IGF2BP2: an m6A reader that affects cellular function and disease progression.

Insulin-like growth factor 2 messenger RNA (mRNA)-binding protein 2 (IGF2BP2) is a widely studied N6-methyladenosine (m6A) modification reader, primarily functioning to recognize and bind to m6A modification sites on the mRNA of downstream target genes, thereby enhancing their stability. Previous studies have suggested that the IGF2BP2-m6A modification plays an essential role in cellular functions and the progression of various diseases. In this review, we focus on summarizing the molecular mechanisms by which IGF2BP2 enhances the mRNA stability of downstream target genes through m6A modificat…

An Influence of Modification with Phosphoryl Guanidine Combined with a 2'-O-Methyl or 2'-Fluoro Group on the Small-Interfering-RNA Effect.

Small interfering RNA (siRNA) is the most important tool for the manipulation of mRNA expression and needs protection from intracellular nucleases when delivered into the cell. In this work, we examined the effects of siRNA modification with the phosphoryl guanidine (PG) group, which, as shown earlier, makes oligodeoxynucleotides resistant to snake venom phosphodiesterase. We obtained a set of siRNAs containing combined modifications PG/2'-O-methyl (2'-OMe) or PG/2'-fluoro (2'-F); biophysical and biochemical properties were characterized for each duplex. We used the UV-melting approach to esti

Reference genes for proximal femoral epiphysiolysis expression studies in broilers cartilage.

The use of reference genes is required for relative quantification in gene expression analysis and the stability of these genes can be variable depending on the experimental design. Therefore, it is indispensable to test the reliability of endogenous genes previously to their use. This study evaluated nine candidate reference genes to select the most stable genes to be used as reference in gene expression studies with the femoral cartilage of normal and epiphysiolysis-affected broilers. The femur articular cartilage of 29 male broilers with 35 days of age was collected, frozen and further subm

Role of senataxin in DNA damage and telomeric stability.

Ataxia with oculomotor apraxia type 2 (AOA2) is an autosomal recessive neurodegenerative disorder characterized by cerebellar ataxia and oculomotor apraxia. The gene mutated in AOA2, SETX, encodes senataxin (SETX), a putative DNA/RNA helicase. The presence of the helicase domain led us to investigate whether SETX might play a role in DNA damage repair and telomere stability. We analyzed the response of AOA2 lymphocytes and lymphoblasts after treatment with camptothecin (CPT), mitomycin C (MMC), H₂O₂ and X-rays by cytogenetic and Q-FISH (quantitative-FISH) assays. The rate of chromosomal aberra

G-quadruplex stabilization via small molecules as a potential anti-cancer strategy.

G-quadruplexes (G4s) are noncanonical nucleic acid secondary structures formed by guanine-rich DNA or RNA sequences, predominantly located at telomeric ends and within the promoter regions of oncogenes. These structures play essential roles in maintaining telomere stability, regulating DNA replication, and modulating gene transcription and translation. Stabilization of G4 structures can induce cellular senescence and apoptosis while suppressing oncogene expression, positioning them as highly promising therapeutic targets in anticancer research. In recent years, a variety of small-molecule G4 l

LncRMRP drives bladder cancer invasion via a lactylation-driven positive feedback circuit.

Muscle-invasive bladder cancer (MIBC) remains highly aggressive. We identified the long non-coding RNA RMRP as significantly upregulated in MIBC. Functional studies demonstrated that RMRP promotes invasive phenotypes and glycolytic reprogramming. Mechanistically, RMRP binds pyruvate dehydrogenase complex X component (PDHX) and weakens the PDHX-dihydrolipoamide dehydrogenase (DLD) interaction, thereby impairing pyruvate dehydrogenase complex (PDC) function, reducing pyruvate dehydrogenase (PDH) activity, and shifting pyruvate metabolism toward lactate production. The resulting lactate promotes

Reactive Polymer-Engineered Dual-Targeting Fluorescent Lipid Nanoparticles for Enhanced saRNA Delivery.

Lipid nanoparticles (LNPs) are widely explored as delivery vehicles for self-amplifying RNA (saRNA); however, incorporation of surface functionalities, such as fluorescent dyes for imaging and ligands for targeted delivery, remains challenging. In this study, we introduce a reactive poly(pentafluorophenyl acrylate) (PPFPA)-based copolymer platform containing a terminal C12 alkyl chain for LNP incorporation and reactive sites for modular conjugation of near-infrared dye IR775, targeting peptides, and polyethylene glycol chains. The resulting copolymers are successfully incorporated into LNPs, e

m6A-Modulated RNF14 Favors Fatty Acid Oxidation to Drive Colorectal Cancer Progression via Regulation of TAF1/PINK1 Axis.

