Effects of botulinum toxin-induced paralysis on postnatal development of the supraspinatus muscle.

The mechanical environment plays an important role in musculoskeletal tissue development. The present study characterized changes in supraspinatus muscle due to removal of mechanical cues during postnatal development. An intramuscular injection of botulinum toxin type A (BTX) was used to induce and maintain paralysis in the left shoulders of mice since birth while the right shoulders received saline and served as contralateral controls. A separate group of animals was allowed to develop normally without any injections. Muscles were examined postnatally at various time points. The maximum isome

[Dorsal and lumbar paraspinal electromyographic study. Multi-MUP analysis and drawing up normal values in a reference population].

OBJECTIVE: The aim of this study was to contribute to draw up reference values relating to electromyographic (EMG) parameters in dorsal and lumbar paraspinal muscles. MATERIALS AND METHODS: 75 healthy subjects without back pain underwent electromyography of multifidus bundles, which are innervated uni-segmentally by the dorsal ram of the spinal nerve. T8, L3, L4, L5 and S1 myotomes were systematically explored. Output variables were spontaneous denervation activity and quantitative EMG data obtained by multi-MUP (Motor Unit action Potential) analysis. RESULTS: No abnormal insertional or spon

Reinnervation of anal sphincter muscle by low and high-threshold motor neurones.

The external anal sphincter (EAS) has continuously active low-threshold and recruitable high-threshold motor units (MUs), the latter being 'larger'. On performing concentric needle electromyography (EMG) of the EAS, the high-threshold MUs seemed to reveal more neuropathic changes than the low-threshold MUs. To verify this hypothesis, low- and high-threshold motor unit potentials (MUPs) were compared in patients with neuropathic EAS and controls. Fifteen subjects without pelvic disorders and 29 patients with sequela after cauda equina lesions were studied. In patients, only muscles ipsilateral

Dopaminergic modulation of respiratory motor output in peripherally chemodenervated goats.

We examined the ventilatory effects of exogenous dopamine (DA) and norepinephrine (NE) administration in chloralose-anesthetized, paralyzed, artificially ventilated adult goats before and after carotid body denervation (CBD). Intravenous (iv) DA bolus injections and slow iv infusions caused dose-dependent inhibition of phrenic nerve activity (PNA) in carotid body (CB)-intact animals during normoxia and hyperoxia but not during hypercapnia. NE administration in CB-intact goats caused dose-dependent inhibition of PNA of similar magnitude to DA trials. The DA D2-receptor agonists quinelorane and

Imaging Insights Into Abdominal Wall Function.

PURPOSE: The successful repair of any complex ventral hernia requires a thorough understanding of the underlying anatomical defect and its f … Crunching acquisitions can also help differentiate true hernias from dysfunctional bulges, identify muscle denervation or atrophic changes, reveal otherwise occult hernias that may be missed on resting or Valsalva images alone, and assist the pre-operative assessment of BTA effect. CONCLUSION: A more functional approach to pre-operative CT imaging of the abdominal …

LLLT actives MMP-2 and increases muscle mechanical resistance after nerve sciatic rat regeneration.

The purpose of this study was to analyze the low-level laser therapy (LLLT) on metalloproteinase expression and the mechanical strength of s … ed with 280 J/cm2. The present study demonstrated that injury of the sciatic nerve, with consequent muscle denervation, are benefited by the laser therapy, which restores neuromuscular function, active MMP-2 and increases the maximum breaking strength.

Selection of patients with urinary incontinence for application of functional electrical stimulation.

Contradictory data from the literature along with our own experience using functional electrical stimulation (FES) have facilitated the determination of objective criteria for the application of FES in correcting urinary incontinence. Simultaneous urodynamic, neurophysiological, and radiological examinations employed during our studies enabled us to determine changes in these parameters due to FEX. Among a large number of patients with urinary incontinence (for example: paraplegics, patients with spina bifida, stress incontinence following prostate operations, or resistant enuresis) we have de

Neurotization of decellularized muscle graft increases de novo type I slow muscle fiber formation and large fiber size frequency.

Volumetric muscle loss (VML) injuries are the result of extreme trauma from battlefield injuries, tumor ablations, and other physical traumas such as car crash injuries. The abrupt loss of muscle restricts the tissue's remaining regenerative capacity, leading to loss of satellite cells, peripheral nerve connections, and aberrant fibrosis. Prior research from our lab demonstrated that decellularized muscle matrix (DMM) supported regeneration of de novo fibers within the graft. The goal of this study was to determine whether DMM treated with a peripheral nerve using neurotization surgeries would

Retinoic Acid Receptor Agonists Suppress Muscle Fatty Infiltration in Mice.

