Population genomic and evolutionary modelling analyses reveal a single major QTL for ivermectin drug resistance in the pathogenic nematode, Haemonchus contortus.

BACKGROUND: Infections with helminths cause an enormous disease burden in billions of animals and plants worldwide. Large scale use of anthelmintics has driven the evolution of resistance in a number of species that infect livestock and companion animals, and there are growing concerns regarding the reduced efficacy in some human-infective helminths. Understanding the mechanisms by which resistance evolves is the focus of increasing interest; robust genetic analysis of helminths is challenging, and although many candidate genes have been proposed, the genetic basis of resistance remains poorly

Monitoring the Evolution of Resistance in Spodoptera frugiperda (Lepidoptera: Noctuidae) to the Cry1F Protein in Argentina.

Maize (Zea mays L.) is one of the most important and widely cultivated crops in Argentina. Fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is a common maize pest capable of causing significant yield losses and is most destructive in late-planted maize in subtropical regions, going through five to six generations per growing season. The Bacillus thuringiensis (Bt) trait Herculex I Insect Protection technology by Dow AgroSciences and Pioneer Hi-Bred (HX I, event DAS-Ø15Ø7-1), expressing Cry1F protein, was launched in the 2005-2006 season in Argentina and was widely adopted because of th

Insect pathogens as biological control agents: Back to the future.

The development and use of entomopathogens as classical, conservation and augmentative biological control agents have included a number of successes and some setbacks in the past 1years. In this forum paper we present current information on development, use and future directions of insect-specific viruses, bacteria, fungi and nematodes as components of integrated pest management strategies for control of arthropod pests of crops, forests, urban habitats, and insects of medical and veterinary importance. Insect pathogenic viruses are a fruitful source of microbial control agents (MCAs), particu

Flea control in cats: new concepts and the current armoury.

PRACTICAL RELEVANCE: Flea allergic dermatitis is one of the most common skin diseases of cats presented for veterinary attention. It is therefore important for the practitioner to be able to design an appropriate flea management plan for their patients. CLINICAL CHALLENGES: There is no 'one size fits all' flea control programme for cats. Successful flea management requires an understanding of flea biology and knowledge of the mode of action of commercial flea products, of which there is a wide range available. Management of owner expectations can often present a challenge. Cat owners generall

Spinosad resistance in field populations of melon fly, Zeugodacus cucurbitae (Coquillett), in Hawaii.

BACKGROUND: Control of Zeugodacus cucurbitae, a serious agricultural pest worldwide, often includes or is dependent on the use of spinosad-based insecticides. This is especially the case in Hawaii, where GF-120, a protein bait containing spinosad as the active ingredient, has been in use as a key integrated pest management (IPM) tool against this Tephritid for the last two decades. Here, we report on resistance to spinosad [resistance ratios (RRs) and median lethal concentration (LC50 )] in Hawaii's populations of Z. cucurbitae. RESULTS: High resistance was found in populations from three far

Larval movement and survival of western corn rootworm (Coleoptera: Chrysomelidae) and European corn borer (Lepidoptera: Crambidae) on short-stature corn and tall corn.

Short-stature corn is approximately 60 cm shorter than conventional corn at maturity and is expected to soon be commercially available in the United States. One advantage of short-stature corn is lodging resistance, which should help facilitate its adoption by producers. However, knowledge gaps exist concerning how insect pests interact with short-stature corn. Western corn rootworm, Diabrotica virgifera virgifera LeConte, and European corn borer, Ostrinia nubilalis Hübner, are important pests of corn in the United States. To manage these pests, transgenic corn producing insecticidal toxins de

Molecular analysis of knockdown resistance (kdr) mutations in the voltage-gated sodium channel gene of Aedes aegypti populations from Saudi Arabia.

BACKGROUND: The Aedes aegypti mosquito is the primary vector for dengue, chikungunya, yellow fever and Zika viruses worldwide. The first rec … that are fully susceptible, in Saudi Arabia indicates that further consideration should be given to insecticide resistance management strategies that could restore pyrethroid sensitivity to the populations of Ae. aegypti in Saudi Arabia as part of an integrative vector control strategy.

