Spatial immobility and the dynamics of loneliness, social capital, and mental health during the COVID-19 pandemic.

OBJECTIVES: COVID-19 lockdowns and wider mixing restrictions severely disrupted people's lives with potentially acute implications for their mental health. This study examines how mixing restrictions affected people's loneliness and how far loneliness, in turn, can explain any impact of restrictions on psychological distress. In addition, the study explores whether local social capital (LSC) in residential communities buffered any impact of mixing restrictions on loneliness, and subsequently protected people's mental health, during the pandemic. STUDY DESIGN: Individuals are drawn from three

Simultaneous detection and characterization of common respiratory pathogens in wastewater through genomic sequencing.

Genomic surveillance of SARS-CoV-2 has given insight into the evolution and epidemiology of the virus and its variant lineages during the COVID-19 pandemic. Expanding this approach to include a range of respiratory pathogens can better inform public health preparedness for potential outbreaks and epidemics. Here, we simultaneously sequenced 38 pathogens including influenza viruses, coronaviruses and bocaviruses, to examine the abundance and seasonality of respiratory pathogens in urban wastewater. We deployed a targeted bait capture method and short-read sequencing (Illumina Respiratory Virus

Recently emerged swine influenza A virus (H2N3) causes severe pneumonia in Cynomolgus macaques.

The triple reassortant H2N3 virus isolated from diseased pigs in the United States in 2006 is pathogenic for certain mammals without prior adaptation and transmits among swine and ferrets. Adaptation, in the H2 hemagglutinin derived from an avian virus, includes the ability to bind to the mammalian receptor, a significant prerequisite for infection of mammals, in particular humans, which poses a big concern for public health. Here we investigated the pathogenic potential of swine H2N3 in Cynomolgus macaques, a surrogate model for human influenza infection. In contrast to human H2N2 virus, whic

Improving risk analysis of environmentally driven zoonotic biological threats as a primary pandemic prevention approach: A case study of the Tripartite Joint Risk Assessment Operational Tool operationalization in Kenya.

Kenya faces a heightened risk of emerging and/or reemerging infectious disease (EID) outbreaks of zoonotic origins due to climate-induced extreme weather events and other environmental drivers. We aimed to obtain the perspectives of stakeholders on the development of a newly conceptualized integrated risk analysis (IRA) framework to address environmentally driven EIDs. We addressed the study aim by learning from stakeholders' experiences with the operationalization of the Joint Risk Assessment Operational Tool (JRA OT) in Kenya. We conducted an exploratory and explanatory qualitative study usi

Orally bioavailable RORγ/DHODH dual host-targeting small molecules with broad-spectrum antiviral activity.

Host-directed antivirals (HDAs) represent an attractive treatment option and a strategy for pandemic preparedness, especially due to their potential broad-spectrum antiviral activity and high barrier to resistance development. Particularly, dual-targeting HDAs offer a promising approach for antiviral therapy by simultaneously disrupting multiple pathways essential for viral replication. Izumerogant (IMU-935) targets two host proteins, (i) the retinoic acid receptor-related orphan receptor γ isoform 1 (RORγ1), which modulates cellular cholesterol metabolism, and (ii) the enzyme dihydroorotate d…
