Oxidant stress has been implicated in the pathogenesis of Parkinson's disease. To test the oxidant stress hypothesis of dopaminergic degeneration, age-related changes in the mesostriatal dopamine neuron system were compared between zitter mutant rats which have abnormal metabolism of oxygen species in the brain and Sprague-Dawley rat as a control using the neurochemistry and immunohistochemistry. Dopamine content in the caudate-putamen, nucleus accumbens and olfactory tubercle of zitter rats decreased significantly with age, and was lower than that found in corresponding age-matched controls.

To investigate the role of small ubiquitin-like modification (SUMO) in Alzheimer's disease (AD) and its pathogenesis, this study employed bioinformatics methods to identify diagnostic biomarkers for AD based on SUMO-related genes (SRGs). It further conducted preliminary explorations into their role in AD pathogenesis and potential therapeutic targets. Datasets related to AD (GSE140831, GSE63060), a dementia dataset (GSE140830), and 189 SRGs were retrieved from public databases. Candidate genes were identified by intersecting differentially expressed genes (DEGs) with SRGs. A protein-protein in

Polypropylene microplastics (PP-MPs) are ubiquitous in our daily lives, but their toxicological effects on mammals remain poorly understood. This study investigated the toxicity effects of PP-MPs on C57BL/6 mice using 16S rRNA gene amplicon sequencing and transcriptome sequencing. Female and male mice were randomly classified into the control and PP-MPs-treated groups, respectively, and the experiment lasted for 5 weeks. We found that PP-MPs exposure did not affect the levels of immunoglobulin G (IgG), interleukin-4 (IL-4), and interferon gamma (IFN-γ), indicating that humoral immunity and inf

Elderly patients exhibit heightened susceptibility to postoperative complications following general anesthesia and surgery, yet the molecular mechanisms driving this age-dependent vulnerability remain poorly defined. We performed RNA sequencing on olfactory bulb (OB), hippocampus (HI), lung, and spleen from young (3-month, m), late middle-aged (17 m), and geriatric (27 m) male C57BL/6 mice 24 h after 2 h of exposure to isoflurane anesthesia and laparotomy (ISO/OP). Short-term ISO/OP elicited pronounced, age-dependent transcriptional remodeling across tissues. Late middle-aged mice exhibited ro

INTRODUCTION: Australia's native fauna is unique through millions of years of isolated evolution resulting in a significant divergence of marsupial brain anatomy, and thus neural pathways, from eutherian mammals. This study provides a morphometric description of a mature female koala (Phascolarctos cinereus) through segmentation of high-resolution magnetic resonance images. As the availability of brain specimens of the endangered koala is scarce, brain segmentation has been performed on a single specimen.
METHODS: The brain of a naturally deceased koala was removed and fixed in 10% neutral bu

BACKGROUND: Chronic respiratory diseases (CRDs), driven by dysregulated Th2/Th17 inflammation, represent a global health challenge with limited therapeutic options. Interleukin (IL)-25 has emerged as a key upstream regulator of both pathways, yet its exact role in CRDs' pathogenesis remains underexplored. Here, our aim is to evaluate the efficacies of SM17, a novel anti-IL-25 receptor (IL-17RB) antibody, in preclinical models of chronic rhinosinusitis with nasal polyps (CRSwNP) and idiopathic pulmonary fibrosis (IPF).
METHODS: Eosinophil, peripheral blood mononuclear cell, primary macrophage

Major depression (MDD) is a complex and multifactorial mental illness. Despite advances in treatment, about 30% of patients still do not see improvement, underscoring the need for new insights into its pathophysiology. Clinical studies suggest that zinc deficiency may be an essential risk factor for the development of depressive disorder and may also contribute to reduced antidepressant efficacy. We developed a novel animal model combining olfactory bulbectomy (OB) with dietary zinc deficiency (ZnD; 3 mg Zn/kg) to further explore this relationship. After three weeks of OB + ZnD treatment, rats

Exosomes, lipid bilayer-enclosed small cellular vesicles, are actively secreted by various cells and play crucial roles in intercellular communication. These nanosized vesicles transport internalized proteins, mRNA, miRNA, and other bioactive molecules. Recent findings have provided compelling evidence that exosomes derived from stem cells hold great promise as a therapeutic modality for central nervous system disorders. These exosomes exhibit multifaceted properties including anti-apoptotic, anti-inflammatory, neurogenic, and vasculogenic effects. Furthermore, exosomes offer several advantage

Hesperidin is a flavonoid glycoside that is frequently found in citrus fruits. Our group have demonstrated that hesperidin has neuroprotective effect in 6-hydroxydopamine (6-OHDA) model of Parkinson's disease (PD), mainly by antioxidant mechanisms. Although the pathophysiology of PD remains uncertain, a large body of evidence has demonstrated that mitochondrial dysfunction and apoptosis play a critical role in dopaminergic nigrostriatal degeneration. However, the ability of hesperidin in modulating these mechanisms has not yet been investigated. In the present study, we examined the potential

BACKGROUND: The characteristic progression of Lewy pathology in Parkinson's disease likely involves intercellular exchange and the accumulation of misfolded α-synuclein amplified by a prion-like self-templating mechanism. Silencing of the α-synuclein gene could provide long-lasting disease-modifying benefits by reducing the requisite substrate for the spreading aggregation.
OBJECTIVES: As a result of the poor penetration of viral vectors across the blood-brain barrier, gene therapy for central nervous system disorders requires direct injections into the affected brain regions, and invasivenes

