAS indirectly by targeting its immediate downstream effectors. RAS binds and activates three main effector classes to drive transformation: RAF kinases, phosphoinositide 3 (PI-3) kinase and Ras-like (RAL) small GTPases (RALGEF) exchange factors. Multiple FDA-approv…

Regulation of cardiac hypertrophic signaling by prolyl isomerase Pin1.

RATIONALE: Cardiac hypertrophy results from the complex interplay of differentially regulated cascades based on the phosphorylation status of involved signaling molecules. Although numerous critical regulatory kinases and phosphatases have been identified in the myocardium, the intracellular mechanism for temporal regulation of signaling duration and intensity remains obscure. In the nonmyocyte context, control of folding, activity, and stability of proteins is mediated by the prolyl isomerase Pin1, but the role of Pin1 in the heart is unknown. OBJECTIVE: To establish the role of Pin1 in the

[Sorafenib and radiotherapy association for hepatocellular carcinoma].

Conformal radiotherapy is a promising therapeutic strategy for hepatocellular carcinoma (HCC), producing local control rates above 90% within the radiation beam. However, survival after radiotherapy remains limited by the high frequency of intra- and extra-hepatic recurrences, which occurs in 40-50 and 20-30% of cases, respectively. Interaction of fibroblast growth factor and C-natriuretic peptide signaling in regulation of chondrocyte proliferation and extracellular matrix homeostasis.

Overexpression of C-natriuretic peptide (CNP) in cartilage partially rescues achondroplasia in the mouse. Here, we studied the interaction of fibroblast growth factor (FGF) and CNP signaling in chondrocytes. CNP antagonized FGF2-induced growth arrest of rat chondrosarcoma (RCS) chondrocytes by inhibition of the Erk mitogen activated protein kinase pathway. This effect of CNP was protein kinase G-dependent and was mimicked by the cGMP analog pCPT-cGMP. FGF2-mediated activation of both MEK and Raf-1 but not Ras or FRS2 was abolished by CNP demonstrating that CNP blocks the Erk pathway at the lev
