Behavioural topography in the striatum: differential effects of quinpirole and D-amphetamine microinjections.

Behavioural evidence has accumulated that supports the hypothesis that specific territories of the striatum contribute differentially to the control of motor behaviours. The present experiments compare the behavioural effects of microinjections of amphetamine (20 microg/0.5 microl) with those elicited by the D2-class dopamine receptor agonist quinpirole (3 microg/0.5 microl) following direct microinjection into three anatomically distinct sectors of the striatum: the nucleus accumbens, the ventrolateral striatum and the anterodorsal striatum. Our findings demonstrate that site-specific behavio

Carbachol-induced pressor responses and muscarinic M1 receptors in the central nucleus of amygdala in conscious rats.

The type of muscarinic receptor in the central nucleus of the amygdala that mediates the carbachol-evoked pressor responses was investigated in conscious unrestraint Sprague-Dawley rats. Carbachol (100 ng) injected into the lateral cerebral ventricle caused a significant rise in blood pressure of 31.8+/-4.5 mmHg and a decrease in heart rate of 80.0+/-12.2 beats/min. Pirenzepine (10-75 nmol) injected into the central nucleus of the amygdala inhibited carbachol-induced pressor responses dose-dependently. The bradycardic response to carbachol was also inhibited by pirenzepine, but no dose-depende

Role of brain angiotensin in thirst and sodium appetite of rats.

The role of brain angiotensin II (ANG II) in water, Na and food intake of rats was studied. Intracerebroventricular (i.c.v.) infusion (100 micrograms/h) of the non-peptide ANG II receptor antagonist losartan (type 1), but not PD123319 (type 2), completely blocked water intake caused by i.c.v. infusion of ANG II at 50 ng/h. Following food deprivation, food intake was reduced by PD123319 and associated water intake was decreased by losartan or PD123319. Neither water intake after water deprivation nor Na intake after Na depletion was altered by losartan or PD123319. In conclusion, evidence was c

Involvement of dopamine D3 receptors in the area postrema in R(+)-7-OH-DPAT-induced emesis in the ferret.

We investigated the possible involvement of dopamine D3 receptors in R(+)-7-hydroxy-2-(N,N-di-n-propylamino)tetraline (R(+)-7-OH-DPAT)-induced emesis in the ferret. The R(+)enantiomer of 7-OH-DPAT (0.03-1 mg/kg, s.c.) caused emesis in a dose-dependent manner, whereas the S(-)enantiomer, even at 1 mg/kg s.c. failed to induce emesis. Quinpirole (0.1-1.0 mg/kg) and apomorphine (0.3 mg/kg, s.c. only) also elicited an emetic response. S(-)-Eticlopride, which has a high affinity for the dopamine D3 receptor, antagonized R(+)-7-OH-DPAT (0.3 mg/kg, s.c.)-induced emesis (ID50 1.4 micrograms/kg, s.c.).

Enhanced restoration of striatal dopamine concentrations by combined GM1 ganglioside and neurotrophic factor treatments.

Intraperitoneal injection of GM1 ganglioside or intracerebroventricular infusion of basic fibroblast growth factor (FGF-2) or epidermal growth factor (EGF) partially restored dopamine and 3,4-dihydroxyphenylacetic acid (DOPAC) levels in the striatum of young MPTP-treated mice. Combined treatments of GM1 ganglioside with FGF-2 or EGF produced a greater restoration of striatal dopamine levels than treatments with GM1 or either of the neurotrophic factors alone. GM1 treatment, but not trophic factor treatments caused significant sparing of substantia nigra pars compacta (SNc) tyrosine hydroxylase

Pharmacological evidence for the involvement of endogenous alpha-MSH-like peptides in peripheral nerve regeneration.

