Acetylcholinesterase of the sand fly, Phlebotomus papatasi (Scopoli): cDNA sequence, baculovirus expression, and biochemical properties.

BACKGROUND: Millions of people and domestic animals around the world are affected by leishmaniasis, a disease caused by various species of flagellated protozoans in the genus Leishmania that are transmitted by several sand fly species. Insecticides are widely used for sand fly population control to try to reduce or interrupt Leishmania transmission. Zoonotic cutaneous leishmaniasis caused by L. major is vectored mainly by Phlebotomus papatasi (Scopoli) in Asia and Africa. Organophosphates comprise a class of insecticides used for sand fly control, which act through the inhibition of acetylchol

[The worldwide expansion of bed bugs also constitutes a problem in Germany].

Worldwide, reports of the spread of the bed bugs Cimex lectularius and C. hemipterus (in Australia) are increasing. Irrespective of hygiene conditions, the insects can be especially found in hotels and hostels as well as in other holiday accommodation, health facilities, transport and in the secondhand trade. From these localities the insects spread to private households. A bed bug infestation may well have health consequences and constitutes a heavy burden for those affected. The global spread of bed bugs appears to be multifactorially conditioned and the causes are controversially discussed.

Larvicidal Activity of Synthesized Silver Nanoparticles from Curcuma zedoaria Essential Oil against Culex quinquefasciatus.

Culex quinquefasciatus is the major vector of the bancroftian filarial parasite which causes human lymphatic filariasis and St. Louis encephalitis. The simple way to stop the transmission is to control the vector by using synthetic chemicals. However, herbal essential oils have biological properties, such as a larvicidal effect and are ecofriendly to use. In this study, we investigated the larvicidal activity of Curcuma zedoaria essential oil (ZEO) and biosynthesized silver nanoparticles using this essential oil (ZEO-AgNPs). The larvicidal activity against both insecticide-susceptible and -res

Transit through the flea vector induces a pretransmission innate immunity resistance phenotype in Yersinia pestis.

Yersinia pestis, the agent of plague, is transmitted to mammals by infected fleas. Y. pestis exhibits a distinct life stage in the flea, where it grows in the form of a cohesive biofilm that promotes transmission. After transmission, the temperature shift to 37 degrees C induces many known virulence factors of Y. pestis that confer resistance to innate immunity. These factors are not produced in the low-temperature environment of the flea, however, suggesting that Y. pestis is vulnerable to the initial encounter with innate immune cells at the flea bite site. In this study, we used whole-genom

Gene silencing through RNAi and antisense Vivo-Morpholino increases the efficacy of pyrethroids on larvae of Anopheles stephensi.

BACKGROUND: Insecticides are still at the core of insect pest and vector control programmes. Several lines of evidence indicate that ABC transporters are involved in detoxification processes against insecticides, including permethrin and other pyrethroids. In particular, the ABCG4 gene, a member of the G subfamily, has consistently been shown to be up-regulated in response to insecticide treatments in the mosquito malaria vector Anopheles stephensi (both adults and larvae). METHODS: To verify the actual involvement of this transmembrane protein in the detoxification process of permethrin, bio

Pyrazole Amide Scaffolds in Agricultural Chemistry: From Molecular Design to Applications.

Pyrazole amide compounds exhibit a wide range of biological activities, which enables them to hold an important position in the field of pesticide research. The pyrazole ring has characteristics such as multiple substitution sites, a wide optimization space, a unique mechanism of action, and no cross-resistance; the amide moiety represents a highly prevalent structural motif. It is frequently chosen as a pivotal pharmacophore and incorporated into the molecular architecture of compounds to enhance the relevant biological activities. Analyzing the concise structure-activity relationships (SAR)

Engineered mosquitoes to fight mosquito borne diseases: not a merely technical issue.

Malaria, dengue and other mosquito-borne diseases pose dramatic problems of public health, particularly in tropical and sub-tropical countries. Historically, vector control has been one of the most successfully strategies to eradicate some mosquito-borne diseases, as witnessed by malaria eradication in Mediterranean regions such as Italy and Greece. Vector control through insecticides has been used worldwide; unfortunately, it is losing effectiveness due to spread of resistances. Control of mosquito-borne diseases through field-releases of genetically engineered mosquitoes is an innovative and

Sulfonamide inhibition studies of the β carbonic anhydrase from Drosophila melanogaster.

An inibition study of the β-carbonic anhydrase (CA, EC 4.2.1.1) DmBCA from the insect Drosophila melanogaster with sulfonamides and sulfamates is reported. Among the panel of 40 investigated compounds, the best DmBCA inhibitors were the sulfonylated benzenesulfonamides and ethoxzolamide, which showed inhibition constants in the range of 65.3-138 nM. Methazolamide and sulthiame were also effective inhibitors with KIs ranging between 237 and 249 nM, whereas most of the simple aromatic/heterocyclic sulfonamides showed inhibition constants in the range of 0.47-6.40 μM. Prevalence of rat fleas in and around Delhi (India) area and their susceptibility status to insecticides.

A rapid survey in Delhi and surrounding areas was undertaken in September 1994, during plague outbreak in District Beed (Maharashtra) and Surat (Gujarat) to monitor rodent/flea population and to determine susceptibility status of fleas to insecticides. The five rodent species in order of their prevalence were Rattus rattus (86.06%), Mus musculus (15.33%), Suncus murinus (2.47%), Bandicoota indica (0.48%) and B. bengalensis (0.48%). Two flea species, Xenopsylla cheopis and X. astia, were captured from these rodents. The absolute flea index was found to be ranging between 0.08 to 4.0 in various

Selective and irreversible inhibitors of mosquito acetylcholinesterases for controlling malaria and other mosquito-borne diseases.