The contribution of fatty acid oxidation (FAO) to colorectal cancer (CRC) progression has been recognized. However, the detailed mechanisms underlying FAO remain obscure. This study explored the influence of ring finger protein 14 (RNF14) on FAO and its related mechanisms in CRC. We found that RNF14 was up-regulated in CRC, which was positively associated with FAO level. High expression of RNF14 promoted FAO to facilitate growth of CRC cells in vitro and in vivo. Mechanistically, RNA binding motif protein 15 (RBM15)/YTH N6-methyladenosine RNA binding protein 1 (YTHDF1)-mediated N6-methyladenos

Nestin/CXCL12 immunohistochemistry and RNA sequencing map the bone marrow microenvironment in aplastic anemia.

Aplastic anemia (AA) is a rare bone marrow failure syndrome characterized by immune-mediated destruction of hematopoietic stem and progenitor cells (HSPCs). The contribution of the bone marrow microenvironment remains incompletely understood. Here, we analyzed 29 bone marrow biopsies from patients with moderate (mAA), severe (sAA) and very severe (vsAA) AA, along with 12 unaffected controls and seven subcortical pseudohypocellular samples. Immunohistochemistry for nestin and CXCL12 was performed to quantify stromal niches. RNA sequencing was carried out to investigate immune and niche-related

Comprehensive evaluation of Ganoderma lucidum extracts: digestion kinetics, gut microbiota modulation, and immunoregulatory mechanisms.

Ganoderma lucidum (Reishi mushroom) exhibits medicinal-edible functionality mediated by bioactive components such as β-glucans and ganoderic acids that regulate immune responses through multiple pathways. However, the digestive transformation and microbiota-mediated interactions of G. lucidum extracts remain insufficiently characterized, and oral delivery results in substantial degradation during digestion, leading to limited bioavailability (<10%). The impact of extraction methods on component stability, digestive behavior, and metabolite profiles also remains unclear. To address these gaps,

TET2: a critical regulatory hub with broad therapeutic implications across human diseases.

Ten-Eleven Translocation 2 (TET2) is a pivotal α-ketoglutarate and Fe2+-dependent dioxygenase belonging to the TET family, governing epigenetic homeostasis through DNA, RNA, and histone modifications. Its central function involves the iterative oxidation of 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC) and further derivatives, initiating active DNA demethylation. Beyond this, TET2 catalyzes RNA m5C oxidation and interacts with histone modifiers, thus modulating diverse cellular processes. Physiologically, TET2 is indispensable for hematopoietic stem cell (HSC) function, genomic stab

Environmental arsenic hijacks DDX5-mediated FANCA splicing to impair R-loops resolution and drive ovarian aging.

Arsenic, a widespread environmental toxicant, is increasingly implicated in female reproductive dysfunction. Long-term exposure to low concentrations of arsenic leads to diminished ovarian reserve. However, the mechanisms by which arsenic exposure accelerates ovarian aging remain unclear. Here, we demonstrate that arsenic exposure induces widespread disruption of pre-mRNA splicing programs in granulosa cells, and these aberrantly spliced genes are predominantly responsible for maintaining genomic stability. Arsenic exposure induces proteasomal degradation of the RNA helicase DDX5 through the U

Epitranscriptomic Role of m6A in Obesity-Associated Disorders and Cancer Metabolic Reprogramming.

The global rise in obesity and its associated metabolic disorders underscores the need for a deeper investigation into their underlying molecular mechanisms. While genetic factors are well-established contributors, recent research has increasingly focused on epigenetic regulators, particularly N6-methyladenosine (m6A)-the most prevalent internal RNA modification in eukaryotes. This post-transcriptional modification plays a crucial role in RNA metabolism by regulating mRNA stability, splicing, nuclear export, and translation efficiency. Notably, emerging evidence implicates m6A in both adipogen…

Staple oligomers induce a stable RNA G-quadruplex structure for protein translation inhibition in therapeutics.

Gene-silencing mechanisms using RNA interference (RNAi) and antisense oligomers have drawn attention to nucleic acid medicines. However, several challenges remain such as low in vivo stability and inadequate target selectivity. Here we report a versatile and highly selective method for suppressing gene expression with a short oligonucleotide. The oligonucleotide, a Staple oligomer, hybridizes specifically to a target mRNA and artificially induces a higher-order structure, an RNA G-quadruplex (rG4), on the mRNA. This results in the rG4 effectively suppressing the target protein's translation. T

Surface Design Options in Polymer- and Lipid-Based siRNA Nanoparticles Using Antibodies.

The mechanism of RNA interference (RNAi) could represent a breakthrough in the therapy of all diseases that arise from a gene defect or require the inhibition of a specific gene expression. In particular, small interfering RNA (siRNA) offers an attractive opportunity to achieve a new milestone in the therapy of human diseases. The limitations of siRNA, such as poor stability, inefficient cell uptake, and undesired immune activation, as well as the inability to specifically reach the target tissue in the body, can be overcome by further developments in the field of nanoparticulate drug delivery
