Beyond text: Using network centralities and AI models to detect suicide risk on Reddit.

Suicide prevention research increasingly leverages social media data, where individuals often share personal experiences and emotions. While most studies rely on textual analysis to detect suicidal ideation, the structural characteristics of online social networks remain underexplored. In this exploratory pilot-scale study, we investigate whether network centrality metrics derived from Reddit interaction graphs can provide complementary signals for suicide risk detection beyond text-centric approaches. We construct directed user interaction graphs and extract 14 centrality metrics, including d

Ensemble learning-guided discovery of anti-tuberculosis phytochemicals: computational prediction and mechanistic insights.

Tuberculosis (TB) remains a major global health challenge driven by persistent Mycobacterium tuberculosis infection and increasing drug resistance. Phytochemicals represent a structurally diverse and underexplored chemical space for anti-TB drug discovery, yet systematic prioritization strategies integrating machine learning and structure-based validation are limited. A curated phenotypic anti-TB dataset of 425,180 compounds was used to train ensemble ExtraTrees models based on ECFP4 fingerprints and physicochemical descriptors. The models achieved strong predictive performance (ROC-AUC up to

CD44 and CLDN3 as immune-metabolic regulators in acute pancreatitis: a multi-modal transcriptomics study and experimental validation.

Acute pancreatitis (AP) is an inflammatory disorder of exocrine pancreas regulated by a complex interaction between injured pancreatic acinar cells and immune cells. Recent studies indicated the crucial role of glycolysis in regulating immune cell function and inflammation. Here, we identified 43 glycolysis-related differentially expressed genes (DEGs) from transcriptomic datasets (GSE65146 and GSE109227). Through three machine learning algorithms,Claudin-3 (CLDN3) and CD44 were identified as key glycolysis-related DEGs. Their significant upregulation was further validated in an independent da

Analysis of immunoinfiltration and EndoMT based on TGF-β signaling pathway-related genes in acute myocardial infarction.

Acute myocardial infarction (AMI), a critical manifestation of coronary heart disease, presents a complex and not entirely understood etiology. This study investigates the potential role of immune infiltration and endothelial-mesenchymal transition (EndoMT) in AMI pathogenesis. We conducted an analysis of the GSE24519 and MSigDB datasets to identify differentially expressed genes associated with the TGF-β signaling pathway (DE-TSRGs) and carried out a functional enrichment analysis. Additionally, we evaluated immune infiltration in AMI and its possible link to myocardial fibrosis. Key genes we

Pattern and predictors of death from aluminum and zinc phosphide poisoning using multi-kernel optimized relevance vector machine.

The use of metal phosphides, particularly aluminum phosphide, poses a significant threat to human safety and results in high mortality rates. This study aimed to determine mortality patterns and predictive factors for acute zinc and aluminum phosphide poisoning cases that were admitted to Menoufia University Poison and Dependence Control Center from 2017 to 2021. Statistical analysis revealed that poisoning was more common among females (59.7%), aged between 10 and 20 years, and from rural regions. Most cases were students, and most poisonings were the result of suicidal intentions (78.6%). A

Understanding the mutational frequency in SARS-CoV-2 proteome using structural features.

The prolonged transmission of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus in the human population has led to demographic divergence and the emergence of several location-specific clusters of viral strains. Although the effect of mutation(s) on severity and survival of the virus is still unclear, it is evident that certain sites in the viral proteome are more/less prone to mutations. In fact, millions of SARS-CoV-2 sequences collected all over the world have provided us a unique opportunity to understand viral protein mutations and develop novel computational approach

Automatic Lacunae Localization in Placental Ultrasound Images via Layer Aggregation.

