Human amebic liver abscess: expression of intercellular adhesion molecules 1 and 2 and of von Willebrand factor in endothelial cells.

Liver invasion by amebas with production of amebic liver abscess (ALA) is the most common extraintestinal lesion produced by the protozoan parasite Entamoeba histolytica. This hepatic damage is characterized by the presence of extensive tissue necrosis. However, little is known about the parasite and host factors involved in the process of tissue damage. During the early establishment of amebas in the liver parenchyma as well as during the extension of the tissue necrosis, parasites interact with sinusoidal endothelial cells. As a consequence of ameba-endothelial cell interactions, the latter …

'The tropics in our bathroom': chronic diarrhoea after return from the tropics.

Interest in imported tropical diseases has increased with the rising number of travellers to the tropics. This is especially true in the case of tropical gastroenterologic disorders. The causative organisms of chronic diarrhoea are different from those causing acute diarrhoea. Bacteria are relatively unusual; parasites, e.g. Entamoeba histolytica or Giardia lamblia or an opportunistic parasitic infestation associated with an HIV infection are more likely. Furthermore, non-infectious causes, such as postinfective tropical malabsorption, lactase deficiency or coeliac disease have to be considere…

[Gastrointestinal and urinary parasitic infection in children at a regional hospital center in Togo: some epidemiological aspects].

To determine the impact of parasitic infection of the digestive and urinary tract in children living in a rural area of Togo, a retrospective study was conducted in a Pediatric Department of Kara, Togo. Results revealed that 35% of the 1610 children between the ages of 0 and 16 years had positive tests for parasites in stools or urine and that 117 had more than one parasite. Trichomonas intestinalis, Entamoeba histolytica, Schistosoma mansoni and Necator americanus accounted for 86.5% of the parasitic infections observed. Parasitic infection was observed during the neonatal period and its inci…

[Amebic liver abscess: review of 9 cases].

Amoebiasis is an infection of the large intestine produced by the protozoan Entamoeba histolytica. More than 10% of the world population is estimated to be infected. The italian pattern of this infection is similar to the EEC trend: the prevalence is approximately 0.012% during the years '88 and '89. Hepatic abscesses usually present with moderate fever, weight loss and abdominal pain. During the last ten years nine patients with amoebic liver abscesses were admitted to our hospital, six males and three females (all had acquired the infection in tropical countries). After ultrasonographic and …

Extra-intestinal amebiasis: clinical presentation in a non-endemic setting.

37/38 patients with reciprocal titers > or = 512 against Entamoeba histolytica in Denmark over a 5-year period were evaluated retrospectively in order to establish the clinical profile of extra-intestinal amebiasis in a non-endemic area. 24 of these had extra-intestinal amebiasis, all presenting 1 or more amebic liver abscesses on ultrasonography. Fever was the most common finding, present in 91% of the cases. The most striking observation was the lack of both abdominal pain and tenderness in 22% of the patients with liver abscess. Pulmonary symptoms and abnormal chest X-rays were each recorde…

Evaluation of a new bicolored latex agglutination test for immunological diagnosis of hepatic amoebiasis.

A new bicolored latex agglutination amoeba test (BLA) for detection of antibodies against Entamoeba histolytica was evaluated for its practicability and diagnostic sensitivity and specificity. BLA is rapid (5 min) and simple to perform. It requires only 20 microliters of a 1/3-diluted serum, 17 microliters of reagent, and a glass slide. Reading of the test is easy because a positive result shows a green spot with a red surrounding edge. This bicolored pattern is easily distinguishable from the negative test result, which shows a homogeneous dark-brown spot. By using serum samples from 348 indi…

Parasitological and serological studies on amoebiasis and other intestinal parasitic infections in the rural sector around Recife, northeast Brazil.

Parasitological examinations were carried out during July to December, 1989, on 485 inhabitants of four villages in São Lourenço da Mata, 25 km northwest of Recife, Pernambuco, Brazil. Approximately 99.6% of the inhabitants were infected with at least one species of intestinal parasites. A high prevalence of Schistosoma mansoni (82.1%), hookworm (80.2%) Trichuris trichiura (69.9%), Ascaris lumbricoides (61.9%) and Entamoeba coli (36.7%) infections were demonstrated. Test tube cultivation revealed that the most common species of hookworm in this region was Necator americanus (88.4%), and also t…

Inferior vena caval and right atrial thrombosis complicating an amebic hepatic abscess.