The possible involvement of alpha-MSH-like peptides in the regenerative response of peripheral nerves was investigated with a competitive antagonist of alpha-MSH, the synthetic hexapeptide [D-Trp7,Ala8,D-Phe10)alpha-MSH(6-11)-amide. Subcutaneous administration of the alpha-MSH antagonist during the first 10 days following sciatic nerve crush significantly decreased functional recovery as measured by the foot flick withdrawal test and the walking pattern analysis. Hypophysectomy delayed both the initial sprouting response and the outgrowth rate after major caudal nerve crush. When hypophysectom

Central neurogenic antiinflammatory action of alpha-MSH: modulation of peripheral inflammation induced by cytokines and other mediators of inflammation.

The neuropeptide alpha-melanocyte-stimulating hormone (alpha-MSH) has potent antipyretic and antiinflammatory properties. When administered systemically, the naturally occurring molecule and its COOH-terminal tripeptide sequence inhibit inflammation induced by peripherally applied irritants and intradermal injections of mediators of inflammation such as interleukin-1 (IL-1), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF alpha). We recently found that alpha-MSH can act solely within the brain to inhibit inflammation caused by a general irritant applied to the skin. This activity ap

Calcium channel blockade attenuates angiotensin II-induced drinking in rats.

Lateral ventricular administration of angiotensin II (ANG II) produces potent dipsogenic effects in water-sated rats. ANG II seems to require functional voltage-gated calcium channels on neurons throughout circumventricular brain sites to exert its effects. Although there are at least three types of calcium channels, only L-type calcium channel-blocking drugs have been reported to decrease drinking. Microinjection of neurotensin into the CNS induces hyperdipsia in the rat.

Neurotensin (NT) is a neuropeptide and putative neurotransmitter that has been shown to exert a variety of effects on digestive and ingestive processes. In order to address the possibility that NT might play a role in the regulation of water intake as well, the peptide was infused into the lateral cerebral ventricle, amygdala, ventral tegmental area, lateral hypothalamus, and preoptic area of the anterior hypothalamus of rats deprived of water for 16 hours. Neurotensin produced a significant and dose-dependent increase in water intake when injected into the ventricular system but had no effect

Effects of enkephalins and the analogue FK-33824 on mean arterial pressure and heart rate in conscious sheep.

Experiments were designed to evaluate the central and systemic effects by enkephalins and the enkephalin analogue FK-33824 on mean arterial pressure (MAP) and heart rate (HR) in conscious sheep. Intracerebroventricular infusion of FK-33824 increased both MAP and HR in a dose-dependent manner in normal sheep. The increases in MAP and HR were attenuated by naloxone administered centrally, but not systemically. Intracerebroventricular infusion of met-enkephalin, leu-enkephalin and naloxone failed to change both MAP and HR significantly. However, intravenous infusion of met-enkephalin, leu-enkepha

Neonatal diethylstilbestrol alters blood pressure and CNS drinking response in SHR and WKY rats.

Neonatal SHR and WKY rats were treated with diethylstilbestrol or propylene glycol (controls). Control SHR had higher blood pressures than WKY rats. Neonatal diethylstilbestrol treatment delayed the onset and full expression of hypertension in the male SHR but had no effect on blood pressure in the other animals. Neonatal diethylstilbestrol treatment had an adverse effect on body weights of male SHR and WKY rats through 98 days of age (not statistically decreased thereafter). Adult female SHR and WKY rats treated neonatally with diethylstilbestrol had body weights significantly greater than th

Assessment of the effects of a synthetic gonadotropin-releasing hormone associated peptide on hormone release from the in situ and ectopic pituitaries in adult male rats.

The in vivo effects of synthetic human gonadotropin-releasing hormone associated peptide (GAP) were evaluated in adult male rats. In normal rats, intracerebroventricular (III ventricle) injection of 5 ng GAP significantly increased plasma LH levels after 60 minutes. Intracerebroventricular administration of 5, 25 or 125 ng of GAP elevated circulating LH levels also at 120 minutes of injection but did not alter plasma FSH, prolactin or testosterone concentrations. In hypophysectomized-pituitary-grafted rats injection of 125 ng GAP directly into the ectopic pituitary induced no changes in periph

Effect of cholinergic deficit induced by ethylcholine aziridinium on serotonergic parameters in rat brain.

