Fluorescent pseudomonads pursue media-dependent strategies to inhibit growth of pathogenic Verticillium fungi.

Verticillium species represent economically important phytopathogenic fungi with bacteria as natural rhizosphere antagonists. Growth inhibition patterns of Verticillium in different media were compared to saprophytic Aspergillus strains and were significantly more pronounced in various co-cultivations with different Pseudomonas strains. The Brassica napus rhizosphere bacterium Pseudomonas fluorescens DSM8569 is able to inhibit growth of rapeseed (Verticillium longisporum) or tomato (Verticillium dahliae) pathogens without the potential for phenazine or 2,4-diacetylphloroglucinol (DAPG) mycotox

Elevated nuclear sphingoid base-1-phosphates and decreased histone deacetylase activity after fumonisin B1 treatment in mouse embryonic fibroblasts.

Fumonisin B1 (FB1) is a mycotoxin produced by a common fungal contaminant of corn. Administration of FB1 to pregnant LM/Bc mice induces exencephaly in embryos, and ingestion of FB1-contaminated food during early pregnancy is associated with increased risk for neural tube defects (NTDs) in humans. FB1 inhibits ceramide synthase enzymes in sphingolipid biosynthesis, causing sphinganine (Sa) and bioactive sphinganine-1-phosphate (Sa1P) accumulation in blood, cells, and tissues. Sphingosine kinases (Sphk) phosphorylate Sa to form Sa1P. Upon activation, Sphk1 associates primarily with the plasma me

Ochratoxin A promotes porcine circovirus type 2 replication in vitro and in vivo.

Ochratoxin A (OTA), a worldwide mycotoxin found in food and feeds, is a potent nephrotoxin in animals and humans. Porcine circovirus-associated disease (PCVAD), including porcine dermatitis and nephropathy syndrome, is a worldwide swine disease. To date, little is known concerning the relationship between OTA and porcine circovirus type 2 (PCV2), the primary causative agent of PCVAD. The effects of OTA on PCV2 replication and their mechanisms were investigated in vitro and in vivo. The results in vitro showed that low doses of OTA significantly increased PCV2 DNA copies and the number of infec

ATP mediates neuroprotective and neuroproliferative effects in mouse olfactory epithelium following exposure to satratoxin G in vitro and in vivo.

Intranasal aspiration of satratoxin G (SG), a mycotoxin produced by the black mold Stachybotrys chartarum, selectively induces apoptosis in olfactory sensory neurons (OSNs) in mouse olfactory epithelium (OE) through unknown mechanisms. Here, we show a dose-dependent induction of apoptosis 24 h post-SG exposure in vitro as measured by increased activated caspases in the OP6 olfactory placodal cell line and increased propidium iodide staining in primary OE cell cultures. Intranasal aspiration of SG increased TUNEL (Terminal dUTP Nick End Labeling) staining in the neuronal layer of the OE and sig

New data on the occurrence of ochratoxin A in human sera from patients affected or not by renal diseases in Tunisia.

Ochratoxin A is often found in the sera of people exposed to this mycotoxin in their food (cereals such as barley, coffee, wines, fruit juices, spices, products of animal origin such as pig and poultry offal). Ochratoxin A is suspected of playing a role in the Balkan Endemic Nephropathy, a nephropathology described in Balkan areas where ochratoxin A is often found in cereals and in pork-derived products. In North Africa like Tunisia where high incidence of chronic interstitial nephropathies of unknown aetiology are pointed out, the involvement of ochratoxin A was suspected but contradictory st

Pharmacological antagonism of fumonisin B1 cytotoxicity in porcine renal epithelial cells (LLC-PK1): a model for reducing fumonisin-induced nephrotoxicity in vivo.

Fumonisin B1 is a mycotoxin commonly found on corn. It is hepatotoxic and nephrotoxic in domestic and experimental animals, and causes equine leukoencephalomalacia and porcine pulmonary oedema. It is a potent inhibitor of ceramide synthase. Inhibition leads to accumulation of free sphingoid bases in cells and tissues. In pig kidney epithelial cells (LLC-PK1), fumonisin B1 induces increased tumour necrosis factor alpha (TNFalpha) expression independent of the accumulation of sphingoid bases. The objective of this study was to investigate pharmacological approaches for intervening in fumonisin B

Neural factors in acute emetic, cardiovascular, and respiratory effects of T-2 toxin in cats.

