Host mouse strain is not selective for a laboratory adapted strain of Schistosoma mansoni.

We genotyped pooled adult worms of Schistosoma mansoni from infected CF1, C57BL/6, BALB/c, and BALB/c interferon gamma knockout mice in order to establish if mouse strain differences selected for parasite genotypes. We also compared differentiation in eggs collected from liver and intestines to determine if there was differential distribution of parasite strains in the vertebrate host that might account for any genotype selection. We found that mouse strains with differing immune responses did not differ in resistance to infection and did not select for parasite genotypes. Schistosoma mansoni

Cloning, heterologous expression in Escherichia coli and characterization of a protein disulfide isomerase from Fasciola hepatica.

A Fasciola hepatica cDNA clone of 1752 bp was isolated from an adult worm cDNA expression library by immunological screening using a rabbit serum against the excretory-secretory antigens. The nucleotide sequence of the cDNA revealed the presence of an open reading frame of 489 codons which encoded a 55 kDa polypeptide, showing a high degree of homology to protein disulfide isomerases. This putative antioxidant protein cDNA was expressed in Escherichia coli as a GST fusion protein. The cleaved recombinant protein was shown to be biologically active in vitro by mediating the oxidative refolding

Molecular evidence for human Metagonimus kogai and M. saitoi infection in Korea: Detection of COI genes in the feces of riverside people along the Seomjin‑gang (river).

Until now, 3 Metagonimus spp. (M. yokogawai, M. takahashii, and M. miyatai) causing human metagonimiasis have been reported in Korea. In this study, we investigated the possible presence of Metagonimus spp. other than these 3 species using human fecal samples from an endemic area in Korea. DNA was extracted from Metagonimus egg-positive fecal samples collected from residents of Gurye-gun, Jeollanam-do. A total of 21 representative mitochondrial cytochrome c oxidase subunit I sequences were obtained by PCR and cloning, and sequencing. Phylogenetic analysis revealed 1 cluster corresponding to M.

Riding South American rivers: a multilocus assessment of Creptotrematina batalhensis (Digenea: Allocreadiidae).

The Allocreadiidae comprise a diverse group of digeneans infecting freshwater fishes worldwide, with South America harbouring a particularly rich fauna. Within this family, Creptotrematina includes five valid species parasitizing primarily characid fishes, most of which have been recorded from the La Plata Basin. Despite the large geographic extent and hydrological complexity of this basin, the genetic structure and evolutionary relationships of Creptotrematina species across its sub-basins remain poorly understood. The inclusion of a new ITS sequence of Martorellia liciae recovered the same t

Molecular prevalence of zoonotic nematode parasite Calodium hepaticum in household rat population in Chennai city, India.

Calodium hepaticum (syn. Capillaria hepatica) is a zoonotic nematode parasite occurring commonly in the liver of rats, which act as primary reservoir hosts. In the present study, 55 household rats in Chennai, Tamil Nadu, India, were trapped and screened for C. hepaticum infection in the liver. All the collected rat liver samples were subjected to macroscopic examination. Rat liver samples with yellowish-white patch lesions were confirmed for the presence of bi-operculate and ellipsoidal shape eggs by parasitological and histopathological examination. A parasite-specific semi-nested PCR was dev

Development of a loop-mediated isothermal amplification with lateral flow dipstick (LAMP-LFD) assay for the detection of Dactylogyrus monogeneans in goldfish (Carassius auratus).

The monogenetic trematode, genus Dactylogyrus, is an important group of ectoparasites that causes disease in fish, which can result in economic losses, particularly in the ornamental fish market. Dactylogyrus monogeneans attach to the host's gills, causing respiratory disorders and potentially leading to fish morbidity and mortality. In this study, we developed a detection method for Dactylogyrus spp. infection in goldfish (Carassius auratus) using a loop-mediated isothermal amplification (LAMP) assay coupled with a lateral flow dipstick (LFD). The LAMP-LFD assay, which targets the 28S rDNA ge

In situ collected small intestinal mucosal swabs as alternative analytes to diagnose Echinococcus multilocularis infection in its definitive hosts by real-time polymerase chain reaction.

The cestode Echinococcus multilocularis parasitizes small intestines of carnivores such as foxes (Vulpes vulpes) and raccoon dogs (Nyctereutes procyonoides) as definitive hosts and represents an important zoonotic threat. This study aimed to determine whether E. multilocularis DNA detection methods by real-time PCR involving easy-to-collect sample types, such as intestinal swabs could represent alternatives to traditional parasitological techniques. Three analytes from foxes and raccoon dogs were tested. i. Swabs taken immediately during necropsy (in situ) from the small intestinal mucosa (Fas

The diversity of Khawia tapeworms (Cestoda: Caryophyllidea) of barbels (Cyprinidae: Barbinae) in the western Palearctic: the existence of vicariant lineages and the description of a new species from the Iberian Peninsula.

The caryophyllidean tapeworm Khawia armeniaca has long been regarded as an exceptionally widespread species within its genus, notable for its significant morphological variability. However, with the accumulation of molecular data from different fish hosts, K. armeniaca was suspected to represent a species complex. To clarify the true identity of these parasites, a comprehensive morphological and molecular study (using 18S, 28S and ITS2 ribosomal regions) of K. armeniaca tapeworms from barbels (Barbinae) across the Iberian Peninsula and the Middle East has been conducted. The results revealed t

Detection and quantification of species-specific gastrointestinal nematode DNA in goat feces using quantitative PCR.