The consequence of loss of cholinergic input on the function of serotonergic neurons has been studied in rat brain after bilateral intracerebroventricular injections of various doses of the cholinotoxin ethylcholine aziridinium ion (1 to 5 nmoles/ventricle). This treatment resulted in a dose-dependent decrease in acetylcholine content in hippocampus, which occurred 2 days after injection and persisted during the 28 day observation period. The reduction in acetylcholine content ranged from 50.3 +/- 6.0% to 76.9 +/- 3.8% when compared to vehicle-injected rats. Other brain areas, including cortex

Central effects of leukotriene C4 and D4 in rats and mice.

Leukotrienes (LTs) C4 or D4 in a dose of 12 nmoles were administered into the lateral ventricle (i.c.v.) of the rat brain under urethane anaesthesia, and the changes in the blood pressure, heart and respiratory rate were investigated. In other animals the behaviour was evaluated by means of the open field test, and the rectal body temperature was measured. Besides, the content of noradrenaline, 5-hydroxytryptamine and 5-hydroxyindole acetic acid was estimated in different parts of the brain following i.c.v. application of LTs. In mice the antinociceptive effect of LTs in a dose of 1.5 nmoles w

Isolation and characterization of a putative endogenous benzodiazepineoid (endozepine) from bovine and human brain.

A protein termed endozepine (EP) which inhibits the binding of benzodiazepines to synaptosomal membranes (Ki approximately 5 microM) has been purified to electrophoretic homogeneity from bovine and human brain using acidic ethanol/chloroform extraction, Bio-Sil TSK-250 gel permeation chromatography, and reverse-phase high performance liquid chromatographies. Bovine and human EP are single-chain polypeptides and have molecular weights of approximately 10,000. Both proteins are very hydrophilic and contain an abundance of lysine, glutamic, and aspartic residues. Antisera prepared against bovine

Attempted antagonism of adenosine analogue induced depression of respiration.

Intracerebroventricular (ICV) administration of the stable adenosine analogue 2-chloroadenosine (2CA) to hyperoxic halothane-anesthetized rats produced a dose-dependent depression of respiration largely as a result of a decrease in tidal volume. Similar changes were noted after another adenosine analogue, phenylisopropyladenosine (PIA). Higher doses shifted the minute ventilation-PaCO2 curve to the right and decreased its slope. Bradycardia and hypotension were produced at doses which altered respiration. Neonatal destruction of brain serotonin or dopamine-containing nerve terminals did not al

Convulsant activity of pyridoxal sulphate and phosphonoethyl pyridoxal: antagonism by GABA and its synthetic analogues.

Pyridoxal phosphate and its synthetic analogues--pyridoxal 5'-sulphate and the 5-phosphonoethyl analogue of pyridoxal (phosphonoethyl pyridoxal) in doses of 0.125-0.250 (mumol/10 microliters/i.c.v./rat), caused epileptic seizures characterized by running fits, vocalization, muscular fasciculation and tonic-clonic convulsions. These effects were specific and could not be demonstrated with 5'-deoxypyridoxal, N-methylpyridoxal phosphate or the 5-trans-carboxyethenyl analogue of pyridoxal phosphate (carboxyethenyl pyridoxal). Structure-activity relationships of these analogues indicated that the p

The rate of CSF formation in man: preliminary observations on metrizamide washout as a measure of CSF bulk flow.

Metrizamide, a nonionic, water-soluble contrast agent, was injected into the ventricular system of 3 patients with indwelling ventricular catheters and subcutaneous cerebrospinal fluid reservoirs. A first-order rate constant, k, for metrizamide washout from the right lateral ventricle was determined with reference to mean ventricular metrizamide concentration on serial CAT scans; right ventricular volume, V, was estimated after metrizamide injection by multiplying the appropriate ventricular area on consecutive CAT slices by the nominal slice thickness.
