Molecular basis of familial growth hormone deficiency.

A significant proportion of cases of GH deficiency (5-30%) may be due to genetic causes. At least four Mendelian types of isolated GH deficiency (IGHD) have been delineated based on the mode of inheritance and the degree of GH deficiency: IGHD type IA, autosomal recessive with absent endogenous GH; type IB, autosomal recessive with diminished GH; type II, autosomal dominant with diminished GH; and type III, X-linked with diminished GH. Most patients with IGHD type IA have heterogeneous deletions, ranging in size from 6.7 kb to 45 kb, that encompass the entire gene encoding for pituitary GH, GH

The EXTra-uterine Environment for Neonatal Development (EXTEND) - future treatment for prematurity and potentially other congenital disease.

The EXtra-uterine Environment for Neonatal Development (EXTEND) was developed to support extremely premature infants (EPIs) with the goal to mimic in-utero conditions as closely as possible and therefore reduce morbidity and mortality associated with organ immaturity and iatrogenic injury caused by current neonatal management. In an animal model using fetal lambs at a gestational age equivalent to EPIs of 22-24 weeks of gestation, EXTEND was able to support fetuses for up to four weeks while enabling normal organ growth, particularly normal lung development. Recent advances in EXTEND have allo

Sodium taurocholate cotransporter polypeptide deficiency combined with novel PYGL mutations in glycogen storage disease type VI: a rare case report.

Both Sodium Taurocholate Cotransporting Polypeptide Deficiency (NTCPD) and Glycogen Storage Disease Type VI (GSD-VI) are autosomal recessive (AR) genetic disorders that affect liver metabolism in newborns. NTCPD is primarily caused by mutations in the Solute Carrier Family 10 Member 1 (SLC10A1) gene, resulting in decreased bile acid transport function, while GSD-VI is caused by mutations in the phosphorylase glycogen liver (PYGL) gene, leading to a deficiency of liver glycogen phosphorylase. Both diseases are rare, and there have been no previous reports of their coexistence in a single patien

Dynamic subtype- and context-specific subcellular RNA regulation in growth cones of developing neurons of the cerebral cortex.

Neurons of distinct subtypes compartmentalize subtype-specific function in part by differentially localizing and translating specific RNAs, but underlying mechanisms are not understood. Here we investigate messenger RNA localization and stability within subtype-specific growth cones (GCs), leading tips of growing axons, of long-range projection neurons (PNs) of the developing cerebral cortex. Comparison of GC-localized transcriptomes between two subtypes of PNs (interhemispheric-callosal and corticothalamic) across developmental stages identified both distinct and shared subcellular machinery

Tissue engineering approaches for lymphedema: biomaterial innovations and clinical potential.

The lymphatic system plays a critical role in maintaining fluid balance and immune regulation. Lymphedema, and other lymphatic disorders, highlight the need for advanced therapeutic approaches, including tissue engineering. This review examines the latest developments in artificial lymphatic tissue engineering, focusing on scaffold materials, lymphangiogenic factors, and regenerative strategies to replicate the intricacy of lymphatic vessels and nodes. We conducted a thorough literature review of current practices and applications in lymphatic tissue engineering. Findings show that biomaterial

Metabolic functions for molecular chaperones and stress proteins in microcephaly.

Neurometabolic disruptions during brain development are central drivers of the structural and functional impairments observed in neurodevelopmental disorders such as microcephaly. While these deficits are often attributed to inherited mutations in metabolic enzymes, emerging evidence highlights the critical role of cellular stress response pathways, particularly those governed by proteostatic machinery, such as molecular chaperones. Although molecular chaperones support brain growth through diverse mechanisms, this review focuses on their role in stabilising metabolic enzymes, preventing their

Intrauterine growth restriction: the controversial role of perinatal adipocytokines in the prediction of metabolic adult disease.

"Prenatal/fetal programming," implying structural/functional disorders of developing tissues/organs, consequent to an adverse intrauterine environment leading to asymmetric intrauterine growth restriction (IUGR), predisposes to metabolic syndrome and noncommunicable diseases in adulthood, in the framework of the "Developmental Origins of Health and Disease" (DOHaD) concept. DOHaD consequences are associated with adipose tissue, particularly the visceral one, occurring in relative abundance in IUGR infants. Adipose tissue secretes numerous hormones, collectively called adipocytokines, as leptin

Neurotrophic natural products: chemistry and biology.

Neurodegenerative diseases and spinal cord injury affect approximately 50 million people worldwide, bringing the total healthcare cost to over 600 billion dollars per year. Nervous system growth factors, that is, neurotrophins, are a potential solution to these disorders, since they could promote nerve regeneration. An average of 500 publications per year attests to the significance of neurotrophins in biomedical sciences and underlines their potential for therapeutic applications. Nonetheless, the poor pharmacokinetic profile of neurotrophins severely restricts their clinical use. On the othe

Angiogenesis and lymphangiogenesis in bronchial asthma.

