[Dicentric Y chromosome].

Dicentric Y chromosomes are the most common Y structural abnormalities and their influence on gonadal and somatic development is extremely variable. Here, we report the third comprehensive review of the literature concerning dicentric Y chromosomes reported since 1994. We find 78 new cases for which molecular studies (PCR or FISH) have been widely applied to investigate SRY (68% of cases), GBY, ZFY, RFS4Y, GCY and different genes at AZF region. For dic(Yq), all cases (n = 20) were mosaic for 45,X and 4 of them were also mosaic for a 46,XY cell line. When breakpoints were available (15/20 cases

Abnormalities of the Eyelashes in Turner's Syndrome.

Turner's syndrome (TS) is a sex chromosome disorder caused by a partial loss, complete absence or structural abnormality of one X chromosome in females. Special ocular features are often found. Some of the abnormalities are only cosmetic, such as the abnormalities of the eyelashes. The present prospective study with 12 TS and 12 control patients demonstrates the higher number of eyelashes as well as the greater vertical distance between the roots of the eyelashes in patients with TS compared with the control group. Increased awareness of this ophthalmological abnormality could be an additional

[Genetic analysis and prenatal diagnosis of a fetus with Xq25 microduplication].

OBJECTIVE: To explore the genetic basis for a fetus with structural brain abnormalities. METHODS: The karyotypes of the fetus and its parents were analyzed by conventional G-banding. Chromosome microarray analysis (CMA) was carried out to detect chromosomal microdeletion and microduplication. RESULTS: No kartotypic abnormality was detected in the fetus and its parents. CMA has identified a 194 kb microduplication at Xq25 in the fetus, which encompassed exons 4-35 of the STAG2 gene and was derived from its mother. CONCLUSION: The Xq25 duplication encompassing part of the STAG2 gene probably

Clinical implications of first-trimester screening.

First-trimester aneuploidy screening is associated with very high detection rates for Down syndrome and other chromosomal abnormalities. The clinical implications of early screening relate to prenatal diagnosis of chromosomal abnormalities and important information obtained at screening. Early screening can change the rate of invasive prenatal diagnosis and the gestational age of prenatal diagnosis and abortion, and lead to earlier prenatal diagnosis of certain major structural abnormalities. The detection of biochemical abnormalities associated with obstetric complications, such as low pregna

Analysis of TSC1 truncations defines regions involved in TSC1 stability, aggregation and interaction.

Tuberous sclerosis complex (TSC) is an autosomal dominant disorder characterised by the development of hamartomas in a variety of organs and tissues. The disease is caused by mutations in either the TSC1 gene on chromosome 9q34, or the TSC2 gene on chromosome 16p13.3. The TSC1 and TSC2 gene products, TSC1 and TSC2, interact to form a protein complex that inhibits signal transduction to the downstream effectors of the target of rapamycin complex 1 (TORC1). Here we investigate TSC1 structure and function by analysing a series of truncated TSC1 proteins. We identify specific regions of the protei

Cytogenetic monitoring of men occupationally exposed to airborne pollutants.

A study of structural chromosomal aberration frequencies in peripheral blood lymphocytes was performed in a group of 20 professional drivers exposed to airborne pollutants and 20 matching controls. The subjects in the latter group were of the same sex (males) and of similar age as the exposed ones, and also had similar habits of smoking and alcohol. A statistically significant increase of chromosomal aberration was observed in the exposed subjects over the control group. An increasing trend of aberrations was observed with the duration of service (exposure) in the exposed individuals. This stu

Breakpoints of 19q13 translocations of benign thyroid tumors map within a 400 kilobase region.

Structural rearrangements involving the long arm of chromosome 19 characterize a cytogenetic subgroup of benign thyroid tumors. To localize the breakpoint of the 19q13 aberrations, we have established three cell lines derived from benign thyroid tumors showing translocations in this region. We have used these cell lines and four additional primary tumors with 19q13 abnormalities for fluorescence in situ hybridization (FISH) mapping studies with ten cosmid clones located between the molecular markers POLD1 and TNNT1. The breakpoints of all chromosome 19 abnormalities mapped within a 400 kb regi

Chromosomal abnormalities in patients with familial polyposis and colorectal cancer.

A search for chromosomal (cytogenetic, karyotypic) changes with specificity for familial polyposis coli (FPC) and related conditions has not been successful. The cytogenetic anomalies described to date (chromosomal instability, tetraploidy, structural anomalies) are not only nonspecific and inconsistent but may also be seen in other congenital conditions predisposing to cancer of various organs. However, this does not mean that some (eg, tetraploidy) of the changes cannot be used as indices in the study and evaluation of affected individuals and their family members. Future application of DNA

A novel gene of beta chain of the IFN-gamma receptor of Huiyang chicken: cloning, distribution, and CD assay.

The beta chain of the interferon-gamma receptor (IFNGR-2) plays a critical role in signal transmission to the nucleus by IFN-gamma. Here, we cloned the full-length cDNA of IFNGR-2 of Huiyang chicken using RACE. mRNA transcripts of IFNGR-2 were detected in peripheral blood leukocytes (PBL) and various organs using Northern blot analysis. The extracellular region of IFNGR-2 (IFNGR-2EC) was expressed in Pichia pastoris, and its secondary structure was investigated by circular dichroism (CD). The Huiyang chicken IFNGR-2 gene is 2221 bp with a polyA+ tail, and it encodes 334 amino acids sharing 30%

Ambiguous Genitalia: An Unexpected Diagnosis in a Newborn.

