Assays for the analysis of synaptic proteins in neurodegenerative disorders.

Neurodegenerative disorders are characterized by damage to selective neuronal populations (1) that could be followed or proceeded by synapti … s disease (HD) (13,14), Parkinson's disease (PD) (15-17), Creutzfeldt-Jakob disease (CJD) (18), and myotonic dystrophy (19) have turned out to be the result of mutations in proteins that concentrate at the synaptic site. Therefore, in order to better understand the mechanisms of central nervous system (CNS) dysfunction in neurodegenerative disorders, it is important …

[Endocrinology 1996-1997].

Post-traumatic stress-induced disorders are still the focus of interest and most recently discussions are under way whether stress-induced c … f recombinant preparations will be available, they will be tested in amyotrophic lateral sclerosis, myotonic dystrophy, multiple sclerosis, catabolic conditions, osteoporosis, in renal failure and to promote wound healing. STH may also prove useful in cardiac failure, in particular in cardiac cachexia. That TRH has receptors in the gut is not surprising, it acts, ho …

Evolution of the Friedreich's ataxia trinucleotide repeat expansion: founder effect and premutations.

Friedreich's ataxia, the most frequent inherited ataxia, is caused, in the vast majority of cases, by large GAA repeat expansions in the fir … gle generation. This stepwise evolution to large trinucleotide expansions already was suggested for myotonic dystrophy and fragile X syndrome and may relate to a common mutational mechanism, despite sequence motif differences.

Comparison of genetic services with and without genetic registers: knowledge, adjustment, and attitudes about genetic counselling among probands referred to three genetic clinics.

Genetic register services incorporating long term follow up and a proactive approach to at risk subjects have been recommended as a way of i … hree regional genetic clinics because of a family history of Duchenne or Becker muscular dystrophy, myotonic dystrophy, or chromosome translocations, and 43 of the referring GPs. Probands attending the centre using a genetic register approach were compared with those from the two centres offering the standard clinical genetic service. A very high proportion of proba …

Regional variation of thigh muscle fat infiltration in patients with neuromuscular diseases compared to healthy controls.

BACKGROUND: Chemical shift encoding-based water-fat magnetic resonance imaging (CSE-MRI) measures a quantitative biomarker: the proton density fat fraction (PDFF). The aim was to assess regional and proximo-distal PDFF variations at the thigh in patients with myotonic dystrophy type 2 (DM2), limb-girdle muscular dystrophy type 2A (LGMD2A), and late-onset Pompe disease (LOPD) as compared to healthy controls. METHODS: Seven patients (n=2 DM2, n=2 LGMD2A, n=3 LOPD) and 20 controls were recruited. A 3D-spoiled gradient echo sequence was used to scan the thigh musculature. Muscles were manually se…

Efficacy and safety of glucocorticoids in the treatment of progressive muscular dystrophy in children: a systematic review and meta-analysis.

BACKGROUND: Hormonal drug therapy has been widely used in clinical practice for the treatment of progressive muscular dystrophy (PMD). Glucocorticoids, as a common drug in the clinical treatment of PMD, have been reported in several clinical studies. METHODS: Chinese and English databases were respectively searched using "randomized controlled trials", "Duchenne-type myotonic dystrophy", "glucocorticoids", Prednisone", "Prednisolone", and "Methylprednisolone", and "Defibrotide" were used as search terms. The meta-analysis was performed using the RevMan 5.3 and Stata 13 software provided by th…

Impaired pre-mRNA processing and altered architecture of 3' untranslated regions contribute to the development of human disorders.

The biological fate of each mRNA and consequently, the protein to be synthesised, is highly dependent on the nature of the 3' untranslated region. Despite its non-coding character, the 3' UTR may affect the final mRNA stability, the localisation, the export from the nucleus and the translation efficiency. The conserved regulatory sequences within 3' UTRs and the specific elements binding to them enable gene expression control at the posttranscriptional level and all these processes reflect the actual state of the cell including proliferation, differentiation, cellular stress or tumourigenesis.

Pituitary-testicular axis in cardiomyopathic Syrian hamsters.

