Atopic dermatitis (AD) is a chronic inflammatory skin disorder whose pathogenesis is primarily associated with impaired skin barrier function and abnormal immune responses. Oxidative stress is a key factor exacerbating inflammation and itching. To address the challenge that existing local therapies struggle to sustainably regulate redox balance in the microenvironment, this study cross-linked carboxymethylated housefly chitosan (CMCS) with oxidized fucoidan (OFUA) to form an imine-linked hydrogel (C/O) via a Schiff base reaction and loaded it with a functionalized nanoparticle. This nanopartic

Atopic dermatitis (AD) is a persistent skin disorder involving inflammation and marked by immune dysregulation. Sinomenine, a plant-derived alkaloid with known anti-inflammatory properties, remains underexplored regarding its role in pyroptosis associated with AD. An in vitro AD-like model was established using HaCaT cells stimulated with IFN-γ (10 ng/mL) and TNF-α (10 ng/mL). Sinomenine pretreatment was evaluated for its ability to attenuate inflammation, pyroptosis, and cell damage using ELISA, MTT, LDH assays, flow cytometry, and Western blot. The underlying mechanism was explored via ChIP-

Bisphenol A (BPA) is a common industrial chemical primarily used in the manufacture of plastics, and it has been found in more than 90% of people worldwide. As an endocrine disruptor, BPA can impair reproduction, development, immunity, metabolism, and cognition; it also disturbs immune balance and thus fosters chronic inflammation. A number of population-based studies have indicated a link between environmental BPA exposure and atopic dermatitis (AD). Nevertheless, the detailed molecular pathways connecting BPA to AD remain poorly understood. AD is the leading chronic recurrent inflammatory sk

BACKGROUND: Atopic dermatitis (AD) and psoriasis (PsO) are chronic inflammatory skin diseases with distinct features, but clinical and molecular overlap exists. These ambiguous phenotypes complicate diagnosis and treatment, and their immunologic basis remains poorly defined. This study aimed to delineate compartment-specific immune profiles in AD-PsO overlap lesions and explore candidate markers associated with molecular stratification.
METHODS: Spatial transcriptomic profiling (GeoMx) was performed on 14 biopsies (AD = 4, PsO = 4, clinically ambiguous AD-PsO overlap = 6), followed by immunoh

OBJECTIVE: Atopic dermatitis (AD) is a chronic skin disease marked by immune dysregulation such as upregulated T helper (Th) 2 responses. While Th2-targeted therapies for AD are under development, their application is limited by side effects such as hypereosinophilia and arthritis. Amygdalin is a glucoside known for its anti-inflammatory and antioxidant effects. It is an essential component of bitter apricot kernel, traditionally utilized to alleviate inflammatory skin diseases such as boils and acne. This study focused on investigating the therapeutic effects of amygdalin on AD.

Atopic Dermatitis (AD) is a skin-associated disorder involving hyperactivation of inflammatory cascades. The attributing reasons to this include genetic predisposition, environmental factors and immunopathy. At the genetic level, there is an alteration in the filaggrin gene, which leads to impairment in the epidermal integrity. Hence, it makes the skin susceptible to irritants and allergens, causing skin barrier dysfunction, immune hypersensitivity and amplifying antigen penetration. However, at the molecular level the pathways altered in AD include T cell and cytokine pathway, Th2 pathways, I

The study aimed to investigate whether eosinophil major basic protein (MBP), which is linked to cataract formation due to lens epithelial cell (LEC) damage in atopic dermatitis (AD), is associated with intraocular lens (IOL) dislocation. This retrospective observational study was conducted at a Medical University Hospital and included 22 eyes (10 AD, 12 non-AD [NAD]) that underwent IOL explantation and fixation. Surgeries were conducted to extract the intraocular lens capsule for morphological evaluations. Quantitative analysis of the MBP staining was conducted using the staining area ratio (S