New insecticides are urgently needed because resistance to current insecticides allows resurgence of disease-transmitting mosquitoes while concerns for human toxicity from current compounds are growing. We previously reported the finding of a free cysteine (Cys) residue at the entrance of the active site of acetylcholinesterase (AChE) in some insects but not in mammals, birds, and fish. These insects have two AChE genes (AP and AO), and only AP-AChE carries the Cys residue. Most of these insects are disease vectors such as the African malaria mosquito (Anopheles gambiae sensu stricto) or crop

Endophyte-mediated pest resistance: mechanisms, evidence gaps, and translational opportunities.

Escalating crop losses caused by insect pests, together with pesticide resistance, environmental persistence, and increasing regulatory scrutiny of conventional agrochemicals, have intensified interest in biologically based pest-management strategies. Endophytic bacteria and fungi represent a promising avenue for crop protection because they can influence plant defense, pest performance, and multitrophic interactions within plant tissues. This review critically synthesizes the 'landscape' of endophyte-mediated pest resistance-the 'journey' of microbes from colonization to field protection-thro

Detection of West Nile Virus - Lineage 2 in Culex pipiens mosquitoes, associated with disease outbreak in Greece, 2017.

During July-October 2017 a WNV outbreak took place in the Peloponnese, Southern Greece with five confirmed deaths. During routine monitoring survey in the Peloponnese, supported by the local Prefecture, we have confirmed the presence of all three Culex pipiens biotypes in the region, with a high percentage of Culex pipiens/molestus hybrids (37.0%) which are considered a highly competent vector of WNV. Kdr mutations related to pyrethroid resistance were found at relatively low levels (14.3% homozygosity) while no mosquitoes harboring the recently identified chitin synthase diflubenzuron-resista

[Therapy and prevention of malaria].

Malaria is still, in spite of intensive efforts to reduce its transmission, the most serious and widespread protozoal infection in man. More than 100 million people suffer of malaria each year and one million, mostly children, die for it. Widespread resistance of P. falciparum to drugs, especially 4-aminoquinoline, has been progressing to such a speed in many endemic malarious areas that therapy and prophylaxis procedures have been changing and new drugs or associations of them have been introduced. Quinine and chloroquine are still the main therapeutic agents against the blood forms of plasmo

Macrocyclic lactone resistance in Dirofilaria immitis: risks for prevention of heartworm disease.

Heartworm disease, caused by Dirofilaria immitis, can be lethal in dogs and cats. It is transmitted by mosquitoes, and occurs in many parts of the world. Prevention relies on macrocyclic lactones. Macrocyclic lactones used are ivermectin, selamectin, abamectin, eprinomectin, milbemycin oxime and moxidectin, administered at 30-day intervals during the transmission season. Some moxidectin formulations are long-acting injectables. In the USA, preventives are recommended throughout the year. Loss of efficacy of macrocyclic lactone preventives was reported in 2005 and proof of resistance in the USA

Acaricide and ivermectin resistance in a field population of Rhipicephalus microplus (Acari: Ixodidae) collected from red deer (Cervus elaphus) in the Mexican tropics.

In the Neotropics the control of tick infestations in red deer (Cervus elaphus) is achieved primarily through the use of acaricides and macrocyclic lactones. In Mexico, resistance to one or multiple classes of acaricides has been reported in Rhipicephalus microplus infesting cattle, but information on acaricide susceptibility in R. microplus infesting red deer is lacking. In this study we report the level of resistance to different classes of acaricides and ivermectin in R. microplus collected from red deer in the Mexican tropics. Engorged R. microplus females were collected from a red deer fa

Towards a rational policy for dealing with tsetse.

The past 20 years have seen the development of bait technologies that enable livestock keepers to control tsetse flies and, hence, African trypanosomiasis. The techniques have, however, often been applied on too small a scale, without due regard to the realities of tsetse population dynamics. The consequent lack of progress has led to calls for a return to large-scale operations. Analysis of successful programmes to control or eliminate tsetse in southern Africa suggests that the combined use of recently improved bait methods and insecticide spraying will provide the building blocks for achiev

Identifying hazards in complex ecological systems. Part 3: Hierarchical Holographic Model for herbicide tolerant oilseed rape.

This paper is the third in a series designed to demonstrate the application of rigorous, systematic hazard identification techniques to ecological systems. Here we use Hierarchical Holographic Modelling to identify the potential ecological hazards associated with the commercial release of herbicide tolerant oilseed rape. Hierarchical Holographic Models decompose complex systems into a series of sub-systems and consider interactions between the components and processes of these sub-systems in order to identify hazards. In this example we considered 1356 potential interactions between the biolog

Treatment of ectoparasitic infections: review of the English-language literature, 1982-1992.

We reviewed the English-language literature of 1982-1992 for papers concerning scabies, pediculosis, and their treatments; this information was supplemented with opinions of workers in the field of ectoparasitic disease. Six treatment trials for scabies, one study of the treatment of pediculosis pubis, and additional studies on the treatment of pediculosis capitis were included. Both lindane 1% and permethrin 5% are effective treatments for scabies, although resistance to lindane does exist. Lindane 1%, permethrin 1%, and pyrethrins with piperonyl butoxide are all effective treatments for pedi
