Brain-derived neurotrophic factor delivered to the brain using poly (lactide-co-glycolide) nanoparticles improves neurological and cognitive outcome in mice with traumatic brain injury.

Currently, traumatic brain injury (TBI) is the leading cause of death or disabilities in young individuals worldwide. The multi-complexity of its pathogenesis as well as impermeability of the blood-brain barrier (BBB) makes the drug choice and delivery very challenging. The brain-derived neurotrophic factor (BDNF) regulates neuronal plasticity, neuronal cell growth, proliferation, cell survival and long-term memory. However, its short half-life and low BBB permeability are the main hurdles to be an effective therapeutic for TBI. Poly (lactic-co-glycolic acid) (PLGA) nanoparticles coated by sur…

Commentary on Kamper et. al., juvenile traumatic brain injury evolves into a chronic brain disorder: The challenges in longitudinal studies of juvenile traumatic brain injury.

Juvenile traumatic brain injury (TBI) leaves survivors facing a potential lifetime of cognitive, somatic and emotional symptoms. A recent study published in Experimental Neurology (Kamper et al., 2013) explored the chronic consequences of focal brain injury induced in the juvenile animal, extending their previous observations out to 6months post-injury. The results demonstrate transient, persistent, and late onset behavioral dysfunction, which are associated with subtle evidence for enduring histopathology. In line with investigations about chronic traumatic encephalopathy from brain injury in…

Molecular species of phosphatidylcholine containing very long chain fatty acids in human brain: enrichment in X-linked adrenoleukodystrophy brain and diseases of peroxisome biogenesis brain.

Molecular species of phosphatidylcholine containing unsaturated (i.e., monoenoic and polyenoic) 32- to 40-carbon (very long chain) fatty acids (VLCFA-PC) are present in normal human brain, the fatty acid composition changing significantly with development. There is a marked increase in the concentration and a change in the polyenoic VLCFA composition of these molecular species in brains of patients with inherited defects in peroxisomal biogenesis [Zellweger's syndrome, neonatal adrenoleukodystrophy (ALD), and infantile Refsum's disease]. In contrast, there is a marked increase in monoenoic VLC…

[Analysis of computed X-ray tomography of the brain in incontinence patients with senile dementia. Relationship between circulatory defect of the brain, brain atrophy and urinary incontinence].

To evaluate the condition of incontinence in patients with senile dementia, we performed computed tomography X-rays to the brain and analyzed the relationship among the circulatory defect of the brain, the brain atrophy and the degree of incontinence. There were 92 patients subjected to this study who were hospitalised due to senile dementia; 74 patients had vascular dementia, 10 patients had senile dementia of Alzheimer type, and 8 patients had the mixed type. (age ranged: 54-95 years; mean: 80.3 years). The degree of incontinence in these patients varied as follows: 18 patients with continen…

Evidence for Brain-To-Gut and Gut-To-Brain Pathways in Primary Care Patients With Disorders of Gut-Brain Interaction, Inflammatory Bowel Disease and Gastroesophageal Reflux Disease.

BACKGROUND: Apart from disorders of gut-brain interaction (DGBI), little data exist on the magnitude of the brain-to-gut pathway in other chronic gastrointestinal conditions such as gastroesophageal reflux disease (GERD) or inflammatory bowel disease (IBD) and what factors modify order of diagnosis. We aimed to determine the proportion of patients who received a diagnosis of a DGBI, GERD, or IBD prior to a new psychological diagnosis (gut-to-brain), and vice versa (brain-to-gut), and whether specific factors moderate the order of diagnosis. METHOD: Data was collected from a retrospective stud…

The "Yoking" of glutamatergic brain mechanisms involved in controlling brain neuronal excitability and psychosis to brain mechanisms involved in appetite regulation: a new hypothesis on the origin of psychosis.

The authors speculate that the human primate evolved psychosis generating brain mechanisms in the service of certain feeding behaviors (i.e., appetite, foraging) during the course of evolution. Furthermore, these "psychosis generating brain mechanisms" may have grown directly out of brain mechanisms servicing appetite, of which neuropeptide Y (NPY) played an important role. A case is made for an NPY contribution to the pathophysiology of psychosis. We hypothesize that the psychomimetic effects of NPY extend to supporting certain "psychomotor" functions that might have been useful for obtaining…

Continuous osmotic minipump infusion of alcohol into brain decreases brain [Mg2+] and brain bioenergetics and enhances susceptibility to hemorrhagic stroke: an in vivo 31P-NMR study.

31P-NMR spectroscopic studies were performed in vivo on brains of rats chronically infused for 7 and 14 days with 30% ethanol (in the third cerebral ventricle). Peripheral blood alcohol concentration (BAC) rose to between 16.5-30.5 mg/dl. Brain intracellular free Mg2+ ([Mg2+]i) fell 33-39%, brain mitochondrial cytosolic phosphorylation potential (CPP) fell 31-48%, and brain phosphocreatine (PCr) fell approximately 15%; however, neither brain intracellular free hydrogen ion concentration (pHi) nor brain intracellular inorganic phosphate (Pi) were affected significantly by the chronic release of…

The impact of creatine on brain functions.

