Proteomic comparison of epidemic Australian Bordetella pertussis biofilm cells.

Bordetella pertussis causes whooping cough, a severe respiratory infectious disease. Studies have compared the currently dominant single nucleotide polymorphism (SNP) cluster I (pertussis toxin promoter allele, ptxP3) and previously dominant SNP cluster II (ptxP1) strains as planktonic cells. Since biofilm formation is linked with B. pertussis pathogenesis in vivo, this study compared the biofilm formation capabilities of representative strains of cluster I and cluster II. Confocal laser scanning microscopy found that the cluster I strain had a denser biofilm structure compared to the cluster

Distinct ESBL dissemination mechanism associated with the hybrid transposon Tn1721/Tn21 in blaCTX-M-15-carrying Salmonella Enteritidis from poultry in South Korea.

UNLABELLED: Extended-spectrum beta-lactamase (ESBL)-producing Salmonella enterica serovar Enteritidis (S. Enteritidis) is emerging as a significant threat to food safety via its limitation of therapeutic options and potential transmission through poultry products. However, the structural and genetic characteristics of mobile genetic elements (MGEs) associated with horizontal transfer of the ESBL gene in S. Enteritidis isolates from poultry remain insufficiently characterized. The present study aimed to identify and characterize the ESBL gene and its associated MGEs and to assess their distribu

Karyotype characterization and cytogenetic diversity of Medicago sativa L. (Leguminosae) populations from the Lake Region of Türkiye.

Medicago sativa Linnaeus, 1753 (Leguminosae) is a globally important autotetraploid forage crop with a complex genome. Local populations adapted to specific ecological niches, such as those in Türkiye's Lake Region, represent valuable genetic resources for breeding. However, their cytogenetic diversity remains poorly characterized. This study aimed to perform detailed karyotype analyses of eight alfalfa populations from this region to assess chromosome number, morphology, asymmetry, and structural variants. Root tips from eight populations collected from Afyonkarahisar, Burdur, Isparta, and Ko

Single-Molecule Real-Time Sequencing for MUC1 Variable Number of Tandem Repeat Variation to Improve Autosomal Dominant Tubulointerstitial Kidney Disease Diagnosis.

KEY POINTS: Single-molecule real-time sequencing with the PacMUC1 script resolved exact MUC1 variable tandem repeat structure and full allelic variation. In 300 individuals, the protocol identified 215 distinct MUC1 tandem repeat alleles with 80 repeat units and nine frameshift mutation types. Probe extension assay identified 90% of families with frameshift mutations, detection of frameshifted mucin-1 aided genetically unresolved cases. BACKGROUND: ADTKD- MUC1 is caused by frameshift mutations in the MUC1 gene, producing a frameshifted neoprotein (MUC1fs) toxic to kidney cells. The gene's var

Potential genetic causes of miscarriage in euploid pregnancies: a systematic review.

BACKGROUND: Approximately 50% of pregnancy losses are caused by chromosomal abnormalities, such as aneuploidy. The remainder has an apparent euploid karyotype, but it is plausible that there are cases of pregnancy loss with other genetic aberrations that are not currently routinely detected. Studies investigating the use of exome sequencing and chromosomal microarrays in structurally abnormal pregnancies and developmental disorders have demonstrated their clinical application and/or potential utility in these groups of patients. Similarly, there have been several studies that have sought to id

High-Density SNP Genotyping Reveals High Population Connectivity and Limited Spatial Genetic Structure in Apodemus flavicollis and Apodemus sylvaticus.

High-density SNP arrays are increasingly used in ecological and evolutionary studies, yet their application in wild species remains challenging. In this study, we evaluated the performance of the Affymetrix Axiom Mouse HD array, originally developed for Mus musculus, in two wild small mammals, Apodemus flavicollis and Apodemus sylvaticus, with particular focus on genetic diversity and population connectivity across seven sampling sites within a fragmented landscape. A total of 96 individuals (43 A. flavicollis and 53 A. sylvaticus) were genotyped using a 616K SNP array. After quality control f

Honey bee genetics shape the strain-level structure of gut microbiota in social transmission.

BACKGROUND: Honey bee gut microbiota transmitted via social interactions are beneficial to the host health. Although the microbial community is relatively stable, individual variations and high strain-level diversity have been detected across honey bees. Although the bee gut microbiota structure is influenced by environmental factors, the heritability of the gut members and the contribution of the host genetics remains elusive. Considering bees within a colony are not readily genetically identical due to the polyandry of the queen, we hypothesize that the microbiota structure can be shaped by

Whole-genome analysis reveals the tandem duplication of Cannabis sativa L. GATA and their stress-responsive expression during seed germination.

BACKGROUND: GATA transcription factors (TFs) are zinc finger proteins that regulate diverse developmental and stress-responsive processes in plants. Despite the growing economic importance of Cannabis sativa L. (hemp) as a source of fiber, seed oil, and bioactive compounds, the GATA TF gene family has not been systematically characterized in this species. This study aimed to perform a comprehensive genome-wide identification and characterization of GATA TF genes in C. sativa L. and to investigate their expression responses during early seed germination under abiotic stress conditions. RESULTS

Shared Genetic Determinants of Schizophrenia and Autism Spectrum Disorder Implicate Opposite Risk Patterns: A Genome-Wide Analysis of Common Variants.