Inheritance of larval resistance to permethrin in Aedes aegypti and association with sex ratio distortion and life history variation.

The genetic mechanisms that confer larval permethrin resistance were investigated in two strains of Aedes aegypti, vectors of yellow fever and dengue hemorrhagic fever. Larval resistance to permethrin in an Ae. aegypti field-collected resistant Couva (R) strain was associated with the sex-determining locus by analysis of backcrosses to the susceptible Rockefeller (S) strain. The median lethal concentrations (LC50s) of these strains were 23.1 (95% confidence interval = 22.0-24.3) and 2.2 (2.0-2.3) parts/billion of permethrin, respectively. The estimated resistance ratio (RR) for the R strain wa

Costs of single-phase door-to-door insecticide-treated net mass distribution campaigns in Nigeria: a case study in Ondo and Anambra states.

BACKGROUND: The distribution of insecticide-treated nets (ITNs) through mass campaigns remains a cornerstone of malaria vector control. As new ITN products are introduced to address rising insecticide resistance, understanding the cost of such campaigns is essential for informed policy decisions. This study aimed to estimate the programmatic and annualized economic costs of campaigns that distributed pyrethroid-PBO ITNs in Ondo and Anambra States, Nigeria, using a single-phase, door-to-door strategy. METHODS: A micro-costing approach was used to estimate campaign economic costs from the provi…

Induction of RNA interference to block Zika virus replication and transmission in the mosquito Aedes aegypti.

The yellow fever mosquito, Aedes aegypti, serves as the primary vector for epidemic transmission of yellow fever, dengue, Zika (ZIKV), and chikungunya viruses to humans. Control of Ae. aegypti is currently limited to insecticide applications and larval habitat management; however, to combat growing challenges with insecticide resistance, novel genetic approaches for vector population reduction or transmission interruption are being aggressively pursued. The objectives of this study were to assess the ability of the Ae. aegypti antiviral exogenous-small interfering RNA (exo-siRNA) response to i…

Efficacy of pirimiphos-methyl (50% EC), as a larvicide for the control of Culex quinquefasciatus.

BACKGROUND & OBJECTIVE: Pirimiphos-methyl, an organophosphorus insecticide is known to overcome resistance mechanisms of mosquitoes to other organophosphates. Information on the duration of efficacy of pirimiphos-methyl (50% emulsifiable concentrate) for the control of immatures of Culex quinquefasciatus, the vector of bancroftian filariasis, is scanty and hence site specific field trials are necessary to determine the field dosage. Pirimiphos-methyl (50% EC) was tested for its efficacy in cesspits and drains against the immatures of Cx. quinquefasciatus at Puducherry, an endemic area for fila

Adaptive Evolution and Transcriptomic Specialization of P450 Detoxification Genes in the Colorado Potato Beetle Across Developmental Stages and Tissues.

The Colorado potato beetle is a notorious agricultural pest with remarkable adaptability and insecticide resistance. This study conducted comprehensive transcriptome analysis across multiple developmental stages and tissues, focusing on the cytochrome P450 monooxygenase gene family. Using RNA-seq data from 65 datasets, we identified significant transcriptional reprogramming during key developmental transitions, particularly from egg to first instar larva and from third instar larva to adult, reflecting critical biological processes, including feeding initiation and metamorphosis. We identified…

CRISPR-mediated germline mutagenesis for genetic sterilization of Anopheles gambiae males.

Rapid spread of insecticide resistance among anopheline mosquitoes threatens malaria elimination efforts, necessitating development of alternative vector control technologies. Sterile insect technique (SIT) has been successfully implemented in multiple insect pests to suppress field populations by the release of large numbers of sterile males, yet it has proven difficult to adapt to Anopheles vectors. Here we outline adaptation of a CRISPR-based genetic sterilization system to selectively ablate male sperm cells in the malaria mosquito Anopheles gambiae. We achieve robust mosaic biallelic muta…
