Bancroftian filariasis in Pondicherry, south India: 1. Pre-control epidemiological observations.

A 5-year Integrated Vector Management (IVM) project was implemented in Pondicherry, South India, for the control of Bancroftian filariasis. The efficacy of the IVM strategy was compared with routine control strategy under the national programme. The present paper describes the pre-control epidemiological features of filariasis as determined by a mass blood survey in 1981. Of 24946 persons examined 8.41% were microfilaraemic. Microfilaraemia prevalence was homogeneous throughout the study area. The prevalence and intensity of microfilaraemia were age dependent, and increased monotonically until

Demonstration of species-specific and cross-reactive components of Taenia solium metacestode antigens.

Analysis of human serum reactivities to antigenic components of soluble Taenia solium metacestode proteins showed the predominant presence of determinants shared by T. solium, Echinococcus multilocularis and E. granulosus. Two polypeptides were demonstrated by SDS-PAGE and Western blot or enzyme-linked immunoelectrotransfer blot (EITB) assay to bind serum and CSF antibodies only from T. solium cysticercosis patients. These species-specific antigenic polypeptides focused between pH 4.6 and 3.9 after resolution by isoelectric focusing followed by EITB. The high species-specificity demonstrated b

Evaluating the impact of triple-drug mass drug administration using human and mosquito-based indicators of lymphatic filariasis in Samoa, 2019 - 2024.

BACKGROUND: For lymphatic filariasis (LF) elimination, the World Health Organization recommends multiple rounds of mass drug administration (MDA) to interrupt transmission. While LF antigen (Ag) is routinely used to monitor progress, recent evidence suggests more time-sensitive indicators are needed during the immediate post-MDA period. In Samoa, triple-drug MDA was distributed in 2018 and 2023. This study aimed to evaluate the impact of Samoa's second round of triple-drug MDA on human and mosquito-based indicators of transmission. METHODOLOGY: Surveys were conducted in eight primary sampling

Systematic sampling of adults as a sensitive means of detecting persistence of lymphatic filariasis following mass drug administration in Sri Lanka.

BACKGROUND: Sri Lanka's Anti-Filariasis Campaign conducted 5 annual rounds of mass drug administration (MDA) with diethylcarbamazine (DEC) plus albendazole to eliminate lymphatic filariasis (LF) in all endemic districts between 2002 and 2006. Post-MDA surveillance has consistently documented Wuchereria bancrofti microfilaremia (Mf) rates below 1% in all sentinel and spot check sites since that time, and all implementation units easily satisfied WHO's target for school-based transmission assessment surveys (school-TAS) in 2013. However, more detailed studies have identified foci of persistent i

Immunity to filarial nematodes.

Mosquito-borne filarial nematodes cause the severe, debilitating disease of human lymphatic filariasis. In areas endemic for this disease, differential responses range from putative immunity through asymptomatic microfilaraemic infection to chronic pathology. Current research in mouse models of infection is elucidating the immunological mechanisms that can lead to immunity against this disease. In this review, the importance of different immunological pathways are discussed in relation to their role in human disease and in terms of their ability to kill separate developmental stages of the fil

A review of the present status of lymphatic filariasis in Vietnam.

Establishing the current status of an infectious disease forms the starting point of any attempt at parasite control. Although data on the prevalence and distribution of lymphatic filariasis exist for Vietnam from the early 1900s, the present situation regarding the disease is less well-known. Here, we review the results of recent surveys conducted by the Institute of Malariology, Parasitology and Entomology, Hanoi, to update the existing information on filariasis epidemiology and distribution for this country. The present results are from surveys carried out on some 135,000 individuals in 24

Community perception regarding chronic filarial swellings: a case study of the Duruma of coastal Kenya.

The aim of this study, carried out between August 1991 and August 1992, was to understand community perception regarding chronic filarial swellings in Lutsangani village, Kwale district, Kenya. Interviews were conducted with 65 patients and informal discussions held with family and community members. The results indicate that bodily change is the concern of most patients who felt that their physical appearance influenced community reactions towards them. These reactions varied from sympathy to neglect. The community attributed elephantiasis to causes other than mosquitoes and these included wi

Changes in anti-filarial antibody after control of filariasis in Shandong Province.

The changes in anti-filarial antibody level at different periods in different population groups of various controlled areas were studied by indirect fluorescent antibody test (IFT) using frozen sections of Brugia malayi adult worms as antigen. The positive rates by IFAT in formerly hyper-, moderate- and low-endemic areas decreased to the level as those in non-endemic areas 15, 10 and 10 years respectively after the control of filariasis. In the children born after the control of filariasis, the IFAT positive rate was less than 1%. An analysis of IFAT positive cases showed that the positive rat

Approaching onchocerciasis elimination in Equatorial Guinea: Near zero transmission and public health implication.