We have witnessed an accelerated growth of photonics technologies in recent years to enable not only monitoring the activity of specific neurons, while animals are performing certain types of behavior, but also testing whether specific cells, circuits, and regions are sufficient or necessary for initiating, maintaining, or altering this or that behavior. Compared to other sensory systems, however, such as the visual or olfactory system, photonics applications in pain research are only beginning to emerge. One reason pain studies have lagged behind is that many of the techniques originally deve

In the adult mammalian brain, neurogenesis occurs in the olfactory bulb (OB) and in the dentate gyrus (DG) of the hippocampus. Several studies have shown that multiple stages of neurogenesis are regulated by GABAergic transmission with precise spatio-temporal selectivity, and involving mechanisms common to both systems or specific only to one. In the subgranular zone (SGZ) of the DG, GABA neurotransmitter, released by a specific population of interneurons, regulates stem cell quiescence and neuronal cell fate decisions. Similarly, in the subventricular zone (SVZ), OB neuroblast production is m

Since little information is available regarding cellular antigen mapping and the involvement of non-neuronal cells in the pathogenesis of bovine herpesvirus type 5 (BHV-5) infection, it were determined the BHV-5 distribution, the astrocytic reactivity, the involvement of lymphocytes and the presence of matrix metalloproteinase (MMP)-9 in the brain of rabbits experimentally infected with BHV-5. Twelve New Zealand rabbits that were seronegative for BHV-5 were used for virus inoculation, and five rabbits were used as mock-infected controls. The rabbits were kept in separate areas and were inocula

It has recently been reported that iron oxide nanoparticles (Fe(3)O(4)-NPs, 30 nm) have the ability to translocate directly from the olfactory nerve to the brain. The striatum and hippocampus are important structures in the brain and are associated with the development of Parkinson's and Alzheimer's diseases. Therefore, it is critical to evaluate Fe(3)O(4)-NPs and their potential to confer striatum and hippocampus neurotoxicity. This study focuses on the effects of Fe(3)O(4)-NPs on the striatum and hippocampus, including oxidative injury and the accumulation and retention of Fe(3)O(4)-NPs. Thi

There are three subtypes of alpha2 adrenoceptor, i.e., alpha2A, alpha2B, and alpha2C, mediating the specific effect of epinephrine and norepinephrine in various tissues by means of G protein-coupled signal transduction pathways. In an attempt to delineate the regulatory mechanism of the alpha2B receptor subtype (encoded by subtype gene Adra2b) expression in the central nervous system (CNS), we have established transgenic (Tg) mice lines in which the transgene (NN-lacZ) was composed of the promoter region of Adra2b (NcoI fragment, 4.7 kb immediately upstream from receptor coding region) and a r

PURPOSE OF REVIEW: Targeting serotonergic receptors, as well as the serotonin uptake, is a relevant treatment approach for the treatment of depression. The 5-hydroxytryptamine 7 receptor (5-HT7R) is a G-protein coupled receptor belonging to the serotonergic receptor family, which has received a lot of attention due to its association with mood, circadian rhythm, and cognition. This review discusses the potential of 5-HT7R antagonists to treat depression, especially focusing on the pharmacological and bioengineering approaches.
RECENT FINDINGS: The 5-HT7R is rather common in the central and pe

Cell therapy is a promising strategy for tackling neurodegenerative diseases. The most outstanding results with this approach usually involve neuroprotection of damaged neurons at risk of death, but only with limited success. Current therapies are often based on the idea of "one gene, one disease, one drug" for single targets, a concept that limits their actual effectiveness. In contrast, combining different strategies can establish an advanced cell therapy that can slow down neuronal degeneration. In this study, we took advantage of the combination of cell and gene therapy, by transplanting b

OBJECTIVES: The nose and mouth are the primary entry points for upper respiratory severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection; however, the influence of different entry routes on viral spread remains unclear. Oral and nasal infection routes in terms of viral distribution and presence of inflammation were compared.
METHODS: Syrian hamsters were inoculated with SARS-CoV-2 via three routes: nasal inoculation (NI), simulating conventional upper respiratory infection; lingual (supra-lingual) inoculation (LI), simulating exposure during speaking and eating; and sublingual

BACKGROUND: Neuroinflammation and abnormal energy metabolism have been shown to significantly contribute to the progression of Alzheimer's disease (AD). Adenylate kinase 5 (AK5), an enzyme predominantly expressed in the brain regulates ATP metabolism, has an unclear role in energy metabolism and neuroinflammation in AD.
METHODS: The AD datasets were derived from the GEO public database to analyze the expression levels of AK5 in AD and normal samples and to assess the relationship between AK5 expression and the clinical characteristics of AD patients. Functional enrichment analysis was employe

Non-alcoholic fatty liver disease (NAFLD) is a major public health threat with currently limited therapeutic options. Inhalation therapy shows promise for treating metabolic disorders due to rapid absorption and high patient adherence, though relevant medications remain scarce. Eugenol (EUG), the primary component of Syzygium aromaticum volatile oil, emerges as a promising NAFLD inhibitor from lipid-lowering aromatic Chinese medicine screening. EUG elicited significant anti-steatotic effects in both cultured hepatocytes and comprehensive high-fat diet-induced NAFLD animal models. Mechanistical
