A Bayesian approach to Mendelian randomization with multiple pleiotropic variants.

We propose a Bayesian approach to Mendelian randomization (MR), where instruments are allowed to exert pleiotropic (i.e. not mediated by the exposure) effects on the outcome. By having these effects represented in the model by unknown parameters, and by imposing a shrinkage prior distribution that assumes an unspecified subset of the effects to be zero, we obtain a proper posterior distribution for the causal effect of interest. This posterior can be sampled via Markov chain Monte Carlo methods of inference to obtain point and interval estimates. The model priors require a minimal input from t

Time varying methods to infer extremes in dengue transmission dynamics.

Dengue is an arbovirus affecting global populations. Frequent outbreaks occur, especially in equatorial cities such as Singapore, where year-round tropical climate, large daily influx of travelers and population density provide the ideal conditions for dengue to transmit. Little work has, however, quantified the peaks of dengue outbreaks, when health systems are likely to be most stretched. Nor have methods been developed to infer differences in exogenous factors which lead to the rise and fall of dengue case counts across extreme and non-extreme periods. In this paper, we developed time varyi

Forecasting of salmonellosis epidemic proces in Ukraine using autoregressive integrated moving average model.

The article highlights the problem of salmonellosis among the population of the Kharkov region, Ukraine. Three time series were used for calculations: a series of incidence rates for men, a series of incidence rates for women and a series of incidence rates for the general population, each of the series was an ordered set of monthly values from December 2015 to December 2018. It was revealed that the most effective tool for analyzing these statistical data is the use of the autoregressive moving average model (ARIMA). The following steps were used: identification and replacement of outliers, t

Bayesian inference for network meta-regression using multivariate random effects with applications to cholesterol lowering drugs.

Low-density lipoprotein cholesterol (LDL-C) has been identified as a causative factor for atherosclerosis and related coronary heart disease, and as the main target for cholesterol- and lipid-lowering therapy. Statin drugs inhibit cholesterol synthesis in the liver and are typically the first line of therapy to lower elevated levels of LDL-C. On the other hand, a different drug, Ezetimibe, inhibits the absorption of cholesterol by the small intestine and provides a different mechanism of action. Many clinical trials have been carried out on safety and efficacy evaluation of cholesterol lowerin

Practical guide to the meta-analysis of rare events.

OBJECTIVE: Meta-analysing studies with low event rates is challenging as some of the standard methods for meta-analysis are not well suited to handle rare outcomes. This is more evident when some studies have zero events in one or both treatment groups. In this article, we discuss why rare events require special attention in meta-analysis, we present an overview of some approaches suitable for meta-analysing rare events and we provide practical recommendations for their use. METHODS: We go through several models suggested in the literature for performing a rare events meta-analysis, highlight

Childhood obesity: how long should we wait to predict weight?

Obesity is highly prevalent in children under the age of 5 years, although its identification in infants under 2 years remains difficult. Several clinical prediction models have been developed for obesity risk in early childhood, using a number of different predictors. The predictive capacity (sensitivity and specificity) of these models varies greatly, and there is no agreed risk threshold for the prediction of early childhood obesity. Of the existing models, only two have been practically utilized, but neither have been particularly successful. This commentary suggests how future research ma

Factor Structure of the Eating Disorder Examination-Questionnaire in a Clinical Sample of Adult Women With Anorexia Nervosa.

An exploratory factor analysis on the Eating Disorder Examination-Questionnaire (EDE-Q) is presented for a clinical sample of women with anorexia nervosa. THE EDE-Q was completed by 169 participants after admission to an inpatient unit for eating disorders. Results of the current study did not support the four-factor model presented by the EDE-Q. A new four-factor solution was obtained with two factors showing similarity to the Restraint and Eating Concern subscales of the original model. The Shape and Weight Concern items primarily loaded together on one factor, along with preoccupation with

Mapping the potential distribution of the schistosomiasis intermediate host Biomphalaria straminea in China.

Biomphalaria straminea is native to Brazil but has established a population in Guangdong Province, China. Its potential of expanding and transmitting Schistosoma mansoni is of great concern. We seek to map S. mansoni transmission potential by predicting ecological dimensions and potential distributions of B. straminea using state-of-the-art ecological niche model approaches. Two environmental datasets were selected to fit individual and ensemble niche models, together with the evaluation of niche conservatism during B. straminea invasion in China. A small area is still occupied by the introduc

Spatial PM2.5, NO2, O3 and BC models for Western Europe - Evaluation of spatiotemporal stability.

BACKGROUND: In order to investigate associations between air pollution and adverse health effects consistent fine spatial air pollution surfaces are needed across large areas to provide cohorts with comparable exposures. The aim of this paper is to develop and evaluate fine spatial scale land use regression models for four major health relevant air pollutants (PM2.5, NO2, BC, O3) across Europe. METHODS: We developed West-European land use regression models (LUR) for 2010 estimating annual mean PM2.5, NO2, BC and O3 concentrations (including cold and warm season estimates for O3). The models w

Pathogen seasonality and links with weather in England and Wales: a big data time series analysis.