Goats are important livestock species in tropical and subtropical regions. However, they are susceptible to gastrointestinal nematode (GIN) infections, which can significantly reduce their productivity. Therefore, accurate diagnosis and effective control of GIN are essential for the management of goat production. In this study, we investigated the feasibility of detecting two GIN species in fecal samples using a DNA quantification method and quantifying the parasite load for each species. We identified two species, Haemonchus contortus and Trichostrongylus colubriformis, by direct DNA sequenci

Surveillance for Aelurostrongylus abstrusus in feral cats on Prince Edward Island reveals low prevalence.

Aelurostrongylus abstrusus is a metastrongyloid nematode of domestic and wild felids with a cosmopolitan distribution. In Canada, confirmed cases have been reported from several provinces, but no published data exist for Prince Edward Island (PEI). This study aimed to determine the occurrence of A. abstrusus in feral cats from PEI. Between January and December 2024, fecal samples were collected opportunistically from feral cats presented for trap-neuter-return (TNR) surgery at the Atlantic Veterinary College. The Baermann technique was used to detect first-stage larvae, which were identified m

Challenging the phylogenetic relationships among Echinococcus multilocularis isolates from main endemic areas.

Alveolar echinococcosis (AE) is a rare but severe disease that affects more than 18,000 people worldwide per year. The complete sequencing of the mitochondrial genome of Echinococcus multilocularis has made it possible to study the genetic diversity of the parasite and its spatial and temporal evolution. We amplified the whole mitochondrial genome by PCR, using one uniplex and two multiplex reactions to cover the 13,738 bp of the mitogenome, and then sequenced the amplicons with Illumina technology. In total, 113 samples from Europe, Asia, the Arctic and North America were analyzed. Three majo

Molecular detection of two major gastrointestinal parasite genera in cattle using a novel droplet digital PCR approach.

Cooperia sp. and Ostertagia sp. are two cosmopolitan parasitic nematodes often found in mixed gastrointestinal infections in cattle across temperate regions. In light of the recent increase in the emergence of anthelmintic resistance in these and other nematodes derived from cattle around the globe, and their negative impact on animal health and productivity, novel molecular assays need to be put forth in order to facilitate the monitoring of parasite burden in infected herds, using pasture and/or fecal samples. Here, we describe a novel droplet digital PCR platform-based concept for precise i

Morphological and molecular characteristics of adult worms of Gnathostoma Owen, 1836 (Nematoda) collected from domestic pigs in Dien Bien Province, northern Vietnam.

Gnathostomes are of interest because of their unique appearance and medical importance. Among 13 valid species of the genus Gnathostoma Owen, 1836 (Nematoda: Spirurida), two species, G. doloresi Tubangui, 1925 and G. hispidum Fedtschenko, 1872, are parasites of pigs but their larvae can infect humans to cause gnathostomiasis. In this study, we collected adults of Gnathostoma sp. from the stomach of domestic pigs (Sus scrofa domesticus Linnaeus) from Dien Bien Province, northern Vietnam. Morphologically, nematodes found here are most similar to G. doloresi with a slight difference in the spicul

Application of a household-based molecular xenomonitoring strategy to evaluate the lymphatic filariasis elimination program in Tamil Nadu, India.

BACKGROUND: The monitoring and evaluation of lymphatic filariasis (LF) has largely relied on the detection of antigenemia and antibodies in human populations. Molecular xenomonitoring (MX), the detection of parasite DNA/RNA in mosquitoes, may be an effective complementary method, particularly for detecting signals in low-level prevalence areas where Culex is the primary mosquito vector. This paper investigated the application of a household-based sampling method for MX in Tamil Nadu, India. METHODS: MX surveys were conducted in 2010 in two evaluation units (EUs): 1) a hotspot area, defined as

Mitochondrial genome diversity in dagger and needle nematodes (Nematoda: Longidoridae).

Dagger and needle nematodes included in the family Longidoridae (viz. Longidorus, Paralongidorus, and Xiphinema) are highly polyphagous plant-parasitic nematodes in wild and cultivated plants and some of them are plant-virus vectors (nepovirus). The mitochondrial (mt) genomes of the dagger and needle nematodes, Xiphinema rivesi, Xiphinema pachtaicum, Longidorus vineacola and Paralongidorus litoralis were sequenced in this study. The four circular mt genomes have an estimated size of 12.6, 12.5, 13.5 and 12.7 kb, respectively. Up to date, the mt genome of X. pachtaicum is the smallest genome fo

Evaluation by latent class analysis of a magnetic capture based DNA extraction followed by real-time qPCR as a new diagnostic method for detection of Echinococcus multilocularis in definitive hosts.

A new method, based on a magnetic capture based DNA extraction followed by qPCR, was developed for the detection of the zoonotic parasite Echinococcus multilocularis in definitive hosts. Latent class analysis was used to compare this new method with the currently used phenol-chloroform DNA extraction followed by single tube nested PCR. In total, 60 red foxes and coyotes from three different locations were tested with both molecular methods and the sedimentation and counting technique (SCT) or intestinal scraping technique (IST). Though based on a limited number of samples, it could be establis