Neovascularization plays a prominent role in inflammation and tissue remodeling in several chronic inflammatory disorders. Vessel number and size, vascular surface area and vascular leakage are all increased in biopsies from patients with asthma. High levels of VEGF and other angiogenic factors have been detected in tissues and biological samples of patients with asthma and correlate with disease activity and inversely with airway hyper-responsiveness. Inflammation in the lung stimulates the growth of new blood vessels and these contribute to the airway obstruction or airway hyper-responsivene

Interactions between growth hormone and the thyroid gland -- with special reference to biochemical diagnosis.

The diagnosis of pituitary disorders is difficult because several hormone systems are involved. The clinical presentation is often vague and slowly progressing, and clinicians therefore have to rely very much on correct biochemical measurements. This is also associated with difficulties since the pituitary hormones interact, the binding proteins are influenced by the other axes and a variety of other effects, and finally the hormone measurements in serum are not totally adequate Several studies have investigated the interaction between the thyroid and growth hormone axes with very variable res

[The role of static-dynamic factors in genesis of femoral head and neck deformation in dysplastic hips. A physical-mathematical interpretation ].

The clinical material for a physical-mathematical analysis of femoral head and neck deformations in subluxated and luxated hips consisted of 1.574 case histories of children operated on in 1954-1988. In that material 21.9 per cent of the children had preoperative growth disorders of the head, and in the half of the cases also the femoral neck was involved. In 10.79 per cent of the children pre- and postoperative trophic disorders were also observed. On the basis of the physical-mathematical analysis of static-dynamic factors of the hip, it has been determined that the disordered shape of the f…

The neurocristopathies: reinterpretation based upon the mechanism of abnormal morphogenesis.

This review sets forth a broadened interpretation of the neurocristopathies based on the current understanding of the role of neural crest cells in normal development. Two general types of cristopathies are defined predicated on the abnormal mechanism involved in production of the defect or condition. Defects and disorders which constitute the originally defined neurocristopathies including pheochromocytoma, neurofibromatosis, and the multiple endocrine adenomatoses are best explained as dysplasias of neural crest derivatives. Affected individuals rarely exhibit true malformation of structure

Inflammatory biomarkers and cognitive functioning in individuals with euthymic bipolar disorder: exploratory study.

BACKGROUND: Neurobiological research frequently implicates inflammatory and neurogenic components with core aspects of bipolar disorder. Even in periods of symptom remission (euthymia), individuals with bipolar disorder experience cognitive impairments, which are increasingly being proposed as an outcome for interventions; identifying biomarkers associated with cognitive impairment in people with bipolar disorder could advance progress in this therapeutic field through identifying biological treatment targets. AIMS: We aimed to identify proteomic biomarker correlates of cognitive impairment i

Crosstalk between the growth hormone/insulin-like growth factor-1 axis and the gut microbiome: A new frontier for microbial endocrinology.

Both the GH/IGF-1 axis and the gut microbiota independently play an important role in host growth, metabolism, and intestinal homeostasis. Inversely, abnormalities in GH action and microbial dysbiosis (or a lack of diversity) in the gut have been implicated in restricted growth, metabolic disorders (such as chronic undernutrition, anorexia nervosa, obesity, and diabetes), and intestinal dysfunction (such as pediatric Crohn's disease, colonic polyps, and colon cancer). Over the last decade, studies have demonstrated that the microbial impact on growth may be mediated through the GH/IGF-1 axis,

Progress in Research on the Role of FGF in the Formation and Treatment of Corneal Neovascularization.

Corneal neovascularization (CNV) is a sight-threatening disease usually associated with inflammatory, infectious, degenerative, and traumatic disorders of the ocular surface. Fibroblast growth factor (FGF) family members play an important role in angiogenesis to induce corneal neovascularization, which significantly affects the differentiation, proliferation, metastasis, and chemotaxis of vascular endothelial cells. Both acidic fibroblast growth factor (aFGF) and basic fibroblast growth factor (bFGF) demonstrate positive staining in capillaries and induce corneal stromal cells. The anabolism o

[De Sanctis-Cacchione syndrome].

We present a male patient with photosensitivity since the earliest months of his life, and pigmented macules in exposed areas, some showing clinical atypia, which increased in number over time. Molecular biology studies detected an alteration in DNA repair ability, so xeroderma pigmentosum was diagnosed. Shortly after birth, low weight, microcephaly and psychomotor retardation had been observed, but the cause was not established. The patient progressively showed neurological disorders that included perceptive deafness, hyporeflexia and areflexia, as well as choreoathetotic movements. Therefore