As an endogenous compound essential for energy homeostasis, creatine holds a fundamental role in the bioenergetics of the brain, an organ with high metabolic requirements. This review provides a synthesis of current knowledge on the impact of creatine on the functions of the central nervous system (CNS). The brain, which is capable of de novo creatine synthesis, demonstrates an increase in its internal concentrations following supplementation; however, this effect is less significant than in muscle tissue and is constrained by the blood-brain barrier, among other factors. The development of ef…

The endocannabinoid system and the ageing brain.

Ageing is accompanied by a progressive decline in homeostatic resilience that increases vulnerability to chronic disease, disability and cognitive impairment. Although unhealthy brain ageing can often be prevented by managing the underlying risk factors, the molecular, cellular and physiological mechanisms that connect risk factors to late-life brain dysfunction remain incompletely understood. This review examines the role of the endocannabinoid system in brain ageing. The endocannabinoid system is a short-range lipid signalling network that helps to maintain neuronal, glial, metabolic and inf…

Advances in monitoring the injured brain.

Multimodal neuromonitoring has emerged as a cornerstone of modern neurocritical care for traumatic brain injury (TBI). Traditional monitoring approaches based on intracranial pressure (ICP) and clinical examination alone have proven insufficient to capture the complex, heterogeneous pathophysiology that drives secondary brain injury. Over the past decade, significant advances in monitoring technology, including brain tissue oxygenation, cerebral microdialysis, continuous electroencephalography, near‑infrared spectroscopy, cerebrovascular autoregulation assessment and non‑invasive techniques ha…

Interoception Network in the Rat Brain.

The brain is never truly at rest. Even in the absence of external stimuli or cognitive tasks, the central nervous system continuously receives peripheral signals from visceral organs, such as the heart, lungs, and stomach, and sends motor commands to regulate organ physiology. This bidirectional brain-body interaction, also known as interoception, engages neural pathways via a hierarchical set of brain regions, including the nucleus of the solitary tract, hypothalamus, paraventricular nuclei of the thalamus, insular cortex, and anterior cingulate cortex, among others. However, it is largely un…

Infection control in the brain and the eye.

The Central Nervous System (CNS), comprising the brain and the eye, is considered to have a 'privileged' mechanism for dealing with immunological challenge (immune privilege, IP). CNS IP has been revealed through experiments using foreign protein antigens and cell and tissue alloantigens (grafts), but evidence for a role for IP in modulating host-pathogen interactions in the CNS is limited. However, the low frequency of CNS infection in the face of widespread systemic exposure to CNS-tropic infectious agents, together with the high incidence of CNS infection in immunocompromised individuals, s…

Mapping the Brain's Glymphatic System.

The glymphatic system is a fluid-transport framework in which cerebrospinal fluid (CSF) enters the brain along perivascular routes, exchanges with interstitial fluid (ISF), and exits toward venous, perineural, and meningeal lymphatic pathways enabling waste clearance. Recent studies have clarified the anatomical components that regulate solute movement. The perivascular astrocyte endfeet, which are enriched in polarized aquaporin-4 (AQP4) expression, create a high-permeability water interface that facilitates CSF-ISF exchange. Multiscale physical drivers such as cardiac pulsation, arteriolar v…

The Noradrenergic Brain in Parkinson's Disease.

PURPOSE OF REVIEW: Cerebral noradrenergic activity modulates physiological functions of behaviour, cognition, movement, arousal and sleep. This review aims to provide an accurate summary of the current knowledge on the involvement of the noradrenergic system in Parkinson's Disease (PD) and its clinical correlations based on neuroimaging studies. RECENT FINDINGS: Studies in PD highlight neuromelanin MRI signal loss in the locus coeruleus (LC), and positron emission tomography shows noradrenergic denervation across subcortical and cortical areas. More severe phenotypes of PD, manifesting with c

Paediatric anaesthesia and the developing brain.

Concerns persist regarding the potential long-term effects of general anaesthesia on brain development in children. This narrative review summarises recent preclinical and clinical evidence (2019-25) and updates consensus messages. Preclinical studies show that commonly used anaesthetic agents can interfere with neurodevelopmental processes during vulnerable developmental periods. Clinical evidence is less clear. Randomised trials indicate that a single, short exposure in infancy is not associated with measurable impairment in cognitive outcomes, whereas observational studies report mixed find…

The lipidomic architecture of the mouse brain.

Lipids are fundamental components of the brain, crucial for synaptic transmission and signal propagation. Altered brain lipid composition is associated with common and rare neuropathologies, yet the spatial organization of the mammalian brain lipidome remains insufficiently characterized compared with other modalities1-6. Here we mapped the membrane-lipid architecture of the adult mouse brain at micrometric scale, across sexes and during pregnancy. This lipid brain atlas reveals that lipids describe a fine-grained biochemical structure that aligns with functional anatomy. Membrane-lipid spatia…
