m6A modification in immune-cell inflammatory fate: from RNA fate regulation to therapeutic translation in inflammatory diseases.

Inflammatory disease is sustained not only by immune activation but also by the failure of activated cells to terminate effector programs and return tissues to homeostasis. This review evaluates N6-methyladenosine (m6A) as a post-transcriptional regulator of that persistence. Rather than cataloguing diseases or classifying individual m6A regulators as pro- or anti-inflammatory, we organize the evidence around a causal chain linking a regulator and target RNA to a site- or reader-dependent change in RNA fate, an immune-cell phenotype, and a disease outcome. Mechanistically developed evidence is

JAK2V617F+ single-stranded DNA modulates exosome complex activity and DNA-sensing pathways in myeloproliferative neoplasms.

Philadelphia chromosome-negative myeloproliferative neoplasms (Ph-MPNs) are characterized by excessive myeloid cell proliferation due to mutations in hematopoietic stem/progenitor cells, most notably JAK2V617F mutation. This mutation leads to persistent JAK2 activation, driving genomic instability and accumulation of single-stranded DNA (ssDNA) fragments in the cytoplasm. The RNA-exosome complex, a critical ribonuclease complex, plays a key role in maintaining genomic stability by degrading damaged RNAs and processing ssDNA. In this study, we examined the effects of JAK2V617F+ ssDNA on the DNA

A POLR3B-variant reveals a Pol III transcriptome response dependent on La protein/SSB.

RNA polymerase III (Pol III, POLR3) synthesizes tRNAs and other small non-coding RNAs. Human POLR3 pathogenic variants cause a range of deve … minators comprised of 4Ts than of ≥5Ts. La-knockdown decreased Pol III ncRNA expression unlinked to RNA stability. Consistent with these effects, small-RNAseq showed that POLR3B1625A>G and patient fibroblasts express more tRNA fragments (tRFs) derived from pre-tRNA 3'-trailers (tRF-1) than from mature-tRFs, and higher levels of multiple miRNAs, relative to cont …

Binding induced isothermal amplification reaction to activate CRISPR/Cas12a for amplified electrochemiluminescence detection of rabies viral RNA via DNA nanotweezer structure switching.

Rabies is caused by the infection of Rabies virus, it leads to fatal encephalitis, developing a highly sensitive and specific detection method for Rabies virus remains a challenge. Herein, we report an electrochemiluminescence (ECL) biosensor for Rabies viral RNA based on dual-signal amplification and DNA nanotweezers (DTs). Dual-signal amplification process includes target binding induced isothermal amplification and CRISPR-based amplification. In the presence of target RNA, two assisted probes simultaneously hybridized with it to trigger isothermal amplification with the help of polymerase a

Design of Gaussia luciferase-based bioluminescent stem-loop probe for sensitive detection of HIV-1 nucleic acids.

Here we describe the design of a bioluminescent stem-loop probe for the sensitive detection of HIV-1 spliced RNA. In this study, we employed Gaussia luciferase (GLuc), a bioluminescent protein that has several advantages over other bioluminescent proteins, including smaller size, higher bioluminescent intensity, and chemical and thermal stability. GLuc was chemically conjugated to the DABCYL-modified stem-loop probe (SLP) and was purified with a 2-step process to remove unconjugated GLuc and SLP. The binding of the target RNA to the loop region of the SLP results in the open conformation separ

CELF family of RNA-binding proteins: roles in disease biology and potential for therapeutic intervention.

The CUG-BP and Elav-like (CELF) family of RNA-binding proteins are key regulators of post-transcriptional gene expression, coordinating alternative splicing, mRNA stability, and translation. Although individual members, particularly CELF1 and CELF2, have been extensively characterized, a systematic, paralog-resolved integration of structural determinants, regulatory mechanisms, and disease relevance across all six CELF proteins remains limited. Here, we establish an integrative framework linking conserved RNA recognition motifs and divergent linker domains to context-dependent regulatory outpu…

Exosomal YB-1 facilitates ovarian restoration by MALAT1/miR-211-5p/FOXO3 axis.

Premature ovarian failure (POF) affects many adult women less than 40 years of age and leads to infertility. Mesenchymal stem cells-derived small extracellular vesicles (MSCs-sEVs) are attractive candidates for ovarian function restoration and folliculogenesis for POF due to their safety and efficacy, however, the key mediator in MSCs-sEVs that modulates this response and underlying mechanisms remains elusive. Herein, we reported that YB-1 protein was markedly downregulated in vitro and in vivo models of POF induced with H2O2 and CTX respectively, accompanied by granulosa cells (GCs) senescenc

RNA therapeutics in ophthalmology - translation to clinical trials.