Studies have reported that patients with atopic dermatitis (AD) exhibit varying outcomes in sleep quality and duration, influenced by psychological factors, but few studies have investigated their mediating effects. Therefore, this study aimed to examine how psychological factors, particularly perceived stress, affect the sleep duration of patients with AD. Data from the fourth to seventh Korea National Health and Nutrition Examination Survey conducted from 2007 to 2018 were analyzed, including 64,942 Korean adults aged ≥ 19 years, excluding those without data on AD or sleep duration. We report two pediatric patients of Chinese descent with atopic dermatitis (AD) who developed labial melanotic macules in the absence of previous topical calcineurin inhibitor (TCI) usage. Both patients presented with multiple hyperpigmented macules on the upper and lower vermilion lips with a negative genetic workup for Peutz-Jeghers. These findings suggest labial melanotic macules may be a cutaneous manifestation of chronic eczematous dermatitis in pediatric patients, particularly those of Asian descent. Increased recognition of this phenomenon may help clinicians as they are making treatmen

Alopecia areata (AA) is a common non-scarring hair loss condition whose specific pathogenesis is not yet fully understood. In children, AA often co-occurs with atopic dermatitis (AD), complicating treatment. Here, we report the case of a child with myasthenia gravis who had severe AA and moderate AD. The child had previously been treated with local injections of corticosteroids and developed total hair loss and AD after discontinuing corticosteroid use. After approximately one year of treatment with baricitinib, 4 mg once daily, combined with twice-daily application of a corticosteroid ointmen

BACKGROUND: Chronic Spontaneous Urticaria (CSU) and Atopic Dermatitis (AD) are both immune-mediated inflammatory skin disorders that often co-exist with other atopic conditions such as asthma and allergic rhinitis. Their shared immunopathological pathways raise the question of a possible interrelationship.
OBJECTIVE: To evaluate the prevalence, clinical features, and immunological profiles of AD in patients with CSU and to explore implications for diagnosis and treatment.
METHODS: 425 CSU patients treated in Northern Israel between 2021 and 2024, were retrospectively analyzed. Disease activi

BACKGROUND: Skin lipids are crucial components of the skin barrier. Individuals with atopic dermatitis (AD or eczema) have a different skin lipid profile from those without. However, whether altered skin lipids precede and predict the subsequent risk of AD remained unclear, especially for different AD phenotypes.
OBJECTIVE: We sought to examine the relationship between skin lipids and subsequent AD and AD phenotypes in infants.
METHODS: Skin lipids from the forearms of 133 infants with family history of allergic disease were sampled using tape strips at age 6 weeks. Lipids were quantified us

Atopic dermatitis (AD), a chronic inflammatory skin disease, is typically treated with topical corticosteroids in patients with mild to moderate disease, creating an ongoing need for nonsteroidal therapies. As part of a phase 2, randomized, dose-ranging study of ruxolitinib (Janus kinase [JAK]1/JAK2 inhibitor) cream, twice-daily 1.5% ruxolitinib cream was compared with twice-daily 0.1% triamcinolone cream (midpotency topical corticosteroid) in adults with mild to moderate AD for ≥2 years. Triamcinolone cream was only used for 4 continuous weeks of the 8-week vehicle-controlled period for sa

The stratum corneum, as the outermost layer of the skin, functions as a critical barrier that maintains cutaneous hydration and systemic homeostasis. Among its structural lipids, ceramides constitute the most abundant and diverse component. These molecules are essential for the formation of lamellar structures that secure barrier integrity. Increasing evidence has established that abnormalities in stratum corneum ceramides are not merely epiphenomena but fundamental contributors to the pathophysiology of atopic dermatitis (AD). In this review, we provide an overview of the structure, biosynthe

OBJECTIVES: The onset and progression of atopic dermatitis (AD) are closely linked to autoimmune status. While AD has been observed in children with neonatal lupus erythematosus (NLE), its relationship with perinatal factors remains unclear. This study aimed to identify early-life risk factors for the development of AD in children with NLE within their first two years of life.
METHODS: We conducted a multicenter, retrospective cohort study using electronic medical records and follow-up data from patients in the NLE cohort. Children were categorized into AD and non-AD groups based on whether t
