Impulse control disorders in Parkinson's disease: seeking a roadmap toward a better understanding.

The development of an impulse control disorder (ICD) is now recognized as a potential nonmotor adverse effect of dopamine replacement therapy in Parkinson's disease (PD). Here, recent epidemiological, neurophysiological and genetic advances are summarized to outline potential mechanisms involved. It is safe to say that dopaminergic drugs, particularly dopamine agonists, are able to induce ICDs only in a minority of patients, while the majority are somehow protected from this adverse effect. While it seems clear that men with early-onset PD are more vulnerable, other predisposing factors, such

Renewal and spontaneous recovery, but not latent inhibition, are mediated by gamma-aminobutyric acid in appetitive conditioning.

Previous research has reported a role for the neurotransmitter gamma-aminobutyric acid (GABA) in the extinction and renewal of conditioned fear. Here, the authors examine whether GABA is involved in the acquisition, extinction, renewal, spontaneous recovery, and latent inhibition of appetitive conditioning. Using Long-Evans rats, systemic injection of the GABA A receptor inverse agonist FG 7142 was shown to eliminate ABA renewal (Experiment 1) and spontaneous recovery (Experiment 4) of appetitive responding by selectively reducing the recovery of extinguished magazine approach. Furthermore, tr

Sibutramine sensitivity assay revealed a unique phenotype of bombesin BB3 receptor-deficient mice.

Sibutramine sensitivity assay in genetically obese (bombesin BB3 receptor (BRS-3)-deficient mice, KK-Ay mice, db/db mice and Zucker obese rat) and wild-type animals was examined. The sensitivity of Sibutramine (10 mg/kg, p.o.) in BRS-3-deficient mice was retained as well as normal animals; however, it was decreased in KK-Ay, db/db mice and Zucker obese rat. The suppression values of food intake in BRS-3-deficient, KK-Ay, db/db mice and Zucker obese rat were 49.8+/-5.8%, 16.1+/-4.7%, 0.1+/-2.8% and -2.0+/-2.2% (mean +/- S.E.), respectively. Next, we found that the contribution of hyperphagia wa

Does neuropeptide Y contribute to the anorectic action of amylin?

Neuropeptide Y (NPY) is a potent feeding stimulant acting at the level of the hypothalamus. Amylin, a peptide co-released with insulin from pancreatic beta cells, inhibits feeding following peripheral or central administration. However, the mechanism by which amylin exerts its anorectic effect is controversial. This study investigated the acute effect of amylin on food intake induced by NPY, and the effect of chronic amylin administration on food intake and body weight in male Sprague Dawley rats previously implanted with intracerebroventricular (icv) cannulae. Rats received 1 nmol NPY, follow

Hepato-vagal pathway associated with nicotine's anorectic effect in the rat.

Nicotine reduces appetite and body weight. Because the hepato-portal area senses different types of nutrients that transmit signals via vagal afferent nerves to the hypothalamus to modify food intake and feeding pattern, we investigated the effect of nicotine on a hepato-vagal-hypothalamic pathway. Low doses of nicotine (< 10 ng) injected into portal vein (i.p.v.) decreased, while high doses of nicotine increased (> or = 10 ng) electrophysiological activity of hepatic vagal afferents. Stimulatory effect of high dose of nicotine on vagal hepatic afferents was blocked by a prior i.p.v. injection

Acquisition and retention by hippocampal rats of simple, conditional, and configural tasks using tactile and olfactory cues: implications for hippocampal function.

Hippocampal rats were given simple, conditional, and configural tasks that required them to pull up strings to obtain food pellets. The tasks contrasted string size, odor, location, paired string size and location, or compounds of string size and odor. There was no anterograde or retrograde amnesia for the simple discriminations or for simple discrimination components of the other tasks. Hippocampal rats also learned conditional and configural tasks, although they were impaired on the configural task. Finally, the rats were impaired in postoperative retention of the conditional but not the con

Rotation following intranigral injections of a selective D1 or a selective D2 dopamine receptor agonist in rats.

Injections of various nonselective dopamine agonists into the substantia nigra, pars reticulata (SNpr), have been reported to produce contralateral rotation in rats. Since a number of recent dopamine receptor distribution studies have indicated a preponderance of D1 compared to D2 dopamine receptor subtypes within the SNpr, we examined the relative behavioral functions of these two subtypes within the nigra by studying rotation following unilateral, local injections of a D1 (SKF38393) and D2 (quinpirole) agonist, Significant, dose-dependent contralateral rotation was observed following injecti

Behavioral effects of chronic exposure to selective D-1 and D-2 dopamine receptor agonists.

Chronic treatment with the selective D-1 agonist SKF 38393 induced behavioral supersensitivity (increased stereotypy and decreased locomotor activity) in response to apomorphine, In contrast, chronic treatment with the selective D-2 agonist LY 171555 resulted in a subsensitive behavioral response to apomorphine challenge. Chronic treatment with the combination of these drugs augmented apomorphine-induced stereotypic behaviors, but these were different in nature from those observed in animals treated with SKF 38393 alone. Chronic SKF 38393 treatment resulted in an enhanced behavioral response t

Three anorectic drugs: similar structures but different effects on brain and behavior.

Neurochemical probes are starting to tease apart several neurotransmitter systems that are involved in suppressing feeding. Instead of a classical medial hypothalamic lesion, neurotoxins or neurochemical inhibitors can be used to produced animal models of the obese. These models make it clear that amphetamine and fenfluramine do not act in the same way. In addition to actions on brain synapses, these drugs may act directly or indirectly on the receptors that monitor energy availability. Phenylpropanolamine (PPA) apparently has anorectic action within the lateral hypothalamus, but this could be

Cannabinoid treatment for autism: a proof-of-concept randomized trial.