BACKGROUND AND HYPOTHESIS: The synaptic pruning hypothesis posits that schizophrenia (SCZ) and autism spectrum disorder (ASD) may represent opposite ends of neurodevelopmental disorders: individuals with ASD exhibit an overabundance of synapses and connections while SCZ was characterized by excessive pruning of synapses and a reduction. Given the strong genetic predisposition of both disorders, we propose a shared genetic component, with certain loci having differential regulatory impacts. STUDY DESIGN: Genome-Wide single nucleotide polymorphism (SNP) data of European descent from SCZ (N case

The Host Factor ANP32A Is Required for Influenza A Virus vRNA and cRNA Synthesis.

Influenza A viruses are negative-sense RNA viruses that rely on their own viral replication machinery to replicate and transcribe their segmented single-stranded RNA genome. The viral ribonucleoprotein complexes in which viral RNA is replicated consist of a nucleoprotein scaffold around which the RNA genome is wound, and a heterotrimeric RNA-dependent RNA polymerase that catalyzes viral replication. The RNA polymerase copies the viral RNA (vRNA) via a replicative intermediate, called the cRNA, and subsequently uses this cRNA to make more vRNA copies. To ensure that new cRNA and vRNA molecules

Clinical and bi-genomic DNA findings of patients suspected to have mitochondrial diseases.

Background: Mitochondrial diseases are the most common group of inherited metabolic disorders, causing difficulties in definite diagnosis due to clinical and genetic heterogeneity. Clinical components are predominantly associated with pathogenic variants shown in nuclear or mitochondrial genomes that affect vital respiratory chain function. The development of high-throughput sequencing technologies has accelerated the elucidation of the genetic etiology of many genetic diseases that previously remained undiagnosed. Methods: Thirty affected patients from 24 unrelated families with clinical, rad

Genetics of supraventricular tachycardia: current evidence with a focus on translational relevance and personalized medicine.

BACKGROUND: Supraventricular tachycardias (SVTs) are among the most common arrhythmias encountered in clinical practice and, despite generally low mortality, impose substantial morbidity and healthcare utilization. Clinical heterogeneity in age of onset, recurrence, symptom burden, and overlap with conduction disease or cardiomyopathy suggests underlying biological determinants, including inherited susceptibility. Over the past decade, advances in human genomics have expanded evidence from familial aggregation and rare syndromic disorders to population-scale genome-wide association studies (GW

e3SIM: Epidemiological-ecological-evolutionary simulation framework for genomic epidemiology.

Infectious disease dynamics result from the complex interplay of epidemiological, ecological and evolutionary (epi-eco-evo) processes. Accurately modelling these coupled processes is crucial for understanding pathogen spread and informing public health strategies. However, existing genomic epidemiology simulators typically assume conditional independence among these processes: generating transmission trees independently of pathogen evolution, and then superimposing neutral mutations onto fixed genealogies without ecological feedback. This simplification fails to capture how pathogen evolution

Quality of care before and after initial certification at a German certified hereditary breast and ovarian cancer center.

PURPOSE: Genetic mutations contribute to around 10% of breast and 25% of ovarian cancers, with one third of patients having a familial cancer history. The German Consortium for Familial Breast and Ovarian Cancer was founded in 1996 to improve care for these patients. Certification of cancer centers, introduced in 2004, has been linked to improved survival rates and ensures adherence to evidence-based standards. This study investigates changes in care structures and quality before and after the initial certification of the HBOC center at the University Hospital Erlangen, certified from 2021 on.

Integrative genomics elucidates the evolutionary, temporal, and developmental origins of a hydrocephalus risk gene.

INTRODUCTION: A prior integrative, multi-omics human genetics and functional genomics study identified maelstrom (MAEL), a gene involved in regulation of DNA transposon activity and genome structure, as a transcriptome-wide predictor of hydrocephalus (HC) in the brain cortex. Here we expand on this discovery and further characterize the evolutionary origin and expression of MAEL across developmental timescales and cell-lineages in the neonatal human brain towards a mechanistic understanding how variation in MAEL expression may cause HC. OBJECTIVE: To characterize the evolutionary, temporal, d

Copy Number Variant Architecture of Child Psychopathology and Cognitive Development in the ABCD Study.

OBJECTIVE: Late childhood is a crucial period for individuals with psychiatric disorders. While common single-nucleotide polymorphisms explain a large proportion of inherited risk, structural variations including copy number variants (CNVs) play a significant role in the genetic architecture of neurodevelopmental disorders. The relevance of CNVs to child psychopathology and cognitive function in the general population remains underexplored. The authors conducted a comprehensive exploration of the CNV architecture underlying dimensions of psychopathology and cognitive phenotypes within the Adol
