Lomerizine Prophylaxis for Pediatric Migraine: Efficacy and Predictors of Treatment Response-A Retrospective Cohort Study.

Background/Objectives: There is limited evidence supporting the use of lomerizine, a calcium channel blocker approved for migraine prophylaxis in Japan, in children. This study evaluated the prophylactic efficacy of lomerizine in a pediatric population with migraine and identified the baseline factors independently associated with treatment response. Methods: This single-center, retrospective cohort study enrolled 83 patients aged ≤18 years prescribed lomerizine for migraine prophylaxis. The primary outcome was headache improvement, defined as a ≥50% reduction in monthly headache days from bas

Efficacy of dihydroartemisinin-piperaquine to prevent malaria and complication-related malaria during pregnancy compared to sulfadoxine-pyrimethamine: a systematic review, meta-analysis, and meta-regression.

This study aimed to evaluate the efficacy of DHP as an alternative to SP for the prevention of malaria and malaria-related complications during pregnancy. A systematic review and meta-analysis were conducted using pooled effect sizes extracted from published studies. Compared to SP, DHP significantly reduced maternal malaria infection by 78% (RR 0.22, 95% CI: 0.09-0.55), placental malaria infection by 73% (RR 0.27, 95% CI: 0.08-0.98), and maternal anaemia by 16% (RR = 0.84, 95% CI: 0.79-0.89). Women randomized to the DHP arm had a 28% lower risk of foetal loss (RR 0.72, 95% CI: 0.40-1.31), a 4

[OLAPARIB TREATMENT IN METASTATIC CASTRATION-RESISTANT PROSTATE CANCER WITH BRCA2 MUTATION AND HIGH TUMOR MUTATIONAL BURDEN: A CASE REPORT].

In recent years, pharmacologic treatment for advanced prostate cancer has included novel hormonal agents and chemotherapeutic drugs from the initial stages. When resistance develops, personalized therapy based on genomic profiling has become increasingly common. We report a case of castration-resistant prostate cancer (CRPC) with both a BRCA2 mutation and high tumor mutational burden (TMB-high), in which olaparib produced a temporary response. The patient had received hormone therapy, radiotherapy, and chemotherapy, all of which became ineffective. Based on companion diagnostic testing, both o

Development of MDM2-Targeting PROTAC for Advancing Bone Regeneration.

Proteolysis-targeting chimeras (PROTACs) degrade target proteins through the ubiquitin-proteasome system. To date, PROTACs are primarily used to treat various diseases; however, they have not been applied in regenerative therapy. Herein, this work introduces MDM2-targeting PROTACs customized for application in bone regeneration. An MDM2-PROTAC library is constructed by combining Nutlin-3 and CRBN ligands with various linker designs. Through a multistep validation process, this work develops MDM2-PROTACs (CL144 and CL174) that presented potent degradation efficiency and a robust inductive effec

Lrp Family Regulator SCAB_Lrp2 Responds to the Precursor Tryptophan and Represses the Thaxtomin Biosynthesis in Streptomyces scabies.

Streptomyces scabies is a well-researched plant pathogen belonging to the genus Streptomyces. Its virulence is linked to the production of the secondary metabolite thaxtomin A, which is tightly regulated at the transcriptional level. The leucine-responsive regulatory protein (Lrp) family is conserved in prokaryotes and is involved in various crucial biological processes. However, the regulatory interaction between Lrp protein and pathogenic Streptomyces species remains poorly understood. This study aims to explore the role of SCAB_Lrp2 in regulating thaxtomin biosynthesis and pathogenicity, an

Portable Miniaturized IoT-Enabled Point-of-Care Device for Electrochemical Sensing of Zopiclone in Cocktails.

This study proposes a portable and IoT-based electrochemical point-of-care sensing device for detecting zopiclone in cocktails. The system utilizes an electrochemical laccase biosensor and a potentiostat, offering a low-cost and portable device for detecting this sedative drug in cocktails. The sensor characterization experiments demonstrated the linear behavior of the oxidation and reduction currents for each of the targeted concentrations of zopiclone, enabling their detection and quantification even when mixed with an interfering substance. The proposed system could be used for the in situ

Chemical Genetics in C. elegans Identifies Anticancer Mycotoxins Chaetocin and Chetomin as Potent Inducers of a Nuclear Metal Homeostasis Response.

C. elegans numr-1/2 (nuclear-localized metal-responsive) is an identical gene pair encoding a nuclear protein previously shown to be activated by cadmium and disruption of the integrator RNA metabolism complex. We took a chemical genetic approach to further characterize regulation of this novel metal response by screening 41,716 compounds and extracts for numr-1p::GFP activation. The most potent activator was chaetocin, a fungal 3,6-epidithiodiketopiperazine (ETP) with promising anticancer activity. Chaetocin activates numr-1/2 strongly in the alimentary canal but is distinct from metal exposu

The host-directed therapeutic imatinib mesylate accelerates immune responses to Mycobacterium marinum infection and limits pathology associated with granulomas.

Infections caused by members of the mycobacterium tuberculosis complex [MTC] and nontuberculous mycobacteria [NTM] can induce widespread morbidity and mortality in people. Mycobacterial infections cause both a delayed immune response, which limits rate of bacterial clearance, and formation of granulomas, which contain bacterial spread, but also contribute to lung damage, fibrosis, and morbidity. Granulomas also limit access of antibiotics to bacteria, which may facilitate development of resistance. Bacteria resistant to some or all antibiotics cause significant morbidity and mortality, and new

The antipsychotic medications aripiprazole, brexpiprazole and cariprazine are off-target respiratory chain complex I inhibitors.

Antipsychotic drugs are the mainstay of treatment for schizophrenia and provide adjunct therapies for other prevalent psychiatric conditions, including bipolar disorder and major depressive disorder. However, they also induce debilitating extrapyramidal syndromes (EPS), such as Parkinsonism, in a significant minority of patients. The majority of antipsychotic drugs function as dopamine receptor antagonists in the brain while the most recent 'third'-generation, such as aripiprazole, act as partial agonists. Despite showing good clinical efficacy, these newer agents are still associated with EPS

Targeting BRPF3 moderately reverses olaparib resistance in high grade serous ovarian carcinoma.

PARP inhibitors (PARPi) kill cancer cells by stalling DNA replication and preventing DNA repair, resulting in a critical accumulation of DNA damage. Resistance to PARPi is a growing clinical problem in the treatment of high grade serous ovarian carcinoma (HGSOC). Acetylation of histone H3 lysine 14 (H3K14ac) and associated histone acetyltransferases (HATs) and epigenetic readers have known functions in DNA repair and replication. Our objectives are to examine their expression and activities in the context of PARPi-resistant HGSOC, and to determine if targeting H3K14ac or associated proteins ha