Alterations in gonad formation or function can lead to congenital conditions in which chromosomal, gonadal, or anatomical sex is atypical. These conditions are referred to as disorders of sex development (DSD) and have a heterogeneous etiology. The assessment of these children by a multidisciplinary team is crucial for an accurate diagnosis and should be initiated promptly due to the potentially life-threatening nature of congenital adrenal hyperplasia, a common cause of DSD. We present a neonate born at 39 weeks with a weak cry, slight hypotonia, poor suction reflex, peculiar facies, and ambi

The Role of Repeat DNA Sequences in Human Evolution and Disease.

Segmental chromosome duplications are an important evolutionary mechanism to produce new gene functions. Once an initial duplication takes place, the probability of a second event (structural change) increases. Segmental duplications (SDs) occur in many sizes and configurations. It has long been thought that SDs contribute to rapid evolution in primate genes. SDs tend to cluster around core duplicons, evolutionarily conserved sequences that are often shared between primate species. Many known SDs are associated with predisposition to human chromosome abnormalities and genetic diseases. Chromos

Pathogenic variants in SLF2 and SMC5 cause segmented chromosomes and mosaic variegated hyperploidy.

Embryonic development is dictated by tight regulation of DNA replication, cell division and differentiation. Mutations in DNA repair and replication genes disrupt this equilibrium, giving rise to neurodevelopmental disease characterized by microcephaly, short stature and chromosomal breakage. Here, we identify biallelic variants in two components of the RAD18-SLF1/2-SMC5/6 genome stability pathway, SLF2 and SMC5, in 11 patients with microcephaly, short stature, cardiac abnormalities and anemia. Patient-derived cells exhibit a unique chromosomal instability phenotype consisting of segmented and

Prenatal detection and outcome of congenital diaphragmatic hernia (CDH) associated with deletion of chromosome 15q26: two patients and review of the literature.

Congenital diaphragmatic hernia (CDH) is a severe birth defect characterized by a defect in the diaphragm with pulmonary hypoplasia and postnatal pulmonary hypertension. Approximately 50% of CDH cases are associated with other non-pulmonary congenital anomalies (so called non-isolated CDH) and in 5-10% of cases there is a chromosomal etiology. The majority of CDH cases are detected prenatally. In some cases prenatal chromosome analysis reveals a causative chromosomal anomaly, most often aneuploidy. Deletion of 15q26 is the most frequently described structural chromosomal aberration in patients

Chromosomal translocations are common in natural killer-cell lymphoma/leukemia as shown by spectral karyotyping.

Natural killer (NK)-cell lymphoma/leukemia is a group of rare but highly aggressive neoplasms. The associated genetic aberrations, as defined by conventional cytogenetics, include 6q deletion and chromosome X copy gain, while translocations have been suggested to be uncommon. In this study, three cases of NK cell lymphoma/ leukemia were investigated by spectral karyotyping (SKY). SKY permitted reinterpretation of the chromosomal alterations defined by G-banding and identified several cryptic translocations. In agreement with G-band, 6q deletion was detected in all 3 cases. Structural rearrange

Molecular basis of male (in)fertility.

It is obvious that the molecular basis of male (in)fertility is not a linear order of genetic events but the interaction of complex genetic networks functional in three main developmental pathways: male germ-line development, male gonad development, and male somatic development. Consequently, primary genetic switch signals should exist for linking the different gene networks and/or for starting them. There is some evidence that such switch signals are concentrated on the sex chromosomes. However multiple genes encoding gene products functional for male fertility exist also on other chromosomes

Cytogenetic abnormalities in renal oncocytic neoplasms.

We have performed cytogenetic studies on five renal oncocytic neoplasms (three grade 2 tumors and two grade 1 tumors) identified histologically by light microscopy. One grade 1 tumor failed to produce mitotic cells. The other four tumors exhibited both normal and abnormal cell lines. Numerical abnormalities were found in both the single grade 1 and two of the grade 2 tumors whereas structural abnormalities were limited to grade 2 tumors. Aneuploidy of chromosome 12 was observed in both grade 1 and 2 tumors. Grade 2 tumors showed more extensive numerical change than the grade 1 tumors. Abnormal

Specific chromosomal mutagenesis observed in stimulated lymphocytes from patients with S-ANLL.

An analysis of R-banded PHA-stimulated lymphocytes from 13 patients with secondary acute non-lymphocytic leukemia (S-ANLL) following breast cancer or lymphoma, and treatment by alkylating agents and/or radiotherapy, is reported. We found that chromosomes 5, 7, 11 and 17 are over-involved in structural rearrangements. These anomalies are similar to those observed in the same categories of patients without S-ANLL, and after in vitro treatment of normal lymphocytes by the alkylating agent melphalan. These anomalies are thus likely to be induced by treatment, independently of S-ANLL. However, the

Cytogenetic studies in malignant lymphomas: a study of 8 cases.

Chromosome studies were carried out, by a direct method, in 28 subjects with malignant lymphomas. Lymph node cells were analysed in 24, ascitic fluid sediments in 3 and bone marrow cells in 1. Chromosome abnormalities, both numerical and structural, were found in 12 of 14 cases of well differentiated and poorly differentiated lymphocytic lymphomas, and reticulum cell sarcomas, and in 8 of 14 cases with Hodgkin's disease. The karyotypes were different from case to case and there was no correlation with the histology. In individual cases the abnormalities followed a clonal pattern indicating a c

Canavanine-based assay for gross chromosomal rearrangements reveals genome instability hotspots and modulating genes in fission yeast.

Gross chromosomal rearrangements are a hallmark of many diseases and cancers. The study of their biogenesis and the mechanisms underlying their formation is greatly facilitated by the availability of genetic reporter assays in model organisms. We present here a novel GCR assay developed in fission yeast, a highly relevant model for understanding genome instability related to human biology. The reporter employs canavanine counter-selection to detect GCRs within a chromosomal context. Using this assay, we identified natural hotspots for GCRs, including inverted long terminal repeats (IR-LTRs). S
