Comparative analysis of supervised machine learning algorithms for transdermal drug delivery in brain disorders.

To improve patient compliance and provide stable and prolonged delivery of drugs to the brain, transdermal drug delivery systems (TDDS) are being explored as a viable method of treatment of various brain diseases without the invasion of any organs. Optimization of TDDS remains problematic owing to the complex nonlinear interplay between formulation elements and process factors that affect drug entrapment efficiency, stability, and drug-release properties. Therefore, this study aims to develop a supervised Machine Learning (ML) framework to simultaneously predict multiple critical formulation a

Predicting clinical outcomes in Helicobacter pylori-positive patients using supervised learning through the integration of demographic and genomic features.

BACKGROUND: Helicobacter pylori (H. pylori) infection is widespread globally and is linked to outcomes ranging from chronic gastritis to gastric cancer. However, only a minority of infected individuals progress to malignancy, influenced by a mix of bacterial, host, and environmental factors. Current predictive approaches are limited due to relying mainly on clinical and lifestyle data. Genomic approaches have been sparsely used, and thus their incorporation into machine learning models could ensure early and personalized detection. This study aimed to evaluate the impact of integrating host me

Informed forecasting: Leveraging auxiliary knowledge to boost large language model performance on time series forecasting.

The rapid adoption of large language models (LLMs) has sparked growing interest in extending their capabilities beyond traditional natural language tasks, including applications in time series forecasting. This work explores the enhancement of time series forecasting with LLMs by incorporating time-dependent covariates. We propose a set of representative prompting strategies that span a wide range of formats while incorporating covariates, and then implement the proposed framework to evaluate performance across three real-world time series datasets in healthcare, service operations, and transp

Common Challenges in Predicting Opioid-related Outcomes Using Machine Learning.

Machine learning (ML) models have been commonly utilized to predict various opioid-related outcomes and risks, including post-operative opioid use, opioid use disorder (OUD), misuse, or overdose. Despite their promising performance, the clinical utility and cross-study comparability of ML models are constrained. This is mainly due to variability in outcome definitions, data heterogeneity, class imbalance and analysis concerns, inadequate external or prospective validation, and limited real-world deployment and ethical concerns. Herein, we present an updated overview of these challenges in pred

WaterMAP: A Scalable Machine Learning Framework for Emission-Factor-Derived Spatiotemporal GHG Prediction and Mitigation in Wastewater Treatment Plants.

Greenhouse gas (GHG) emissions from wastewater treatment have gained increasing attention in global climate governance. However, conventional emission-factor (EF)-derived inventories lacked the ability to capture nonlinear variations, while existing machine learning (ML) models remained fragmented in scale. Here, we introduce WaterMAP (Wastewater AI Treatment Emission Regression for Multi-scale Accounting and Prediction), a novel ML framework designed for spatiotemporal EF-derived GHG prediction and mitigation evaluation. Using 40,722 inventory-based GHG records from 5155 WWTPs in China during

A feature-centric predictive and sensitivity analysis framework for bio-oil yield assessment in biomass pyrolysis.

Accurately predicting bio-oil yield from biomass pyrolysis is a real challenge due to nonlinear interactions between feedstock physicochemical properties and operating conditions. In addition, high feature dimensionality, uncertainties in experimental measurements, and multicollinearity make prediction accuracy and interpretability even more difficult, which explains the demand for a feature-driven and explainable modeling framework. We developed an advanced regression feature-based machine learning framework trained on a very comprehensive dataset of both biomass characteristics and pyrolysis

Continuous Evaluation Frameworks for Retrospective Evaluation of Clinical Machine Learning Models.

The performance of clinical machine learning (ML) models that continuously predict the risk of a future event is typically evaluated using metrics at a single time point. While these metrics are widely accepted to evaluate the performance of ML models, they are not comprehensive enough to capture the performance and behavior of the ML models when executing continuously in real-world settings. Additionally, these single time point metrics assess model discriminability, which is different from clinical utility. In this paper, we propose three novel frameworks to continuously evaluate clinical ML

Integrating Target Variable Selection Into Environmental Machine Learning for Surface Water Pollution Management.

Target variable selection is often predefined in machine learning (ML)-based surface water pollution prediction without systematically considering dataset characteristics. This study developed a target variable selection unit (TVSU) integrating Gini feature importance and model pre-evaluation for same-period, cross-station, and cross-variable reconstruction of surface water quality and associated pollution-management decision support. Results showed that the scenario adhering to TVSU achieved the best predictive performance in both shallow and deep learning models (nRMSE values of 0.418 and 0.

Lightweight large language models for early sepsis prediction via a semantic abstraction rule engine.

Sepsis remains a major global health challenge due to its high mortality rate and the difficulty of predicting its onset from nonspecific early symptoms. To address this, we explored the feasibility of using fine-tuned small-scale large language models (LLMs) to perform accurate sepsis prediction from structured clinical time-series data. We developed a Semantic Abstraction Rule Engine (SARE) that transforms numerical clinical data into clinician-readable textual descriptions through temporal feature extraction and semantic transformation. Using these representations, four lightweight LLMs wer
