Probing the phosphoinositide binding site of the clathrin assembly protein AP-2 with photoaffinity labels.

The relative binding specificities of the subunitsof bovine assembly protein AP-2 for the phosphatidylinositol polyphosphates (PtdInsPn) and inositol polyphosphates (InsPn) were determined by photoaffinitylabeling. Three types of benzophenone-containing photoprobes were employed: (i) the water-solubleP-1- or P-2-tethered p-benzoyldihydrocinnamoyl-InsPn (BZDC-InsPn) analogs, (ii) P-1-linked phosphotriester PtdInsPn analogs that sampled the interface between the water and lipid phases, and (iii) sn-1-O-acyl-linked PtdInsPn analogs that interacted with proteins penetrating the bilayer. The InsPn

Further mechanistic study on intestinal absorption enhanced by unsaturated fatty acids: reversible effect by sulfhydryl modification.

In order to study the relationship between the sulfhydryl (SH) modification of membrane-associated proteins and the oleic acid-induced permeability enhancement of the colonic mucosa, in vitro and in situ absorption studies were performed using rat colon and carboxyfluorescein as an impermeable dye. The pretreatment of the mucosa with diamide, a bifunctional sulfhydryl modifier, in in vitro experiments with the everted colonic loops reduced the absorption enhancing effect of oleic acid in a concentration-dependent manner, less inhibitory effect, though just a little, was observed as compared to

Lipid-Inspired Low Melting Ionic Liquids via Synergistic Cyclopropanation and Branching of Terpenoids.

Bacteria employ cyclopropane motifs as bioisosteres for unsaturations to modulate lipid bilayer fluidity and protect cellular membranes under environmental stress. Drawing inspiration from this biological strategy, we investigated how cyclopropanation impacts the thermophysical properties of lipid-inspired ionic liquids. We synthesized a series of imidazolium-based ionic liquids incorporating cyclopropanated derivatives of three renewable terpenoids: phytol, farnesol, and geraniol. Through an integrated approach combining property-driven design, thermophysical analysis, X-ray crystallography,

N-Palmitoylglycine and other N-acylamides activate the lipid receptor G2A/GPR132.

The G-protein-coupled receptor GPR132, also known as G2A, is activated by 9-hydroxyoctadecadienoic acid (9-HODE) and other oxidized fatty acids. Other suggested GPR132 agonists including lysophosphatidylcholine (LPC) have not been readily reproduced. Here, we identify N-acylamides in particular N-acylglycines, as lipid activators of GPR132 with comparable activity to 9-HODE. The order-of-potency is N-palmitoylglycine > 9-HODE ≈ N-linoleoylglycine > linoleamide > N-oleoylglycine ≈ N-stereoylglycine > N-arachidonoylglycine > N-docosehexanoylglycine. Physiological concentrations of N-acylglycines

pH-triggered liposomal delivery of alendronate enables acid-responsive drug release and improves osteoblast viability.

Bisphosphonates, such as sodium alendronate (ALN), are primary treatments for osteoporosis and osteosarcoma; however, their long-term use is limited by causing severe side effects. A pH-sensitive liposomal formulation was developed to enhance ALN delivery, enabling controlled drug release specifically in acidic biological microenvironments and niches. Release efficiency of pH-sensitive liposomes was evaluated and compared with that of standard and PEGylated liposomes. Molecular dynamics simulations demonstrated the stability and structural behavior of liposome formulations under physiological

Thermally Modulated Metasurface Sensor for Dynamic and Time-Resolved Isolation of Extracellular Vesicles.

Extracellular vesicles (EVs) are nanoscale lipid-bilayered structures that mediate intercellular communications by transporting nucleic acids, proteins, and lipids across diverse biological fluids. Their diagnostic potential is immense, yet their heterogeneity poses persistent challenges for isolation and characterization, often leading to low yield, co-isolation of contaminants, and vesicle damage. Here, we present a thermoresponsive polymer-integrated plasmonic metasurface sensor that enables spatiotemporally controlled, label-free EV isolation. The metasurface, engineered by repurposing nan

Bacterial Extracellular Vesicles at the Crossroads of Immune Regulation and Biofilm Dynamics: Biogenesis, Comparative Analysis, and Translational Challenges.

Bacterial extracellular vesicles (BEVs) are nano-sized lipid bilayer particles secreted by bacteria, capable of carrying various proteins, lipids, nucleic acids, and pathogen-associated molecular patterns (PAMPs). The biosynthetic pathway of BEVs determines their load components, physicochemical properties, and different biological activities. Increasing evidence indicates that BEVs play an important role in mediating host immune responses and the dynamic regulation of bacterial biofilms. Additionally, BEVs may serve as a molecular bridge between the two. BEVs derived from pathogens can trigge

TPGS-functionalized liposomal formulation for the transdermal delivery of ellagic acid.

The pursuit of effective anti-aging interventions remains a significant challenge in dermatological research. Ellagic acid (EA), a natural polyphenol with demonstrated anti-skin-aging properties, faces limitations in therapeutic application due to poor percutaneous absorption. To address this challenge, we developed TPGS-functionalized liposomes (EA-TPGS-L) through systematic optimization using a Box-Behnken design, achieving an encapsulation efficiency of 94.19% and an optimal particle size of 152.50 nm. MD simulations revealed that TPGS enhances skin permeation by spontaneously inserting its

Extracellular vesicle signalling in perioperative neurocognitive disorders.