Testicular function and other endocrine parameters were studied in cardiomyopathic hamsters. In these animals, the major defect is an intracellular calcium overload, which is due to defective voltage-sensitive calcium channels. Basal circulating gonadotropin, prolactin, and triiodothyronine levels were lower in cardiomyopathic hamsters than in normal hamsters, but thyroxine, progesterone, and testosterone levels were not. In cardiomyopathic hamsters, the luteinizing hormone receptor (LHR) positive autoregulation by human chorionic gonadotropin (hCG) reached a maximum faster and at a lower dose

F-wave conduction velocity in the deep peroneal nerve: Charcot-Marie-Tooth disease and dystrophia myotonica.

The F-wave has been used to estimate the motor nerve conduction velocity (MNCV) along the proximal segment (spinal cord to knee) of the axons of the deep peroneal nerve in patients with Charcot-Marie-Tooth disease and those with dystrophia myotonica. A new, modified method has been applied to estimate proximal MNCV in patients in whom F-waves or M-responses cannot be obtained from the small muscles of the foot. Terminal latencies and MNCV along the distal nerve segment (knee to ankle) have also been estimated using conventional techniques. The results have been compared with those obtained for

Epigenetics in nucleotide repeat expansion disorders.

Over the past 20 years, nucleotide repeat expansion disorders have informed our broader understanding of neurodevelopmental and neurodegenerative disease. This is especially true with regard to the contributions of epigenetic mechanisms to neurologic disease pathogenesis. In this review, the authors describe a few of the myriad ways in which epigenetic processes underlie aspects of repeat expansion disorder pathophysiology and discuss how therapies targeted at epigenetic modulation hold promise for many of these disorders.

Foreign body ingestion in children: case series, review of the literature and guidelines on minimising accidental ingestions.

Foreign body ingestion in young children is one of the commonest presentations in the Accident and Emergency Department. It has been estimated that 40% of such ingestions may go unnoticed and the child may remain asymptomatic. However, complications can arise which may need urgent medical attention. Unwitnessed cases can delay the diagnosis as foreign bodies may remain in the oesophagus or stomach for a significant period of time causing non-specific symptoms. An index of suspicion is needed amongst primary and secondary health care practitioners to manage these cases early. Prevention of inge

Sleep apnea following a pharyngeal flap: a feared complication.

Pharyngeal flaps are often used to correct velopharyngeal insufficiency. They produce a permanent partial obstruction of the velopharyngeal space. Respiratory obstruction and obstructive sleep apnea have been reported following this surgery. We undertook a study to find out the incidence of sleep apnea associated with pharyngeal flap surgery. Forty-one children (aged 2 to 22 years) admitted for a pharyngeal flap underwent a polysomnographic recording prior to their surgery. One child with Steinert's disease showed some episodes of obstructive apnea, and the surgery was canceled. Forty children

Frequency and satisfaction of conventional and complementary or alternative therapies for neuromuscular disorders.

BACKGROUND: Causal therapies are not yet available for most neuromuscular diseases. Additionally, data on the use of complementary or alternative therapies (CAM) in patients groups with a variety of different neuromuscular diseases are rare. This retrospective cross-sectional study aims to record the frequency of use and satisfaction of conventional therapies and complementary or alternative medicine (CAM) in patients with neuromuscular disorders in order to compare them afterwards. METHODS: Patients from the University of Leipzig (Department and Outpatient Department of Neurology), the Fried

Three family members with elevated plasma cobalamin, transcobalamin and soluble transcobalamin receptor (sCD320).

BACKGROUND: Plasma cobalamin is requested in order to diagnose cobalamin deficiency and low levels confirm a deficient state. Here, we present three family members with unexpected high levels of cobalamin. METHODS: We included a patient referred for cobalamin measurement due to neurological symptoms, her son and her daughter. Mother and son both suffered from myotonic dystrophy type II, while the daughter tested negative for this disease. Blood samples were analyzed for cobalamin, haptocorrin, transcobalamin, holoTC, and sCD320. We employed gel filtration and antibody precipitation for furthe…