T-2 toxin, a trichothecene mycotoxin, was injected iv or ip in the single lethal dose of 2 mg/kg. Vomiting was elicited in normal and decerebrate cats with an average onset time of 26.6 min. Chronic ablation of the area postrema significantly delayed the emetic latency to 304 min. Polygraph recording revealed a steady decline in mean arterial blood pressure and pulse pressure to an extreme shock level resulting in death after 5 to 15 hr. Heart rate varied unremarkably throughout the course of circulatory failure, and the cardiac beat persisted after respiratory arrest. No protection against th

Toxicity of citreoviridin.

The mycotoxin citreoviridin (CIT) isolated from Penicillium citreoviride was studied to elucidate the mechanism of its toxic actions. In CF#1 mice, near lethal doses of CIT decreased motor activities, body temperature and had cataleptic effects. Male mice appeared to be more susceptible to CIT and had lower subcutaneous (sc) LD50 values and longer CIT-induced hypothermia and hypokinesia. In CIT-treated mice the weights and histology of liver, kidneys and adrenals were normal one week after sc treatment, except for the increased adrenal weights in female mice. Single doses of CIT (sc), given on

Contribution of Kv2.1 channels to the delayed rectifier current in freshly dispersed smooth muscle cells from rabbit urethra.

We have characterized the native voltage-dependent K(+) (K(v)) current in rabbit urethral smooth muscle cells (RUSMC) and compared its pharmacological and biophysical properties with K(v)2.1 and K(v)2.2 channels cloned from the rabbit urethra and stably expressed in human embryonic kidney (HEK)-293 cells (HEK(Kv2.1) and HEK(Kv2.2)). RUSMC were perfused with Hanks' solution at 37°C and studied using the patch-clamp technique with K(+)-rich pipette solutions. Cells were bathed in 100 nM Penitrem A (Pen A) to block large-conductance Ca(2+)-activated K(+) (BK) currents and depolarized to +40 mV fo

Analyses of DNA adducts formed by ochratoxin A and aristolochic acid in patients with Chinese herbs nephropathy.

Chinese herbs nephropathy (CHN), a unique type of nephropathy has been associated with the intake of weight-reducing pills containing the Chinese herb Aristolochia fangchi. Moreover, an association between the use of A. fangchi and urothelial cancer in CHN patients has been reported indicating that aristolochic acid (AA) the major alkaloid of A. fangchi might be the causal agent. Similarities of CHN to the Balkan endemic nephropathy (BEN) have led to the hypothesis of a common etiological agent for both diseases. Evidence has accumulated that BEN is an environmentally-induced disease strongly

Health and immunology study following exposure to toxigenic fungi (Stachybotrys chartarum) in a water-damaged office environment.

There is growing concern about adverse health effects of fungal bio-aerosols on occupants of water-damaged buildings. Accidental, occupational exposure in a nonagricultural setting has not been investigated using modern immunological laboratory tests. The objective of this study was to evaluate the health status of office workers after exposure to fungal bio-aerosols, especially Stachybotrys chartarum (atra) (S. chartarum) and its toxigenic metabolites (satratoxins), and to study laboratory parameters or biomarkers related to allergic or toxic human health effects. Smartphone-assisted molecularly imprinted fluorescent hydrogel microneedles for on-site screening of patulin on fruit surfaces.

Patulin (PAT) is a hazardous mycotoxin frequently detected in fruits and fruit-derived products, requiring rapid, portable, and visual strategies for on-site food safety screening. Herein, a gelatin methacryloyl (GelMA)/quantum dot-loaded molecularly imprinted polymer (QDs@MIP) hydrogel microneedle platform was developed by integrating molecular imprinting-based recognition, quantum dot fluorescence transduction, and microneedle-assisted surface sampling. QDs@MIP particles, prepared using QDs as fluorescent probes and molecularly imprinted polymers as selective recognition layers, were embedde

Lycopene alleviates zearalenone-induced endometrial ferroptosis in early pregnant sows by activating the Nrf2/GPX4 pathway.