BACKGROUND: Endocannabinoid dysfunction in animal models of autism spectrum disorder (ASD) and accumulating, albeit anecdotal, evidence for efficacy in humans motivated this placebo-controlled double-blind comparison of two oral cannabinoid solutions in 150 participants (age 5-21 years) with ASD. METHODS: We tested (1) BOL-DP-O-01-W, a whole-plant cannabis extract containing cannabidiol and Δ9-tetrahydrocannabinol at a 20:1 ratio and (2) BOL-DP-O-01, purified cannabidiol and Δ9-tetrahydrocannabinol at the same ratio. Participants (N = 150) received either placebo or cannabinoids for 12-weeks

The accelerator, the brake, and the terrain: associations of reward-related eating, self-regulation, and the home food environment with diet quality during pregnancy and postpartum in the pregnancy eating attributes study (PEAS) cohort.

BACKGROUND: Neurobehavioral factors, including reward-related eating and self-regulation, in conjunction with the food environment, may influence dietary behaviors. However, these constructs have not been examined in pregnancy and postpartum, a time of changing appetite and eating behaviors, and when dietary intake has implications for maternal and child health. This study examined associations of reward-related eating, self-regulation, and the home food environment with pregnancy and postpartum diet quality. METHODS: Participants in the Pregnancy Eating Attributes Study observational cohort

Transcriptional involvement of protein kinase C-alpha isozyme in amphetamine-mediated appetite suppression.

Amphetamine (AMPH) is known as an anorectic agent. The anorectic action of AMPH has been attributed to its inhibitory action on hypothalamic neuropeptide Y (NPY), an appetite stimulant in the brain. The molecular mechanisms behind this anorectic action of AMPH are still unclear. This study investigated the possible role of protein kinase C (PKC) isotypes in this anorectic action. Results revealed that most PKC isotypes (alpha, betaII, gamma, delta, eta, lambda and zeta), except betaI and epsilon isotypes, were stimulated during a repeated treatment of AMPH. Among these stimulated isotypes, thr

Effect of offering a second colostrum feeding and delivery method on transfer of passive immunity, health, and behavior in neonatal calves.

Our objective was to describe the effect of providing a second feeding of maternal colostrum (MC) or colostrum replacement (CR), using either an esophageal tube or nipple bottle delivery method, on transfer of passive immunity, and on measures of calf health and behavior during the first 3 d of life, as compared with providing a milk replacer control. Eligible newborn dairy and dairy x beef calves on a commercial Minnesota dairy were weighed, placed in a bedded pen, then fitted with a tri-axial accelerometer. All calves were tube-fed a first feeding of pooled heat-treated MC (average IgG = 84

Effect of continuous energy restriction combined with probiotic supplementation on parameters related to homeostatic and hedonic systems of food intake in adult men living with obesity: Data from a randomized, parallel, double-blind, placebo-controlled clinical trial.

BACKGROUND AND AIM: Continuous energy restriction (CER) effectively promotes body fat reduction; however, its effects on hunger regulation and the hedonic system (reward-related mechanisms) may compromise long-term weight maintenance. The gut microbiota has emerged as a potential target to support sustained weight loss; however, studies evaluating combined interventions, such as probiotics alongside CER, remain limited. This study aimed to evaluate whether probiotic supplementation enhanced the metabolic and behavioral effects of CER by examining appetite-related hormones (ghrelin, leptin, GLP

Further pharmacological characterization of eltoprazine: focus on its anxiolytic, anorexic, and adverse‑effect potential.

Eltoprazine, a drug that had previously been developed for aggression, has recently been investigated for L-DOPA-induced dyskinesia in animal models of Parkinson´s disease (PD) and in dyskinetic PD patients. Much less is known about effects of eltoprazine in other therapeutic indications. Indeed, the pharmacological profile of eltoprazine might suggest its effects on anxiety and food intake, but also adverse effect potential, which is the focus of the present study. Given for 2 weeks either as infusion or as twice-daily treatment, eltoprazine produced a decrease in food intake and body weight

Acute anorectic response to cannabinoid CB1 receptor antagonist/inverse agonist AM 251 in rats: indirect behavioural mediation.

Despite a large and consistent literature on the suppressant effects of cannabinoid CB1 receptor antagonists/inverse agonists (e.g. rimonabant, AM 251) on food intake and weight gain in rodents, surprisingly little is known about the behavioural selectivity of such effects. In this study, ethological scoring was used to characterize the acute behavioural effects of the rimonabant analogue AM 251 (1.5 and 3.0 mg/kg, intraperitoneally) in nondeprived male rats during a 1-h test with palatable mash. Data were also collected on daily weight gain and on retest food intake 7 days after dosing. Resul

Estradiol-mediated increases in the anorexia induced by intraperitoneal injection of bacterial lipopolysaccharide in female rats.

Lipopolysaccharide (LPS) derived from the cell walls of gram-negative bacteria causes a robust acute phase response (APR) that includes fever, anorexia, and many other elements. Because immune system function, including some models of illness anorexia, is sexually differentiated, we investigated the sexual differentiation of the anorexia induced by intraperitoneal LPS injections in rats. Cycling female Long-Evans rats tested either during diestrus or estrus ate less following 6.25 microg/kg LPS than did intact males. Following 12.5 microg/kg LPS, females in estrus ate less than either females