Accurate localization of structural abnormalities is a precursor for image-based prenatal assessment of adverse conditions. For clinical screening and diagnosis of abnormally invasive placenta (AIP), a life-threatening obstetric condition, qualitative and quantitative analysis of ultrasonic patterns correlated to placental lesions such as placental lacunae (PL) is challenging and time-consuming to perform even for experienced sonographers. There is a need for automated placental lesion localization that does not rely on expensive human annotations such as detailed manual segmentation of anatom

Ultrasound-Based AI in Predicting Hormone Receptor Status in Breast Cancer: Is "Digital Biopsy" Possible.

Accurate and rapid determination of tumor histopathological features and molecular subtypes is critical for breast cancer prognosis and treatment strategies. This study evaluates the feasibility of a "digital biopsy" approach that uses machine learning models to predict hormone receptor status from ultrasound radiomic features non-invasively. A retrospective analysis was conducted on 353 breast tumors from 311 patients who were evaluated between February 2019 and December 2024. Radiomic features were extracted from two-dimensional segmentations of pre-biopsy ultrasound images. The dataset was

Integrating machine learning and metabolomics to identify PFAS-associated metabolic alterations related to lung cancer risk.

Per- and polyfluoroalkyl substances (PFAS) are pervasive environmental contaminants increasingly implicated in lung cancer risk. However, the metabolic features and biological pathways associated with human PFAS exposure in relation to lung cancer remain poorly characterized. We conducted a nested case-control study within the Quzhou Environment Exposure and Human Health cohort (408 lung cancer cases and 421 controls) to examine whether serum PFAS concentrations are associated with lung cancer risk and to identify metabolic alterations associated with both PFAS exposure and lung cancer. Seven

Dynamic albumin-to-RDW trajectories for time-dependent risk stratification in acute brain injury.

BACKGROUND: Acute brain injury (ABI) requires early, interpretable risk stratification to support ICU risk assessment. We evaluated whether longitudinal albumin-to-red cell distribution width ratio (ARR) trajectories provide time-dependent prognostic information and support prediction with multicenter validation. METHODS: In this multicenter, retrospective cohort study (NSICU, MIMIC-IV, eICU), latent class growth modeling identified ARR trajectories. Nonproportional hazards were addressed with time-stratified Cox models (0-7, 7-14, >14 days) and restricted mean survival time (RMST) at 7, 14,

MRI-based risk factors for intensive care unit admissions in acute neck infections.

OBJECTIVES: We assessed risk factors and developed a score to predict intensive care unit (ICU) admissions using MRI findings and clinical data in acute neck infections. METHODS: This retrospective study included patients with MRI-confirmed acute neck infection. Abscess diameters were measured on post-gadolinium T1-weighted Dixon MRI, and specific edema patterns, retropharyngeal (RPE) and mediastinal edema, were assessed on fat-suppressed T2-weighted Dixon MRI. A multivariate logistic regression model identified ICU admission predictors, with risk scores derived from regression coefficients.

A machine learning based framework for identifying consumer product injuries from social media data.

BACKGROUND: Safety is a critical aspect of consumer products. However, there are millions of product related injuries reported each year. Traditional injury surveillance efforts led by public health agencies involve product related injury data collection from hospitals and subsequent injury causation analysis. This approach often requires long processing time and leads to delays in identifying emergent consumer product-related injury patterns and preventive intervention steps such as product recalls, which causes continued product related injuries. METHODS: We propose a machine learning (ML)

Sex dimorphism of IL-17-secreting peripheral blood mononuclear cells in ankylosing spondylitis based on bioinformatics analysis and machine learning.

BACKGROUND: Ankylosing spondylitis (AS) with radiographic damage is more prevalent in men than in women. IL-17, which is mainly secreted from peripheral blood mononuclear cells (PBMCs), plays an important role in the development of AS. Its expression is different between male and female. However, it is still unclear whether sex dimorphism of IL-17 contribute to sex differences in AS. METHODS: GSE221786, GSE73754, GSE25101, GSE181364 and GSE205812 datasets were collected from the Gene Expression Omnibus (GEO) database.
