We examined the effect of pranlukast, the receptor antagonist of the cysteinyl leukotrienes (CysLTs; LTC(4), LTD4 and LTE4), on indomethacin-induced small intestinal lesions in rats. Animals non-fasted were given indomethacin (10 mg/kg) s.c., and killed 24 hr later. Pranlukast (1-10 mg/kg) was given p.o. twice, 30 min before and 6 hr after the administration of indomethacin. A single s.c. administration of indomethacin provoked multiple haemorrhagic lesions in the small intestine, mainly in the jejunum and ileum. This treatment also caused an increase in MPO activity, microvascular permeabilit

Cholera is a potentially fatal bacterial infection caused by the cholera toxin (CT), an AB5 toxin secreted by Vibrio cholera. GM1 has long been known as the receptor of the cholera toxin in the intestine. However, increasing evidence is pointing towards the role of fucosylated conjugates as additional attachment options of the toxin. In the present paper we have synthesized a polymeric hybrid which can inhibit both modes of attachment.

The immune system shapes body metabolism, while interactions between peripheral neurons and immune cells control tissue homeostasis and immunity. However, whether peripheral neuroimmune interactions orchestrate endocrine system functions remains unexplored. After fasting, mice lacking type 2 innate lymphoid cells (ILC2s) displayed disrupted glucose homeostasis, impaired pancreatic glucagon secretion, and inefficient hepatic gluconeogenesis. Additionally, intestinal ILC2s were found in the pancreas, which was dependent on their expression of the adrenergic beta 2 receptor. Targeted activation o

The ascidian Styela clava is an invasive species that easily colonizes new water areas and has a wide distribution. A previously undescribed typhlosole was discovered in S. clava and studied using histological techniques, transmission electron microscopy and computer microtomography. The typhlosole is a large fold of the dorsal intestinal wall that starts in the lower quarter of the stomach and continues to the rectum. Its shape varies greatly: from a rounded protrusion to a spirally twisted fold. The typhlosole is formed by the intestinal epithelium and a thick connective tissue layer, in whi

Postprandial hypotension (PPH) is an important and under-recognised disorder resulting from inadequate compensatory cardiovascular responses to meal-induced splanchnic blood pooling. Current approaches to management are suboptimal. Recent studies have established that the cardiovascular response to a meal is modulated profoundly by gastrointestinal factors, including the type and caloric content of ingested meals, rate of gastric emptying, and small intestinal transit and absorption of nutrients. The small intestine represents the major site of nutrient-gut interactions and associated neurohor

Hypothalamic neuropeptides named hypocretin/orexins which were identified in 1998 regulate critical functions such as wakefulness in the central nervous system. These past 20 years had revealed that orexins/receptors system was also present in the peripheral nervous system where they participated to the regulation of multiple functions including blood pressure regulation, intestinal motility, hormone secretion, lipolyze and reproduction functions. Associated to these peripheral functions, it was found that orexins and their receptors were involved in various diseases such as acute/chronic infl

Guanylyl cyclase C (GC-C) is the receptor for the heat-stable enterotoxin, which causes diarrhea, and the endogenous ligands, guanylin and uroguanylin. GC-C is predominantly expressed in the intestinal epithelium and regulates fluid and ion secretion in the gut. The receptor has a complex domain organization, and in the absence of structural information, mutational analysis provides clues to mechanisms of regulation of this protein. Here, we review the mutational landscape of this receptor that reveals regulatory features critical for its activity. We also summarize the available information o

Bariatric surgery is increasingly recognized as one of the most effective interventions to help patients achieve significant and sustained weight loss, as well as improved metabolic and overall health. Unfortunately, the cellular and physiological mechanisms by which bariatric surgery achieves weight loss have not been fully elucidated, yet are critical to understanding the central role of the intestinal tract in whole-body metabolism and to developing novel strategies for the treatment of obesity. In this review, we provide an overview of potential mechanisms contributing to weight loss, incl

Inflammatory bowel disease (IBD) is characterized by inflammatory reactions, complications, extraintestinal manifestations and a loss of intestinal functions, for example, failures of absorption and secretion. According to intestinal dysfunction, a wide array of pathogenetic pathways is existing leading to iron deficiency and numerous vitamins as well as trace element deficiencies. Complications, symptoms and signs of those deficiencies are common in IBD with varying degrees of clinical significance. This review focuses on selected micronutrients including iron, zinc, magnesium and some vitami

In contrast to gastric dysfunction, diabetes-related functional impairments of the small and large intestine have been studied less intensively. The gastrointestinal tract accomplishes several functions, such as mixing and propulsion of luminal content, absorption and secretion of ions, water, and nutrients, defense against pathogens, and elimination of waste products. Diverse functions of the gut are regulated by complex interactions among its functional elements, including gut microbiota. The network-forming tissues, the enteric nervous system) and the interstitial cells of Cajal, are defini

