Influence of the single or combined administration of cocaine and testosterone in autonomic and neuroendocrine responses to acute restraint stress.

Abuse of cocaine and androgenic-anabolic steroids (AASs) has become a serious public health problem. Despite reports of an increase in the incidence of simultaneous abuse of these substances, potential toxic interactions between cocaine and AASs are poorly known. In the present study, we investigated the effects of either single or combined administration of testosterone and cocaine for one or 10 consecutive days on autonomic (arterial pressure, heart rate and tail cutaneous temperature) and neuroendocrine (plasma corticosterone) responses induced by acute restraint stress in rats. Combined ad

A corticosteroid-responsive transcription factor, promyelocytic leukemia zinc finger protein, mediates protection of the cochlea from acoustic trauma.

Animals can be induced to resist cochlear damage associated with acoustic trauma by exposure to a variety of "conditioning" stimuli, including restraint stress, moderate level sound, heat stress, hypoxia, and corticosteroids. Here we identify in mice a corticosteroid-responsive transcription factor, PLZF (promyelocytic leukemia zinc finger protein), which mediates conditioned protection of the cochlea from acoustic trauma. PLZF mRNA levels in the cochlea are increased following conditioning stimuli, including restraint stress, dexamethasone administration, and moderate-to-high level acoustic s

Activity of the 5-HT1A receptor is involved in the alteration of glucocorticoid receptor in hippocampus and corticotropin-releasing factor in hypothalamus in SPS rats.

Rats exposed to single-prolonged stress (SPS) showed enhanced inhibition of the hypothalamic-pituitary-adrenal (HPA) system and alteration in the glucocorticoid/mineralocorticoid receptor. Dysfunction of the HPA axis is one of the core neuroendocrine abnormalities of post-traumatic stress disorder (PTSD). Serotonergic receptor, glucocorticoid receptor (GR) and corticotropin-releasing factor (CRF) have been proposed to play major roles in dysfunction of the HPA axis. However, the precise molecular mechanism is unknown. In this study, we investigated the relationships between the changes of GR i

Nociception- and anxiety-like behavior in rats submitted to different periods of restraint stress.

The aim of this study was to evaluate the effect of acute, sub-chronic and chronic stress on nociception induced by formalin injection in rats' temporomandibular joint (TMJ). It was evaluated the relation between blood levels of adrenocorticotropin, corticosterone, the levels of anxiety and nociceptive responses recorded after different stress protocols. Animals were initially submitted to acute restraint stress (15; 30 min and 1 h), or exposed to sub-chronic (3 days-1 h/day) or chronic stress (40 days-1 h/day). Then, animals were (1) killed immediately to collect blood for hormonal determinat

Importance of the pineal gland, endogenous prostaglandins and sensory nerves in the gastroprotective actions of central and peripheral melatonin against stress-induced damage.

Melatonin attenuates acute gastric lesions induced by topical strong irritants because of scavenging of free radicals, but its role in the pathogenesis of stress-induced gastric lesions has been sparingly investigated. In this study we compared the effects of intragastric (i.g.) or intracerebroventricular (i.c.v.) administration of melatonin and its precursor, L-tryptophan, with or without concurrent treatment with luzindole, a selective antagonist of melatonin MT2 receptors, on gastric lesions induced by water immersion and restraint stress (WRS). The involvement of pineal gland, endogenous p

Central GABA-ergic mechanism in stress-induced gastric ulceration.

The effect of exogenous administration of central amino acid neurotransmitters gamma-aminobutyric acid (GABA), glycine, glutamic acid and aspartic acid) into the cerebroventricular system was studied on gastric ulceration induced in albino rats either by 2 h restraint at 4 degrees C or by 6 h restraint at room temperature (30 +/- 2 degrees C). GABA (5, 10, 20 and 50 micrograms) injected intracerebroventricularly (i.c.v.) showed a dose-dependent reduction of gastric ulceration induced by 2 h restraint at 4 degrees C (CRU), whereas glycine (5, 10 and 20 micrograms i.c.v.) failed to alter this re

Impacts of the COVID-19 Pandemic on Incidents of Workplace Violence Events as Reported in Patient Safety Event Reports.

BACKGROUND AND OBJECTIVES: Workplace violence (WPV) committed against frontline health care workers is a worldwide concern but relatively little is known about relative rates of reported WPV events as identified through patient safety event (PSE) reports before and during the COVID-19 pandemic. This study sought to understand how the COVID-19 pandemic changed the rates and characteristics of reported WPV events committed by patients against health care workers, as captured in the PSE report system for a Mid-Atlantic health care system. METHODS: This retrospective cohort study analyzed volunta

Ethical experiences of nursing students in inpatient psychiatric units: The autonomy-paternalism tension.

BackgroundSymptoms of mental disorder that impair decision-making capacity in inpatient psychiatric units increase the ethical tension between patient autonomy and paternalistic responsibility. Practices such as involuntary admission, compulsory treatment, and physical restraint further intensify this tension, while nursing students are more vulnerable to these ethical dilemmas because of their limited clinical experience.AimThis study aimed to describe nursing students' ethical experiences regarding the autonomy-paternalism tension in inpatient psychiatric units.Research designA qualitative d

Cultural influences on coping among Chinese children aged 7-14 undergoing chemotherapy: implications for pediatric oncology service delivery.