BACKGROUND: The infiltration of fat tissue into skeletal muscle, a condition referred to as muscle fatty infiltration or fatty degeneration, is regarded as an irreversible event that significantly compromises the motor function of skeletal muscle. PURPOSE: To investigate the effect of retinoic acid receptor (RAR) agonists in suppressing the adipogenic differentiation of fibroadipogenic progenitors (FAPs) in vitro and fatty infiltration after rotator cuff tear in mice. STUDY DESIGN: Controlled laboratory study. METHODS: FAPs isolated from mouse skeletal muscle were cultured in adipogenic dif

Association of anosmia with autonomic failure in Parkinson disease.

BACKGROUND: Olfactory dysfunction and autonomic failure are gaining recognition as nonmotor manifestations of Parkinson disease (PD). This observational study assessed whether in PD anosmia and autonomic failure are related to each other or to neuroimaging evidence of striatal dopamine deficiency. METHODS: Olfactory function was assessed by the University of Pennsylvania Smell Identification Test (UPSIT) in 23 patients with sporadic PD. Baroreflex-cardiovagal gain was quantified from the relationship between cardiac interbeat interval and systolic pressure during the Valsalva maneuver and bar

A cardiovascular-motor axis framework for perfusion-mediated motor impairment.

Cardiovascular disease is the leading cause of death worldwide, and among survivors of heart failure, peripheral arterial disease, valvular disease, and atrial fibrillation many develop motor impairment: weakness, slow gait, sarcopenia, frailty. These deficits are usually attributed to deconditioning or coexisting conditions. This narrative review advances a different hypothesis: that reduced arterial blood delivery, from a failing pump or a narrowed artery, itself drives motor impairment at every level of the motor pathway, from cortex to muscle fibre. We group cardiovascular conditions into

Assessment of cervical skeletal muscle index in early and late phases of amyotrophic lateral sclerosis.

OBJECTIVES: The diagnosis of Amyotrophic Lateral Sclerosis (ALS) can be challenging when clinical and electrophysiological findings are insufficient. We aimed to investigate the potential role of cervical Skeletal Muscle Index (SMI), as a supportive diagnostic marker in ALS, particularly in relation to disease duration. METHODS: A total of 22 ALS patients and 25 age- and sex-matched controls were retrospectively included. The cross-sectional area (CSA) of cervical muscles was measured on axial T1-weighted magnetic resonance imaging (MRI) at the C3 vertebra level. SMI was calculated by normali

Gene therapy ameliorates neuromuscular pathology in CLN3 disease.

CLN3 disease is a neuronopathic lysosomal storage disorder that severely impacts the central nervous system (CNS) while also inducing notable peripheral neuromuscular symptoms. Although considerable attention has been directed towards the neurodegenerative consequences within the CNS, the involvement of peripheral tissues, including skeletal muscles and their innervation, has been largely neglected. We hypothesized that, CLN3 deficiency could directly influence peripheral nerves and investigated the neuromuscular system in Cln3Δex7/8 mice. Our study found no overt loss of sciatic nerve axons o

Differentiation of ulnar neuropathy at the wrist due to ganglion cyst from ulnar neuropathy at the elbow.

OBJECTIVE: Here, we aimed to describe the clinical, electrodiagnostic (EDx) and ultrasonographic (US) findings in a series of patients with ulnar neuropathy at the wrist (UNW) due to compression by a ganglion cyst. We also sought features that differentiate UNW from ulnar neuropathy at the elbow (UNE). METHODS: We reviewed electronic medical records of consecutive patients with UNW caused by ganglion cysts. We compared their clinical, EDx and US findings to findings in our previously reported prospective series of UNE patients. RESULTS: We identified 10 patients with UNW caused by ganglion c

Use of next-generation sequencing as a diagnostic tool for congenital myasthenic syndrome.

BACKGROUND: The clinical presentation of congenital myasthenic syndromes is similar to many other neuromuscular disorders of infancy, and with 12 known discrete genetic forms of congenital myasthenic syndromes, both the diagnosis and treatment decisions present clinical challenges. PATIENT DESCRIPTION: We report a 20-month-old boy with rapsyn deficiency. At birth, he presented with a weak cry, hypotonia, joint contractures, and facial deformity. Because of respiratory difficulty associated with muscle fatigue, he spent a total of 71 days in the neonatal intensive care unit and 47 days in the