Intestinal ulcerogenic properties of NSAIDs require the inhibition of both COX-1 and COX-2, and inhibition of COX-1 up-regulates COX-2 expression and PGs produced by COX-2 counteract the deleterious influences of the COX-1 inhibition. These lesions are prevented by PGE2 mediated by EP3/EP4 receptors and functionally associated with stimulation of mucus secretion, inhibition of intestinal contraction or enterobacterial invasion, and down-regulation of cytokine expression. COX/PGE2 also plays an important role in the healing of these lesions, but the COX isozyme responsible for PG production dif

The study describes the immunohistological localization of the extracellular CuZn superoxide dismutase (SOD2) in the parasitic nematode Onchocerca volvulus. Using specific antiserum raised against a 15-amino-acid peptide from the N-terminal region of the mature protein, this enzyme is detected primarily in the intestinal epithelium of the adult worms and to a lesser extent in the muscle cells of the uterine wall. A blocking experiment with the SOD2 peptide reduced the staining significantly, confirming specificity. The localization profile of SOD2 correlates extremely well with the localizatio

The effects of three bacterial pathogens on the villus architecture of small intestines and the role that bacterial virulence factors play in pathogenesis are described. Bacterial pathogens cause a spectrum of effects ranging from severe tissue damage to a lack of perceptible damage. Enterotoxigenic Escherichia coli, which cause acute and severe diarrhea, does so by producing potent toxins, but these toxins act by altering the biological activity in epithelial cells. However, the cells are not damaged. Enteropathogenic E. coli and Salmonella, on the other hand cause various degrees of tissue d

Histochemical examinations of 28 cancers of Vater's papilla detected argirophilic cells in 7 (25%) and argentaffinic cells in 3 (10.8%) tumors. Neoplastic endocrine cells occurred most frequently in structures of the highly differentiated papillary and tubular adenocarcinoma. The analysis of the qualitative composition of mucus produced by these tumors showed the likelihood of detection of endocrine cells in cancer of Vater's papilla to increase with higher secretion of acid mucopolysaccharides and to decrease with a higher portion of sulphomucines in them. The occurrence of impregnated endocr

Groups of athymic rnu rats were found to be susceptible to SAW 408 Entamoeba histolytica introduced intra-intestinally. The same strain of rat could be infected orally with cultured trophozoites using a gastric secretion inhibitor, cimetidine, as a pretreatment. Groups of Lac:P and Wistar rats similarly treated were also found to be susceptible although the parasite population growth was slow. A similar regimen attempted on susceptible mice was unsuccessful.

Type 2 diabetes mellitus is associated with gut dysbiosis, decreased microbial diversity, short-chain fatty acid synthesis, and altered GLP secretion, which are crucial for intestinal integrity, insulin sensitivity, and postprandial glucose control. Evidence from peer-reviewed mechanistic studies, observational research, clinical trials, and meta-analyses was summarised in this review. Prebiotics, an emerging potential treatment, increase the formation of SCFA during fermentation, thereby enhancing the release of GLP through FFAR2/3 signalling. This chain of events enhances glucose-dependent i

The intestinal epithelium poses a formidable obstacle to the systemic absorption of the oral nanovehicles. Despite the development of extracellular vesicles (EVs) for drug delivery, there has been limited exploration into edible-product-derived EVs (EP-EVs), particularly those derived from plants as carriers to enhance the oral delivery of biomacromolecules. Here, we evaluated the potential of EP-EVs and highlighted their promising application and underlying mechanisms as orally delivered carriers from plant-derived EVs. Grape-derived EVs (Gra-EVs) were found to have superior efficacy in oral

Intervention of the gut microbiome is a promising adjuvant strategy in cancer immunotherapy. Chemotherapeutic agents are recognized for their substantial impacts on the gut microbiome, yet their therapeutic potential as microbiome modulators remains uncertain, due to the complexity of microbiome-host-drug interactions. Here, it is showed that low-dose chemotherapy preferentially shapes the ileal microbiome to augment the extraintestinal immune response to anti-programmed death-1 (anti-PD-1) therapy without causing intestinal toxicity. Mechanistically, low-dose chemotherapy causes DNA damage re

Pancreatic cancer (PC) is a dangerous digestive tract tumor that is becoming increasingly common and fatal. The most common form of PC is pancreatic ductal adenocarcinoma (PDAC). Bile acids (BAs) are closely linked to the growth and progression of PC. They can change the intestinal flora, increasing intestinal permeability and allowing gut microbes to enter the bloodstream, leading to chronic inflammation. High dietary lipids can increase BA secretion into the duodenum and fecal BA levels. BAs can cause genetic mutations, mitochondrial dysfunction, abnormal activation of intracellular trypsin,

Inclusion of additive blends is a common dietary strategy to manage post-weaning diarrhea and performance in piglets. However, there is limited mechanistic data on how these additives improve outcomes during this period. To evaluate the effects of Presan FX (MCOA) on the intestinal microbiota and metabolome, diets with or without 0.2% MCOA were compared. Pigs fed MCOA showed improved whole-body metabolism 7 days post-weaning, with decreased (P < 0.05) creatine, creatinine and β-hydroxybutyrate. Alterations in bile-associated metabolites and cholic acid were also observed at the same time-point
