Periarterial macrophage sheaths (ellipsoids) in cat spleen--an electron microscope study.

Periarterial macrophage sheaths (PAMS), a term we introduce to replace "ellipsoids," surround arterial capillaries in the red pulp of the spleen and are major sites for clearance of blood-borne particles. PAMS and their arterial capillaries in cat spleens in various states of congestion and contraction were studied by transmission electron microscopy. Thorotrast, a colloidal suspension of thorium dioxide, was injected to label macrophages. A PAMS consisted of a fine meshwork of reticular cells and reticular fibers which held macrophages and formed a cylindrical sheath around an arterial capill

[Characterization of the convoluted seminiferous tubule wall. II. Ultrastructural alteration after testis retention in the abdominal cavity].

An ultrastructural analysis has been carried out on the seminiferous tubules after different periods of stay of the rat testis in the abdominal cavity. With respect to control rats, no changes are detectable after 5 days, while after 15 and 30 days considerable modifications take place. A number of cellular features are modified, such as the nuclear shape and the perinuclear vesicles, while the cytoplasmic filaments show a less ordered appearance. In addition, the extracellular amorphous matrix undergoes a large increase, due either to an enhanced production or to a reduced turnover related wi

[Soft tissue neoplasms arising from myofibroblasts].

A group of 14 mainly spindle-cell tumours caused some uncertainly during bioptic classification with diagnosis oscillating between proliferating fibroma, fasciitis, fibrosarcoma, mesenchymal tumour, leimyoma, fibromesothelioma or even polymorphocellular sarcoma. A common feature was seen in the presence of different proportions of myofibroblasts of largely variable appearance. The only justification for the term "tumours arising from myofibroblasts" is in the predominance of well defined myofibroblasts. In all other cases the presence of such elements is at least a prospective contribution to

Effect of thiol-oxidation of glutathione with diamide on corneal endothelial function, junctional complexes, and microfilaments.

Intracellular-reduced glutathione (GSH) was removed by thiol-oxidation with diamide during in vitro perfusion of the corneal endothelium. By 15 min the normal mosaic-like pattern of the endothelial cells was disrupted by serpentine-like lines of cell separation at the cell juntions. After 45 min of perfusion, infividual clusters of cells formed cup-shaped islands. The resultant exposure of Descemet's membrane to the perfusion solution resulted in corneal swelling. Transmission electron microscopy revealed that the endothelial cells separated at the apical junctions and that the microfilaments

Upregulated CircRNAs With Efficacy in Preclinical In Vivo Models Reveal New Targets for Therapeutic Intervention in Glioma.

There is a significant need to improve treatment efficacy for patients with glioma. Therefore, we have analyzed the literature for the role of circular RNAs (circRNAs) with respect to selected pathology-related topics of glioma. We have focused on upregulated circRNAs with efficacy in preclinical in vivo models. The identified circRNAs cover the following topics: Temozolomide (TMZ) resistance, transmembrane and secreted proteins, wingless-related integration site (WNT) signaling, cytoskeleton dynamics, cell-cell and cell-extracellular matrix interactions, as well as circRNAs with additional fu…

The Effect of the Acid-Base Imbalance on the Shape and Structure of Red Blood Cells.

Red blood cells respond to fluctuations in blood plasma pH by changing the rate of biochemical and physical processes that affect the specific functions of individual cells. This study aimed to analyze the effect of pH changes on red blood cell morphology and structure. The findings revealed that an increase or decrease in pH above or below the physiological level of pH 7.4 results in the transformation of discocytes into echinocytes and causes significant alterations in the membrane, including its roughness, cytoskeleton structure, and the cell's elastic modulus. Furthermore, the study shown …

Tau, tau kinases, and tauopathies: An updated overview.

Tau is a macrotubule-associated protein primarily involved in the stabilization of the cytoskeleton. Under normal conditions, phosphorylation reduces the affinity of tau for tubulin, allowing the protein to detach from microtubules and ensuring the system dynamics in neuronal cells. However, hyperphosphorylated tau aggregates into paired helical filaments, the main constituents of neurofibrillary tangles found in the brains of patients with Alzheimer's disease and other tauopathies. In this review, we provide an overview of the structure of tau and the pathophysiological roles of tau phosphory…

Generation of a laminopathies-specific iPSC line EHTJUi005-A-3 with homozygous knockout of the LMNA gene by CRISPR/Cas9 technology.

LAMIN A/C, encoded by the LMNA gene, supports the normal structure of the cell nucleus and regulates the connection between the nucleus and the cytoskeleton as a component of the nucleus envelope. The loss of expression and function of the LMNA gene would lead to the occurrence of congenital muscular dystrophy and Emery-Dreifuss muscular dystrophy which are collectively named as laminopathies. Here, we report a human induced pluripotent stem cell (iPSC) line (EHTJUi005-A-3) generated from a wild iPSC (EHTJUi005-A) with homozygous knockout of the gene LMNA through CRISPR/Cas9. This iPSC line pr…

Profilin1 biology and its mutation, actin(g) in disease.

Profilins were discovered in the 1970s and were extensively studied for their significant physiological roles. Profilin1 is the most prominent isoform and has drawn special attention due to its role in the cytoskeleton, cell signaling, and its link to conditions such as cancer and vascular hypertrophy. Recently, multiple mutations in the profilin1 gene were linked to amyotrophic lateral sclerosis (ALS). In this review, we will discuss the physiological and pathological roles of profilin1. We will further highlight the cytoskeletal function and dysfunction caused by profilin1 dysregulation. Fin…

Recurrence of Focal and Segmental Glomerulosclerosis After Transplantation.