Zearalenone (ZEA), a feed-borne mycotoxin, induces oxidative stress and triggers ferroptosis-related injury. Lycopene (LYC), a natural antioxidant carotenoid, has not been well characterized for its protective role against ZEA-induced uterine damage in pregnant sows. This study investigated the protective efficacy as well as the underlying molecular mechanisms of LYC against ZEA-induced uterine injury. Thirty-two pregnant sows were randomly assigned to groups and fed basal diets supplemented with ZEA (1 mg/kg) and/or LYC (400 mg/kg) on gestational days 1-35. Porcine endometrial epithelial cell

AHL13, AHL19 and AHL20 Genes Differentially Regulate Cell Wall-Associated Defense Responses under Fumonisin B1-Induced Stress in Arabidopsis thaliana.

Fumonisin B1 (FB1), a mycotoxin produced by Fusarium species, induces oxidative stress and cell wall-associated defense responses in plants. However, the regulatory factors that determine how different components of cell wall defense are coordinated under toxin-induced stress remain poorly understood. AT-hook motif nuclear-localized (AHL) proteins are chromatin-associated transcriptional regulators, but their roles in FB1-induced cell wall remodeling have not been directly examined. In this study, we investigated the distinct roles of AHL13, AHL19, and AHL20 genes in FB1-induced defense respon

Activation of the unfolded protein response via PERK-ATF4 signalling in deoxynivalenol- and PRRSV-co-exposed MARC-145 cells.

IMPORTANCE: Deoxynivalenol (DON) is a prevalent mycotoxin that is highly toxic to animals, particularly pigs. Porcine reproductive and respiratory syndrome virus (PRRSV) is a major swine pathogen with high infectivity and severe clinical outcomes, but the mechanisms underlying the host cell responses to co-exposure of DON and PRRSV remain unclear. OBJECTIVE: This study examined the effects of the co-exposure of Marc-145 cells with DON and PRRSV. METHODS: A DON- and PRRSV-co-exposed MARC-145 cells model was established, and transcriptomic/proteomic sequencing combined with Western blot and im

Influence of interstitial carbon dioxide on deoxynivalenol accumulation in stored wheat under varying Fusarium treatments and airtight storage conditions.

BACKGROUND: Temperature and moisture content are recognized as the main factors responsible for mycotoxin production in stored grain, while the role of gaseous environment, particularly interstitial CO2, remains unexplored. This study investigated the relationship between interstitial CO2 concentration (generated metabolically in air spaces between grain kernels) and deoxynivalenol (DON) production in stored wheat under varying moisture contents (13%, 15%, 17%, 20%, or 23%), Fusarium treatments (naturally infected, Fusarium-inoculated, disinfected, or disinfected + Fusarium-inoculated), and ai

Herbicolin A, an antifungal lipopeptide produced by Pantoea agglomerans APC 4211 is a promising biocontrol agent against food spoilage fungi.

Fungal contamination of food with yeast and molds is associated with major economic losses due to spoilage and also poses health risks in the form of mycotoxin production. The strain Pantoea agglomerans APC 4211 isolated from leaves of Ilex aquifolium (holly tree) has broad spectrum antifungal activity against a variety of food spoilage fungi. Genomic analysis of the strain confirmed the presence of biosynthetic gene clusters potentially encoding for the enzymatic machinery required for the production of the antifungal lipopeptide herbicolin A. Antifungal activity and biochemical characteristics of natamycin against Fusarium graminearum.

Fusarium head blight (FHB) caused by Fusarium graminearum is a destructive fungal disease of wheat. Natamycin is a natural polyene macrolide antifungal compound exhibiting broad-spectrum antifungal activity, but its efficacy and mechanism against F. graminearum remain unclear. In this study, the sensitivity of 47 F. graminearum strains from different regions of China to natamycin was determined. The results showed that natamycin had a good inhibitory effect with the EC50 value ranging from 0.93 to 4.04 μg/mL and a mean EC50 value of 2.20 ± 0.59 μg/mL. No resistance association was found betwee
