Identification and Immunological Characterization of Cuproptosis Related Genes in Preeclampsia Using Bioinformatics Analysis and Machine Learning.

Preeclampsia (PE) is a pregnancy-specific disorder characterized by an unclearly understood pathogenesis and poses a great threat to maternal and fetal safety. Cuproptosis, a novel form of cellular death, has been implicated in the advancement of various diseases. However, the role of cuproptosis and immune-related genes in PE is unclear. The current study aims to elucidate the gene expression matrix and immune infiltration patterns of cuproptosis-related genes (CRGs) in the context of PE. The GSE98224 dataset was obtained from the Gene Expression Omnibus (GEO) database and utilized as the int

Machine Learning in Nonhuman Primate Models of Infectious Diseases: Current Applications and Future Perspectives.

The amount of biologic data produced by biomedical research has increased significantly in both volume and complexity in recent years. Advances in computational power have increasingly enabled the use of machine learning (ML) to analyze and predict patterns from large-scale, complex biologic datasets. In nonhuman primate (NHP) infectious disease models, such high-dimensional datasets containing a large number of features are often obtained by next-generation sequencing-based multiomics and immunologic analyses. As a result, ML is particularly valuable for effective analysis and predictive mode

Integrative bioinformatics analysis identifies placental senescence-associated signatures in early-onset preeclampsia.

BACKGROUND: Early-onset preeclampsia (EOPE) is a severe hypertensive disorder of pregnancy associated with preterm delivery and maternal multi-organ dysfunction. Although placental senescence and immune dysregulation have been implicated in EOPE, the expression profile of senescence-related genes(SRGs) and their mechanistic roles in disease progression remain unclear. METHODS: Multiple placental bulk RNA-seq datasets were integrated to identify senescence-related differentially expressed genes in EOPE. Weighted gene co-expression network analysis (WGCNA) and multiple machine learning algorith

Identifying Mitochondria-Related Candidate Biomarkers of Ligustri Lucidi Fructus in Diabetic Nephropathy.

Mitochondrial dysfunction and excessive oxidative stress within mitochondria are key pathological factors driving renal tubular injury in diabetic nephropathy (DN). Although Ligustri Lucidi Fructus (LLF) is traditionally used to treat DN, the mechanisms involved, particularly those relating to mitochondria-associated genes and pathways, remain poorly understood. This study used differential expression analysis of the GSE142025 dataset to identify differentially expressed genes (DEGs) related to DN. Feature genes were selected by cross-referencing the outputs of four machine learning models. Ge

Integrative multi-omics and machine learning reveal PLAUR as a Pan-Cancer prognostic biomarker and potential therapeutic target.

Tumor heterogeneity remains a major barrier to effective cancer therapy, driving the need for novel biomarkers and targeted strategies. A bibliometric analysis underscored the lack of a systematic pan-cancer study of PLAUR. By integrating functional experiments, multi-omics analyses, and machine learning, we systematically investigated urokinase-type plasminogen activator receptor (PLAUR) across malignancies. PLAUR overexpression was observed in 10 cancer types, with significant prognostic value in 7 of these. Epigenetic analysis revealed its association with specific methylation patterns. Fun

Early-warning signals and the role of H9N2 in the spillover of avian influenza viruses.

BACKGROUND: The spillover of avian influenza viruses (AIVs) presents a significant global public health threat, leading to unpredictable and recurring pandemics. Current pandemic assessment tools suffer from deficiencies in terms of timeliness, capability for automation, and ability to generate risk estimates for multiple subtypes in the absence of documented human cases. METHODS: To address these challenges, we created an integrated database encompassing global AIV-related data from 1981 to 2022. This database enabled us to estimate the rapid expansion of spatial range and host diversity for

RNA m5C Modifications in the Development and Prognosis of Muscle-Invasive Bladder Cancer.

The m5C RNA modifications have been implicated in the pathogenesis of urothelial carcinoma and hold potential as prognostic biomarkers for muscle-invasive bladder cancer (MIBC) patients. In this study, we developed an MIBC-risk model by integrating m5C modification-related genes and differentially expressed genes using Nanopore sequencing and a machine learning approach. Compared to our previous research, we observed that m5C modifications are more functional, with the most enriched regions being the 3'UTR and exons. Our analysis revealed differential m5C methylation sites in several well-char

Magnetic 3D macroporous MOF oriented urinary exosome metabolomics for early diagnosis of bladder cancer.

Bladder cancer (BCa) exhibits the escalating incidence and mortality due to the untimely and inaccurate early diagnosis. Urinary exosome metabolites, carrying critical tumor cell information and directly related to bladder, emerge as promising non-invasive diagnostic biomarkers of BCa. Herein, the magnetic 3D ordered macroporous zeolitic imidazolate framework-8 (magMZIF-8) is synthesized and used for efficient urinary exosome isolation. Notably, beyond retaining the single crystals and micropores of conventional ZIF-8, MZIF-8 is further enhanced with highly oriented and ordered macropores (150

Machine Learning in Tremor Analysis: Critique and Directions.

Tremor is the most frequent human movement disorder, and its diagnosis is based on clinical assessment. Yet finding the accurate clinical diagnosis is not always straightforward. Fine-tuning of clinical diagnostic criteria over the past few decades, as well as device-based qualitative analysis, has resulted in incremental improvements to diagnostic accuracy. Accelerometric assessments are commonplace, enabling clinicians to capture high-resolution oscillatory properties of tremor, which recently have been the focus of various machine-learning (ML) studies. In this context, the application of M

Unraveling corticobasal syndrome and alien limb syndrome with structural brain imaging.

Alien limb phenomenon is a rare syndrome associated with a feeling of non-belonging and disowning toward one's limb. In contrast, anarchic limb phenomenon leads to involuntary but goal-directed movements. Alien/anarchic limb phenomena are frequent in corticobasal syndrome (CBS), an atypical parkinsonian syndrome characterized by rigidity, akinesia, dystonia, cortical sensory deficit, and apraxia. The structure-function relationship of alien/anarchic limb was investigated in multi-centric structural magnetic resonance imaging (MRI) data. Whole-group and single-subject comparisons were made in 2

Structures of the recurrence plot of heart rate variability signal as a tool for predicting the onset of paroxysmal atrial fibrillation.

This paper aims to propose an effective paroxysmal atrial fibrillation (PAF) predictor which is based on the analysis of the heart rate variability (HRV) signal. Predicting the onset of PAF, based on non-invasive techniques, is clinically important and can be invaluable in order to avoid useless therapeutic interventions and to minimize the risks for the patients. This method consists of four steps: Preprocessing, feature extraction, feature reduction, and classification. In the first step, the QRS complexes are detected from the electrocardiogram (ECG) signal and then the HRV signal is extrac

The evolutionary landscape and serotypic dynamics of avian infectious bronchitis virus from spike protein.

Avian infectious bronchitis virus (IBV), a gammacoronavirus with substantial agricultural impact, offers a tractable model for dissecting coronavirus evolution. Here, we integrated 20 years of epidemiological surveillance with whole-genome sequence analysis of 624 IBV strains, including 136 newly isolated field samples, to investigate the evolutionary and structural dynamics of N-linked glycosylation at the spike protein. We identified three dominant glycosylation haplotypes defined by residues 51 and 77 of spike protein, which correlate with receptor-binding interfaces, clinical phenotypes, a