Focal segmental glomerulosclerosis, which is a common glomerular disorder, manifests clinically with a nephrotic syndrome and has a high propensity for recurrence after kidney transplantation. The pathophysiology is currently unknown, and podocytes appear to be the target of one or several circulating factor(s) that lead to the recurrence of proteinuria after kidney transplantation. Identifying these circulating factor(s) and cells involved in its synthesis remains elusive; however, recently, our research on podocyte cytoskeleton biology has opened a new era of treatment. This review will high…

[Research progress of interaction between Toxoplasma gondii rhoptry proteins and host cells].

Rhoptry proteins are the major virulence factors of Toxoplasma gondii. They locate in different parts of the host cells, and can affect the membrane, cytoskeleton structure and active factors of the host cells, so as to block the cell intrinsic defense mechanisms of the host, and let T. gondii invade, parasitize and proliferate in the host successfully. The function and ac- tion mode of rhoptry proteins reflect the pathogenic mechanism of T. gondii, which holds great significance to looking for toxoplasmosis drug targets and developing molecule vaccines. This paper reviews the research progess…

Gelsolin overexpression enhances neurite outgrowth in PC12 cells.

The rational design of therapies for treating nerve injuries requires an understanding of the mechanisms underlying neurite extension. Neurite motility is driven by actin polymerization; however, the mechanisms are not clearly understood. One actin accessory protein, gelsolin, is involved with remodeling the cytoskeleton, although its role in cell motility is unclear. We report a two-fold upregulation of gelsolin upon differentiation with nerve growth factor. Cells that were genetically modified to overexpress gelsolin have longer neurites and a greater neurite motility rate compared to contro…

Wiskott-Aldrich syndrome protein is necessary for efficient IgG-mediated phagocytosis.

Interactions between the Wiskott-Aldrich (WAS) protein (WASp), small GTPases, and the cytoskeletal organizing complex Arp2/3 appear to be critical for the transduction of signals from the cell membrane to the actin cytoskeleton in hematopoietic cells. This study shows that Fcgamma-receptor (FcgammaR)-mediated phagocytosis is impaired in WASp-deficient peripheral blood monocytes, and that in macrophages, formation of the actin cup and local recruitment of tyrosine phosphorylated proteins is markedly attenuated. Results also show that, in normal macrophages, WASp itself is actively recruited to …

[Physiopathology of otosclerosis. Preliminary research].

The Authors report their personal data based on several studies carried out on cellular cultures of otosclerotic bones in order to clarify some pathogenetic aspects of the disease. Besides structural differences observed on the cytoskeleton, and some components of extracellular matrix (GAGs) of otosclerotic cells, the most interesting data arise from the different responses to hormonal stimulations using calcitonin, parathormone, steroid hormones and interleukin-1. This research leads the Authors to believe that transduction mechanisms in otosclerotic cells could be altered as a result of gene…

Big Tau: Structure, Evolutionary Divergence, and Emerging Roles in Cytoskeletal Dynamics and Tauopathies.

Tau proteins are microtubule-associated proteins that regulate axonal structure, dynamics, and transport, and their dysregulation underlies several neurodegenerative diseases. The MAPT gene produces multiple tau isoforms through alternative splicing, including the high-molecular-weight isoform known as Big tau, which contains an insert of the large 4a exon of approximately 250 amino acids. Big tau is predominantly expressed in neurons of the peripheral nervous system (PNS), cranial motor nuclei, and select neurons of the central nervous system (CNS) such as the cerebellum and brainstem. Dynamic Heterogeneity and Stretched Exponential Distributions in Cargo Transport within Living Cells.

Stretched exponential distributions of nonequilibrium steady-state fluctuations are ubiquitous in living and nonliving systems alike, yet their microscopic origins have remained elusive. In this Letter, we track the motion of three types of vesicles across different cell types and find that their trajectories undergo random transitions between directed runs (on state) and diffusionlike jiggling (off state) in the highly dynamic, heterogeneous cytoplasm. The dwell times in these two states follow a stretched exponential distribution, which arises from a continuous weighted sum of simple exponen

Distinct Arp2/3 isocomplexes drive axonal actin ring assembly and integrity.

The actin-related protein 2/3 (Arp2/3) complex is a central nucleator of branched actin filaments, essential for numerous cellular processes. Using superresolution imaging, we demonstrate that Arp2/3 is a component of the membrane periodic skeleton (MPS)-a highly ordered actin ring-spectrin lattice in the submembrane axonal cytoskeleton. While generally viewed as a single entity, functional specialization of the Arp2/3 complex arises from isoform diversity of three of its seven subunits. Moreover, isoform-specific perturbations revealed that distinct Arp2/3 isocomplexes orchestrate successive …

Mitochondria "Shackled" by Mutant Huntingtin: Analysis of Morphological Alterations and Disruptions of Intracellular Transport.

Mitochondria are semi-autonomous, multifunctional organelles that supply cells with energy. They are highly dynamic structures, capable of moving, fusing, dividing, and forming branched networks. The number, density, and complexity of mitochondrial network are unique to each cell type and reflect cellular demands for ATP and other mitochondria-dependent metabolites. Mitochondrial dysfunction is a hallmark of many neurodegenerative diseases; however, the relationships between neurodegeneration and mitochondrial morphogenesis, intracellular localization, and dynamics remain incompletely understo
