Open-label, 2-period sequential drug interaction study to evaluate the effect of a 100-mg dose of desvenlafaxine on the pharmacokinetics of tamoxifen when coadministered in healthy postmenopausal female subjects.

BACKGROUND: Potential drugdrug interactions are a concern for patients taking tamoxifen. OBJECTIVE: This study was designed to determine the effect of coadministering desvenlafaxine on tamoxifen pharmacokinetics. MATERIALS AND METHODS: This open-label, 2-period inpatient and outpatient study enrolled healthy, postmenopausal women. Period 1, day 1, subjects were administered tamoxifen 40 mg followed by 23 days of blood sampling for pharmacokinetic analyses. During period 2, subjects received desvenlafaxine 100 mg/d for 28 days; a single dose of tamoxifen 40 mg was administered with desvenlafa

A joint experimental and theoretical investigation of kinetics and mechanistic study in a synthesis reaction between triphenylphosphine and dialkyl acetylenedicarboxylates in the presence of benzhydrazide.

Stable crystalline phosphorus ylides were obtained in excellent yields from the 1:1:1 addition reaction between triphenylphosphine (TPP) and dialkyl acetylenedicarboxylates, in the presence of NH-acids, such as benzhydrazide. To determine the kinetic parameters of the reactions, they were monitored by UV spectrophotometery. The second order fits were automatically drawn and the values of the second order rate constant (k(2)) were calculated using standard equations within the program. At the temperature range studied the dependence of the second order rate constant (Ln k(2)) on reciprocal temp

[Research advances in molecular biology of carotid body tumor].

The carotid body (CB) is a highly specialized small organ located at the bifurcation of the common carotid artery in the neck and plays an important role in acute adaptation to hypoxia, which is the most common tumor site in head and neck paragangliomas. Although carotid body tumors (CBTs) are mostly slow-growing and benign, they can cause significant morbidity because of their proximity to major arteries and nerves in the head and neck. CBTs can be caused by a genetic predisposition and by chronic hypoxic stimulation. Currently, inactivating germline mutations in the mitochondrial complex II

[The study of the enzyme histochemistry changes of thyroarytenoid when one side of RLN was excised].

OBJECTIVE: To chronologically evaluate the changes in both sides of thyroarytenoid (TA) muscles. when one side of recurrent laryngeal nerve (RLN) was cut off. METHOD: In 7 adult mongrel dogs, 2 dogs for control and the other 5 dogs for the following experiment: a 2 cm segment of right RLN was excised under intraperitoneal sedation. The animals were killed at 2, 4, 6, 8, 10 week respectively and the thyroarytenoid were processed for histochemical reaction and electromicroscope. The mean muscle fiber, standard deviation and muscle fiber type composition were determined. RESULT: For the injured

Respiratory chain components of Leishmania tropica promastigotes.

Crude preparations of kinetoplast vesicles were used to investigate the respiratory chain components in Leishmania tropica promastigotes. In difference spectra from enzymically and chemically reduced preparations, cytochrome b was the predominant component. By utilizing special assays designed to minimize the influence of cytochrome b on difference spectra, cytochromes a, a3 and c555 were demonstrated. Difference spectra from chemically reduced preparations indicated that pyridine nucleotides (NADH) and flavoproteins were also part of the respiratory chain. The presence of these components as

Lysine succinylation: a review of its impacts on the reproductive function and related disease.

Lysine succinylation is a recently identified post-translational modification of proteins that has been demonstrated to play critical roles in various biological activities. In recent years, research on succinylation has progressed considerably in the context of reproductive biology and associated diseases, underscoring the importance of a comprehensive review on this topic. A key mechanism underlying succinylation involves changes in the activity of succinyltransferases and desuccinylases, which in turn regulate alterations in the succinylation level of target proteins, and thereby influence

Itaconate and its derivatives ameliorate autoimmunity by suppressing Th17 cells via regulating mitophagy.

Itaconate (ITA) originates by decarboxylation of cis-aconitate catalyzed by the enzyme aconitate decarboxylase 1. It has immunomodulatory properties, but its role in T cells, especially CD4+ T cells, is mostly unexplored. Here, we examined the effects of ITA and its derivatives 4-octyl itaconate (4-OI) and dimethyl itaconate (DMI) on CD4+ T cells. We demonstrate that itaconate, 4-OI, and DMI specifically inhibit Th17 cells differentiation under cell-polarizing conditions, reduce IL-17 A secretion and mRNA expression of Th17 cell-related genes including Il17a, Il22 and Il17f. Metabolic profilin

The alkylation of AIM2 by itaconate mediates macrophage PANoptosis during sepsis.

Although the immunometabolite itaconate has long been considered an anti-inflammatory, we found that its profound accumulation paradoxically drives macrophage cell death and pro-inflammatory responses. However, the exact molecular mechanisms underlying itaconate-induced macrophage toxicity remain unclear. Here, we demonstrate that pathophysiologically relevant high concentrations of itaconate covalently alkylate the absent in melanoma 2 (AIM2) protein at the cysteine 113 (C113) residue. Itaconate-mediated C113 alkylation structurally stabilizes the AIM2 protein and triggers a conformational ch

Targeting neuroinflammation: itaconate and mesaconate as therapeutic strategies against H7N7 influenza-associated CNS damage.

INTRODUCTION: Influenza A virus (IAV) infection is primarily associated with respiratory disease; however, accumulating evidence indicates that neurotropic strains can induce central nervous system (CNS) inflammation and contribute to persistent neurological dysfunction. Aberrant immune activation is thought to play a critical role in these outcomes, yet therapeutic approaches that effectively attenuate neuroinflammation while preserving antiviral immunity remain limited. Immunometabolic regulators, including the endogenous metabolite itaconate, have recently emerged as key modulators of innat

4-Octyl itaconate ameliorates immune thrombocytopenia by modulating megakaryocyte autophagy and apoptosis.

OBJECTIVE: This study aimed to investigate the effects of 4-Octyl itaconate (4-OI) on immune thrombocytopenia (ITP) mice model and elucidate the underlying mechanism. METHODS: An ITP mouse model was established by intraperitoneal injection of the monoclonal antibody MWReg30 and treated by 4-OI with/without chloroquine (CQ). The mice were divided into four groups: Control, ITP, ITP+4-OI and ITP+4-OI + CQ. Platelet (PLT) content was detected and bone marrow megakaryocytes were quantified using Giemsa staining. Apoptosis was evaluated by TUNEL staining, protein expression of Bax and Bcl-2 in bon

Understanding the structural and functional implications of lysine succinylation in Mycobacterium tuberculosis heat shock protein 16.3.

Heat shock protein 16.3 (Hsp16.3), a major immunodominant antigen of Mycobacterium tuberculosis, exhibits molecular chaperone function that is essential for pathogen's survival and slow growth inside hosts, as well as for enhancing the efficacy of Bacillus Calmette-Guérin (BCG) vaccine. Proteomic studies revealed that Hsp16.3 undergoes lysine succinylation in vivo at all lysine residues (K47, K64, K78, K85, K114, K119 and K132) except K136. However, the effects of succinylation on its structure and function remain unexplored. This study investigated the impact of succinylation, induced by phys