BACKGROUND: Many infectious diseases of public health importance display annual seasonal patterns in their incidence. We aimed to systematically document the seasonality of several human infectious disease pathogens in England and Wales, highlighting those organisms that appear weather-sensitive and therefore may be influenced by climate change in the future. METHODS: Data on infections in England and Wales from 1989 to 2014 were extracted from the Public Health England (PHE) SGSS surveillance database. We conducted a weekly, monthly and quarterly time series analysis of 277 pathogen serotype

Real-time temperature monitoring with fiber Bragg grating sensor during diffuser-assisted laser-induced interstitial thermotherapy.

High-sensitivity temperature sensors have been used to validate real-time thermal responses in tissue during photothermal treatment. The objective of the current study was to evaluate the feasible application of a fiber Bragg grating (FBG) sensor for diffuser-assisted laser-induced interstitial thermotherapy (LITT) particularly to treat tubular tissue disease. A 600 - ? m core-diameter diffuser was employed to deliver 980-nm laser light for coagulation treatment. Both a thermocouple and a FBG were comparatively tested to evaluate temperature measurements in ex vivo liver tissue. The degree of

A bivariate contaminated binormal model for robust fitting of proper ROC curves to a pair of correlated, possibly degenerate, ROC datasets.

PURPOSE: The objective was to design and implement a bivariate extension to the contaminated binormal model (CBM) to fit paired receiver operating characteristic (ROC) datasets-possibly degenerate-with proper ROC curves. Paired datasets yield two correlated ratings per case. Degenerate datasets have no interior operating points and proper ROC curves do not inappropriately cross the chance diagonal. The existing method, developed more than three decades ago utilizes a bivariate extension to the binormal model, implemented in CORROC2 software, which yields improper ROC curves and cannot fit dege

Study protocol for a transversal study to develop a screening model for excessive gambling behaviours on a representative sample of users of French authorised gambling websites.

INTRODUCTION: Since the legalisation of online gambling in France in 2010, gambling operators must implement responsible gambling measures to prevent excessive gambling practices. However, actually there is no screening procedure for identifying problematic gamblers. Although several studies have already been performed using several data sets from online gambling operators, the authors deplored several methodological and clinical limits that prevent scientifically validating the existence of problematic gambling behaviour. The aim of this study is to develop a model for screening excessive gam

Prediction accuracy for the cure probabilities in mixture cure models.

Prediction accuracy of a cure model when it is used to predict the cure probability of a patient is an important but not well-addressed issue in survival analysis. We propose a method to assess the prediction accuracy of a mixture cure model in predicting cure probability based on inverse probability of censoring weights to incorporate the censoring and latent cure status in the data. The inverse probability of censoring weight-adjusted estimator is shown to be consistent for the true expected prediction error for cure probability. A simulation study shows that the estimator performs well with

The application of Bayesian hierarchical models to quantify individual diet specialization.

Intraspecific variation in ecologically relevant traits is widespread. In generalist predators in particular, individual diet specialization is likely to have important consequences for food webs. Understanding individual diet specialization empirically requires the ability to quantify individual diet preferences accurately. Here we compare the currently used frequentist maximum likelihood approach, which infers individual preferences using the observed prey proportions to Bayesian hierarchical models that instead estimate these proportions. Using simulated and empirical data, we find that the

The different faces of impulsivity as links between childhood maltreatment and young adult crime.

Crime is a major public health and safety threat. Many studies have suggested that early exposure to child maltreatment increases an individual's risk for persistent serious crime in adulthood. Despite these findings about the connection between child maltreatment and criminal behavior, there is a paucity of empirically-based knowledge about the processes or pathways that link child maltreatment to later involvement in crime. Using a community sample of 337 young adults (ages 18-25) in a U.S. metropolitan area, the present study examined the role of various facets of impulsivity in linking chi

Explaining the geographic spread of emerging epidemics: a framework for comparing viral phylogenies and environmental landscape data.

BACKGROUND: Phylogenetic analysis is now an important tool in the study of viral outbreaks. It can reconstruct epidemic history when surveillance epidemiology data are sparse, and can indicate transmission linkages among infections that may not otherwise be evident. However, a remaining challenge is to develop an analytical framework that can test hypotheses about the effect of environmental variables on pathogen spatial spread. Recent phylogeographic approaches can reconstruct the history of virus dispersal from sampled viral genomes and infer the locations of ancestral infections. Such metho

The relative importance of frequency of contacts and duration of exposure for the spread of directly transmitted infections.

The recent availability of survey data on social contact patterns has made possible important advances in the understanding of the social determinants of the spread of close-contact infections, and of the importance of long-lasting contacts for effective transmission to occur. Still, little is known about the relationship between two of the most critical identified factors (frequency of contacts and duration of exposure) and how this relationship applies to different types of infections. By integrating data from two independently collected social surveys (Polymod and time use), we propose a mo

Risk pathways for gonorrhea acquisition in sex workers: can we distinguish confounding from an exposure effect using a priori hypotheses?

The population distribution of sexually transmitted infections (STIs) varies broadly across settings. Although there have been many studies aiming to define subgroups at risk of infection that should be a target for prevention interventions by identifying risk factors, questions remain about how these risk factors interact, how their effects jointly influence the risk of acquisition, and their differential importance across populations. Theoretical frameworks describing the interrelationships among risk determinants are useful in directing both the design and analysis of research studies and i