Perioperative neurocognitive disorders (PND), including postoperative delirium, delayed neurocognitive recovery, and postoperative neurocognitive disorder, are important complications in older and vulnerable surgical patients. These conditions are associated with prolonged hospitalization, reduced functional recovery, greater healthcare utilization, and worse longer-term outcomes. Current evidence indicates that PND is not driven by a single mechanism. Instead, systemic inflammation, neurovascular dysfunction, blood-brain barrier injury, glial activation, innate immune signalling, and synaptic

Physical-physiological coupling underlies resistance gene transfer driven by environmental subinhibitory tetracyclines.

Antibiotics at subinhibitory concentrations are key drivers of resistance gene dissemination in the environment; however, the mechanisms operating with recipient bacteria remain poorly understood. Here, we propose a Physical-Physiological Coupling hypothesis to explain how tetracyclines promote plasmid transfer. Using a combination of molecular simulations and biological assays, we show that oxytetracycline exerts the strongest effect, increasing conjugation frequency by 4.1-fold relative to controls. Mechanistically, oxytetracycline acts as a specific modulator that binds tightly to the TolC

Extracellular Vesicles as Emerging Cervical Cancer Diagnostic and Molecular Signatures of Tumor and Immune Dynamics.

Cervical cancer remains a major worldwide health concern and ranks as the fourth most common malignancy affecting women, especially in low- and middle-income countries, where early screening and monitoring facilities are limited. Though several screening strategies, including Pap smears, colposcopy, computed tomography, and HPV testing, are available for the detection of cervical carcinoma, these methods cope with various challenges, such as diagnostic errors, false-positive and false-negative results, invasiveness, and sample collections, which could lead to unnecessary anxiety and procedures

Modeling Cohen Syndrome in Phoenix Cells: VPS13B Loss Causes Organelle Stress, G1/S Delay, and Fibrillary Inclusion Bodies Formation.

Cohen syndrome, caused by pathogenic variants in VPS13B, is characterized by microcephaly, developmental delay, and progressive retinal degeneration, yet the cellular mechanisms linking VPS13B dysfunction to disease pathology remain incompletely understood. Here, we used CRISPR-Cas9 to delete VPS13B exons 2-4 in Phoenix HEK293 cells, generating five independent knockout clones. In all mutant lines, VPS13B disruption caused a marked slowing of cell proliferation due to prolongation of the G1 phase. Immunocytochemistry and transmission electron microscopy revealed that VPS13B mutations causes Go

Unlocking the Unreachable: Cationic ThermoSomes for Next-Generation Posaconazole Therapy.

Invasive fungal infections (IFIs) represent a serious global health threat due to rising incidences and drug resistance. Traditional antifungal therapies face limitations, including toxicity, rapid clearance, and resistance, demanding nanotechnology-driven solutions. While lipid-based formulations, such as AmBisome, have achieved clinical success, scalability is restricted by high costs and complex manufacturing. This study advances the solvent-free approach by developing "ThermoSomes", bilayered lipid vesicles encapsulating the FDA-approved antifungal Posaconazole (PCZ) for IFI treatment. The

Silent messengers in the nano-world: Harnessing extracellular vesicles as theranostic tools for neonatal surgical conditions.

Extracellular vesicles (EVs) are lipid bilayer-enclosed nanoparticles secreted by all cells that mediate intercellular communication by transferring proteins, lipids, and nucleic acids. Among their cargo, microRNAs are key post-transcriptional regulators of organogenesis. In fetal lung development, EV-mediated signaling is essential for the coordination of epithelial, mesenchymal, endothelial, and immune cells. In congenital diaphragmatic hernia (CDH), disruption of these interactions leads to pulmonary hypoplasia and pulmonary hypertension, the primary causes of morbidity and mortality. Amnio

Plasma-Enabled Smart Nanoexosome Platform as Emerging Immunopathogenesis for Clinical Viral Infection.

Smart nanoexosomes are nanosized structures enclosed in lipid bilayers that are structurally similar to the viruses released by a variety of cells, including the cells lining the respiratory system. Of particular importance, the interaction between smart nanoexosomes and viruses can be used to develop antiviral drugs and vaccines. It is possible that nanoexosomes will be utilized and antibodies will be acquired more successfully for the transmission of an immune response if reconvalescent plasma (CP) is used instead of reconvalescent plasma exosomes (CPExo) in this concept. The Role of Lipids in Legionella-Host Interaction.

Legionella are Gram-stain-negative rods associated with water environments: either natural or man-made systems. The inhalation of aerosols containing Legionella bacteria leads to the development of a severe pneumonia termed Legionnaires' disease. To establish an infection, these bacteria adapt to growth in the hostile environment of the host through the unusual structures of macromolecules that build the cell surface. The outer membrane of the cell envelope is a lipid bilayer with an asymmetric composition mostly of phospholipids in the inner leaflet and lipopolysaccharides (LPS) in the outer