BACKGROUND: Children undergoing chemotherapy experience significant psychosocial stress; however, limited research has examined how cultural and relational contexts shape stress appraisal and coping within pediatric oncology services in China. Understanding these mechanisms is essential for developing culturally responsive psychosocial care models. METHODS: A qualitative descriptive study was conducted using individual semi-structured interviews with 22 children aged 7-14 years undergoing chemotherapy for haematological cancers in a tertiary children's hospital in China. Data were analysed us

"Fulfilling the hunger": A qualitative study to understand the etiology of binge eating in adolescents experiencing food insecurity.

Food-insecure populations experience increased risk for binge eating. This study examined how food insecurity contributes to development of binge eating during adolescence-a key period for its onset-alongside broader contributing factors in this life stage. Semi-structured qualitative interviews were conducted with 30 adolescents ages 12-19 years who reported past-year food insecurity (via the Child Food Insecurity Experiences Scale) and any past-month objective and/or subjective binge eating (via items from the Youth Eating Disorder Examination-Questionnaire). Grounded theory analysis yielded

Age-Related Patterns in Pediatric Road Traffic Injuries in Romania.

Background: Pediatric road traffic injuries (RTIs) represent a significant public health concern, particularly in countries like Romania, where road infrastructure and safety remain challenges. Despite recent economic reclassification, Romania continues to report high rates of pediatric traffic-related injuries. Non-fatal RTIs often result in long-term physical and psychological harm. This study aims to assess age- and gender-specific injury patterns and mechanisms of non-fatal RTIs in children and adolescents, using data from "St. Mary's" Emergency Clinical Hospital for Children in Iași over

Prior short-term exercise prevents behavioral and biochemical abnormalities induced by single prolonged stress in a rat model of posttraumatic stress disorder.

Posttraumatic stress disorder (PTSD) is an anxiety disorder that occurs following exposure to somatic or psychotic trauma. Physical activity is known to improve symptoms of certain neuropsychiatric disorders. However, the role of exercise on acquired PTSD-like phenotype was not examined. The present study investigated the effects of prior moderate treadmill exercise on anxiety-like behaviors, serum corticosterone and BDNF levels, hippocampal BDNF and mRNA expression of apoptotic - related proteins in the single prolonged stress (SPS) as an animal model of PTSD in rats. Male and female rats und

A panic attack-like unusual stress reaction.

Ever since the seminal studies of Hans Selye, activation of hypothalamus-pituitary-adrenal (HPA) axis is emblematic of stress. Consequently, the lack of HPA axis responses following the undisputable psychological stress of a panic attack stands out as one of the most intriguing findings of contemporary psychiatry. On the other hand, the defensive behaviors and aversive emotions produced by stimulation of the dorsal periaqueductal gray matter (DPAG) have been proposed as a model of panic attacks. Therefore, we examined whether the plasma levels of 'stress hormones' corticotropin and prolactin s

Routine droperidol pre-medication improves sedation for ERCP.

BACKGROUND: Pre-medication with droperidol has been used to improve sedation during endoscopy, especially in patients with a history of alcohol or narcotic abuse. We studied whether routine use of droperidol pre-endoscopic retrograde cholangiopancreatography (ERCP) could improve patient and physician satisfaction with sedation. METHODS: Sixty-seven patients undergoing routine ERCP were enrolled in this double-blind placebo-controlled study. Patients were given either parenteral normal saline solution or 5 mg of droperidol 15 minutes before the procedure. After the ERCP, several parameters of

Traditional Chinese herbal formula, Fuzi-Lizhong pill, produces antidepressant-like effects in chronic restraint stress mice through systemic pharmacology.

ETHNOPHARMACOLOGICAL RELEVANCE: Fuzi-Lizhong pill (FLP) is a well-validated traditional Chinese medicine (TCM) formula that has long been used in China for gastrointestinal disease and adjunctive therapy for depression. In our previous study, we reported that the principal herb of FLP, Aconitum carmichaelii Debx. (Fuzi), exhibits antidepressant-like effects. However, there have been no reports on whether FLP produces antidepressant-like effects and its potential molecular mechanisms. AIM OF THE STUDY: We aim to demonstrate the antidepressant-like effects of FLP in chronic restraint stress (CR

miR-449a contributes to glucocorticoid-induced CRF-R1 downregulation in the pituitary during stress.

The hypothalamic-pituitary-adrenal axis is controlled by the feedback of glucocorticoids on the hypothalamus and pituitary. Stress increases CRF, ACTH, and glucocorticoid secretion. The expression of not only CRF mRNA in the hypothalamus and proopiomelanocortin mRNA in corticotrophs, but also CRF type 1 receptor (CRF-R1) mRNA and protein on corticotrophs are downregulated through glucocorticoids. However, the mechanisms underlying the glucocorticoid-induced CRF-R1 downregulation are not fully understood. Short RNA molecules, called microRNAs (miRNAs), are posttranscriptional regulators that us

Chronic stress affects the expression of brain-derived neurotrophic factor in rat salivary glands.

Plasma brain-derived neurotrophic factor (BDNF) levels are associated with several neural disorders. Previously, we reported that BDNF is produced from salivary glands under acute immobilization stress. Additionally, salivary glands are the origin of plasma BDNF during stress; however, the association between the expression of BDNF by the salivary glands under chronic stress conditions is not known. In the present study, we investigated whether plasma BDNF levels in chronic stress depend on the salivary glands. Expression of BDNF mRNA and protein were identified in the submandibular glands whe