The use of RNA interference technology has proven to inhibit the expression of many target genes involved in the underlying pathogenesis of several diseases affecting various systems. First established in in vitro and later in animal studies, small interfering RNA (siRNA) and antisense oligonucleotide (ASO) therapeutics are now entering clinical trials with the potential of clinical translation to patients. Gene-silencing therapies have demonstrated promising responses in ocular disorders, predominantly due to the structure of the eye being a closed and compartmentalised organ. However, althou

NR2E3 is a key component in p53 activation by regulating a long noncoding RNA DINO in acute liver injuries.

Damage-induced long noncoding RNA (DINO) is a long noncoding RNA that directly interacts with p53 and thereby enhances p53 stability and activity in response to various cellular stresses. Here, we demonstrate that nuclear receptor subfamily 2 group E member 3 (NR2E3) plays a crucial role in maintaining active DINO epigenetic status for its proper induction and subsequent p53 activation. In acetaminophen (APAP)- or carbon tetrachloride-induced acute liver injuries, NR2E3 knockout (KO) mice exhibited far more severe liver injuries due to impaired DINO induction and p53 activation. ETV4 mediates dosage-dependent prostate tumor initiation and cooperates with p53 loss to generate prostate cancer.

The mechanisms underlying ETS-driven prostate cancer initiation and progression remain poorly understood due to a lack of model systems that recapitulate this phenotype. We generated a genetically engineered mouse with prostate-specific expression of the ETS factor, ETV4, at lower and higher protein dosage through mutation of its degron. Lower-level expression of ETV4 caused mild luminal cell expansion without histologic abnormalities, and higher-level expression of stabilized ETV4 caused prostatic intraepithelial neoplasia (mPIN) with 100% penetrance within 1 week. Tumor progression was limit

Monotropein mitigates atherosclerotic plaque vulnerability through circ_0140736/miR-495/SIRPα-mediated efferocytosis.

Efferocytosis is a biological process in which phagocytic cells engulf and degrade senescent, damaged, or dead cells to prevent secondary necrosis and inflammation. This process is implicated in various diseases, including atherosclerosis (AS). Enhancing efferocytosis efficiency may offer a promising therapeutic strategy for managing AS. Consequently, increasing attention is being paid to pharmacological agents that can promote efferocytosis. Monotropein (MON), the predominant iridoid glycoside in Morinda officinalis, has demonstrated anti-inflammatory and analgesic properties in previous stud

Modular antibody-functionalized poly(β-amino ester) polyplexes: Colloidal and interfacial design for targeted mRNA delivery to human T cells.

The rational design of polymer-nucleic acid nanoparticles requires precise control over colloidal stability, interfacial structure, and assembly under biologically relevant conditions. Here, we report a hierarchically assembled, antibody-functionalized polyplex nanoplatform for targeted messenger RNA (mRNA) delivery. A biodegradable cationic poly(β-amino ester) (PβAE-447) enables efficient condensation of in vitro-transcribed mRNA into nanoscale polyplexes. To enhance stability and introduce modular functionality, polyplexes were coated with a poly(γ-glutamic acid)-avidin (PGA-avidin) corona,

Multiple myeloma associated long non-coding RNA PLUM confers chemoresistance by enhancing PRC2 mediated UPR pathway activation.

Multiple myeloma (MM) is the second most common hematological malignancy that displays diverse genetic heterogeneity leading to treatment resistance. Recurrent mutations causing hyperactivation of the non-canonical NF-ĸB pathway are highly prevalent in relapsed, refractory MM patients, but the precise mechanisms driving chemoresistance are poorly understood. Here, we identify a long non-coding RNA termed PLUM, that is overexpressed in NF-ĸB mutant high-risk MM subtypes and patients who are refractory to VRd treatment regimen. Mechanistically, PLUM interacts with Polycomb Repressive Complex 2 t

TWIST1 drives endothelial-to-mesenchymal-transition to stabilize atherosclerotic plaques.

Rupture of unstable atherosclerotic plaques is a major cause of mortality. Endothelial-to-mesenchymal transition associates with advanced atherosclerotic plaques and contributes to plaque progression. We examined the role of Twist1, a transcription factor that drives endothelial-to-mesenchymal transition, in plaque progression by inducible deletion from endothelial cells in hypercholesterolemic mice (Twist1ECKO Apo-/-). Single-cell RNA sequencing coupled to endothelial cell-tracking reveals that Twist1 promotes endothelial-to-mesenchymal transition in advanced atherosclerotic plaques. Histolog