BACKGROUND: Onchocerciasis and lymphatic filariasis (LF) are endemic in Equatorial Guinea with notable variations in disease incidence between island and mainland regions. Historically, efforts to control and map these diseases were concentrated in Bioko Island, where loiasis is absent, allowing for targeted onchocerciasis interruption strategies. With the cessation of onchocerciasis transmission on Bioko and no reported cases on Annobon island, assessing the transmission status in the previously unaddressed mainland region has become imperative. Mapping efforts in mainland Equatorial Guinea h

Yeast-Based High-Throughput Screens to Identify Novel Compounds Active against Brugia malayi.

BACKGROUND: Lymphatic filariasis is caused by the parasitic worms Wuchereria bancrofti, Brugia malayi or B. timori, which are transmitted via the bites from infected mosquitoes. Once in the human body, the parasites develop into adult worms in the lymphatic vessels, causing severe damage and swelling of the affected tissues. According to the World Health Organization, over 1.2 billion people in 58 countries are at risk of contracting lymphatic filariasis. Very few drugs are available to treat patients infected with these parasites, and these have low efficacy against the adult stages of the wo

The mitochondrial genomes of Culex tritaeniorhynchus and Culex pipiens pallens (Diptera: Culicidae) and comparison analysis with two other Culex species.

BACKGROUND: Culex tritaeniorhynchus and Culex pipiens pallens are the major vectors of the Japanese encephalitis virus and Wuchereria bancrofti, the causative agent of filariasis. The knowledge of mitochondrial genomes has been widely useful for the studies on molecular evolution, phylogenetics and population genetics. METHODS: In this study, we sequenced and annotated the mitochondrial (mt) genomes of Cx. tritaeniorhynchus and Cx. p. pallens, and performed a comparative analysis including four known mt genomes of species of the subgenus Culex (Culex). The phylogenetic relationships of Cx. tr

Midgut barrier imparts selective resistance to filarial worm infection in Culex pipiens pipiens.

Mosquitoes in the Culex pipiens complex thrive in temperate and tropical regions worldwide, and serve as efficient vectors of Bancroftian lymphatic filariasis (LF) caused by Wuchereria bancrofti in Asia, Africa, the West Indies, South America, and Micronesia. However, members of this mosquito complex do not act as natural vectors for Brugian LF caused by Brugia malayi, or for the cat parasite B. pahangi, despite their presence in South Asia where these parasites are endemic. Previous work with the Iowa strain of Culex pipiens pipiens demonstrates that it is equally susceptible to W. bancrofti

Integrated mapping of neglected tropical diseases: epidemiological findings and control implications for northern Bahr-el-Ghazal State, Southern Sudan.

BACKGROUND: There are few detailed data on the geographic distribution of most neglected tropical diseases (NTDs) in post-conflict Southern Sudan. To guide intervention by the recently established national programme for integrated NTD control, we conducted an integrated prevalence survey for schistosomiasis, soil-transmitted helminth (STH) infection, lymphatic filariasis (LF), and loiasis in Northern Bahr-el-Ghazal State. Our aim was to establish which communities require mass drug administration (MDA) with preventive chemotherapy (PCT), rather than to provide precise estimates of infection pr

Precontrol observations on lymphatic filariasis & geo-helminthiases in two coastal districts of rural Orrisa.

BACKGROUND & OBJECTIVE: Lymphatic filariasis (LF) is a major public health problem in India, accounting for 40 per cent of the global burden. The World Health Organization has launched a global programme to eliminate LF by 2020 and India is a signatory to it. Orissa, an eastern Indian State has long been known to be endemic for LF. Prior to implementation of mass drug administration programme it is important to collect baseline data on filariasis and geo-helminthiases in the State. The present cross-sectional survey was therefore carried out between February and December 2001 to obtain baselin

Unraveling cross-reactivity of anti-glycan IgG responses in filarial nematode infections.

Parasitic nematodes responsible for filarial diseases cause chronic disablement in humans worldwide. Elimination programs have substantially reduced the rate of infection in certain areas, but limitations of current diagnostics for population surveillance have been pointed out and improved assays are needed to reach the elimination targets. While serological tests detecting antibodies to parasite antigens are convenient tools, those currently available are compromised by the occurrence of antibodies cross-reactive between nematodes, as well as by the presence of residual antibodies in sera yea