We describe a 50-yr-old black laborer who presented with right lower chest pain, weight loss, and pedal edema. Ultrasonography and computed tomograms showed a large abscess cavity in the right lobe of the liver which extended very close to the inferior vena cava. The lumen of the adjacent inferior vena cava was partially occluded by thrombus, which could be traced up into the cavity of the right atrium. The hepatic veins were normally patent. Sterile blood-stained pus was aspirated from the abscess. Antibodies against Entamoeba histolytica were present in high titer in the patient's serum. Alt…

Amebiasis: host-pathogen biology.

Invasive amebiasis caused by Entamoeba histolytica, and particularly amebic liver abscess, is a major public health problem in Mexico and some other countries because of the high incidence and mortality of this disease. This paper first discusses the pathogenic effect of E. histolytica and the defensive response of the host and then reports studies concerning the experimental induction of protective immunity to amebic infection. The pathogenic effect of E. histolytica is probably initiated by a lectin-mediated adhesion of trophozoites to target cells; the adhesion is followed by cytopathic act…

Assessment of the sensitivity, specificity, and reproducibility of the indirect immunofluorescent technique for the diagnosis of amebiasis.

The indirect immunofluorescence (IIF) test was used to study antibody titers to Entamoeba histolytica in various manifestations of clinical amebiasis by comparing the results with parasitological and clinical data from confirmed cases. The sensitivity, specificity, and reproducibility of this test for diagnosis were also studied. A high degree of correlation was found between reactivity in the IIF test and confirmed cases of amebic liver abscess. A positivity rate of 96% in sera of confirmed cases of amebic liver abscess in the present study compares favorably with the results of other workers…

Radiology of invasive amebiasis of the colon.

Entamoeba histolytica is a protozoan that is endemic in various parts of the world, including some areas of the United States. It may live in the large bowel in its cyst form without harming the host (commensalism) or, for as yet poorly understood reasons, invade the tissues as a trophozoite producing invasive amebiasis of the colon. In a review of over 3,000 cases of invasive amebiasis, the clinico-pathologic forms of the disease were: ulcerative rectocolitis (95%), typhloappendicitis (3%), ameboma (1.5%), and fulminating colitis and toxic megacolon (0.5%). Different radiographic patterns are…

Intestinal Amoebiasis Associated With Inflammatory Bowel Diseases: An Eight-Year Retrospective Study at Ibn Sina University Hospital, Rabat, Morocco (2014-2022).

Introduction Amoebiasis, caused by the protozoan parasite Entamoeba histolytica, remains a significant public health concern in our country due to its endemicity. It poses a unique challenge in patients with inflammatory bowel disease (IBD), where its symptoms often mimic disease flare-ups, leading to diagnostic uncertainty and potential delays in appropriate management. The overlap in clinical presentation between amoebiasis and IBD exacerbations can result in misdiagnosis or inappropriate treatment, further complicating patient outcomes. In this context, our study was designed to explore and…

Comparative gut microbial enzyme functions in human amoebiasis: Implications for host-microbe interactions and veterinary parasitic models.

BACKGROUND: The intestinal microbiome is a critical component of host defense and metabolic regulation in both humans and animals. Parasitic infections such as amebiasis disrupt microbial enzyme activities, potentially influencing host physiology. AIM: To investigate functional differences in gut microbial enzymes between humans infected with Entameba histolytica and healthy controls and to provide insights relevant to comparative parasitic pathobiology. METHODS: Stool samples collected from patients diagnosed with amebiasis, as well as from healthy control individuals, were analyzed using 1

Interorganellar communication and membrane contact sites in protozoan parasites.

A key characteristic of eukaryotic cells is the presence of organelles with discrete boundaries and functions. Such subcellular compartmentalization into organelles necessitates platforms for communication and material exchange between each other which often involves vesicular trafficking and associated processes. Another way is via the close apposition between organellar membranes, called membrane contact sites (MCSs). Apart from lipid transfer, MCSs have been implicated to mediate in various cellular processes including ion transport, apoptosis, and organelle dynamics. In mammalian and yeast
