AI in Drug Discovery: Clinical Failures, Regulatory Reality, and the Validation Crisis Behind the Hype.

The integration of artificial intelligence (AI) into the life sciences has accelerated significantly between 2022 and 2026, accompanied by global investment exceeding USD 100 billion and widespread expectations of a transformative impact in drug discovery. Despite these advances, the extent to which AI has improved clinical outcomes remains unclear. This study presents a structured narrative review evaluating the economic, technical, clinical, and regulatory dimensions of AI adoption in drug discovery. Current evidence indicates that clinical attrition rates remain high, with approximately 90%

Diagnostic Potential of a Recombinant Candida albicans Hyr1 Protein.

Invasive candidiasis (IC) is a life-threatening fungal infection caused by Candida species. Current diagnostic methods are based on blood culture of the fungus, a technique with limited sensitivity and slow turnaround times. To address these limitations, novel diagnostic strategies are under investigation. This study evaluates the diagnostic potential of the Candida albicans germ tube protein Hyr1 and a subterminal Hyr1 fragment (D22b), both produced in an eukaryotic expression system, for the diagnosis of IC; for that purpose, recombinant Hyr1 and D22b were expressed in Pichia pastoris and te

Lysine acetylation modulates s-OPA1 GTPase activity and oligomerization in mitochondrial membrane remodeling.

Mitochondrial dynamics are regulated by coordinated fission and fusion events that rely on key proteins and lipids organized spatially within the mitochondria. The dynamin-related GTPase Optic Atrophy 1 (OPA1) is essential for inner mitochondrial membrane fusion and cristae structure maintenance. While post-translational modifications, particularly lysine acetylation, are emerging as critical regulators of mitochondrial protein function, their impact on OPA1 remains poorly characterized. In this study, we explored the effects of lysine acetylation on the short form of OPA1 (s-OPA1) using acety

Mitochondrial tRNA pseudouridylation governs erythropoiesis.

Pseudouridine is the most prevalent RNA modification, and its aberrant function is implicated in various human diseases. However, the specific impact of pseudouridylation on hematopoiesis remains poorly understood. Here, we investigated the role of transfer RNA (tRNA) pseudouridylation in erythropoiesis and its association with mitochondrial myopathy, lactic acidosis, and sideroblastic anemia syndrome (MLASA) pathogenesis. By using patient-specific induced pluripotent stem cells (iPSCs) carrying a genetic pseudouridine synthase 1 (PUS1) mutation and a corresponding mutant mouse model, we demon

The Molecular Basis of FIX Deficiency in Hemophilia B.

Coagulation factor IX (FIX) is a vitamin K dependent protein and its deficiency causes hemophilia B, an X-linked recessive bleeding disorder. More than 1000 mutations in the F9 gene have been identified in hemophilia B patients. Here, we systematically summarize the structural and functional characteristics of FIX and the pathogenic mechanisms of the mutations that have been identified to date. The mechanisms of FIX deficiency are diverse in these mutations. Deletions, insertions, duplications, and indels generally lead to severe hemophilia B. Those in the exon regions generate either frame sh

Nrf2/Keap1 Signaling Axis in the Brain: Master Regulator of Oxidative Stress in Neurodegenerative and Psychiatric Disorders.

Oxidative stress is a crucial factor in the development of CNS disorders, including neurodegenerative and psychiatric conditions. The Nrf2/Keap1 signaling axis plays a central role in defending against oxidative damage by regulating antioxidant and cytoprotective gene expression. Beyond its antioxidant function, Nrf2 influences neurogenesis, synaptic plasticity, mitochondrial bioenergetics, and glial neuronal interactions, all of which are vital for maintaining neural integrity and cognitive performance. Dysregulation of this pathway through altered dimerization, post-translational modificatio

The Interplay Between Antioxidant and Chaperone Functions of α-Crystallin.

α-Crystallin, the predominant protein of the eye lens, possesses molecular chaperone activity and antioxidative properties, both of which are essential for maintaining lens transparency. Its chaperone function prevents the formation of light-scattering protein aggregates, while its antioxidative activity mitigates oxidative stress through both direct and indirect mechanisms. However, with aging, α-crystallin undergoes cumulative post-translational modifications and oxidative damage, leading to protein crosslinking and a decline in chaperone efficacy. Notably, α-crystallin exhibits free radical

[Molecular bases of prion diseases].

Prion diseases are transmissible neurodegenerative conditions that include Creutzfeldt-Jakob disease (CJD) in humans and bovine spongiform encephalopathy and scrapie in animals. The normal cellular prion protein (PrP(c)) is a membrane sialoglycoprotein of unknown function having the unique property of adopting an abnormal tertiary conformation. The pathological conformer (PrP(sc)) would be the agent of transmissible spongiform encephalopathies or prion diseases. Prion propagation involves recruitment of host cellular prion protein, primarily of alpha-helical structure, into a disease-specific

Lipocalins of boar salivary glands binding odours and pheromones.

Large amounts of an odorant-binding protein have been isolated from submaxillary glands of mature male pig. This polypeptide molecule is sex-specific, being absent in females. On electrophoretic gels under denaturing conditions it migrated as a broad band with an apparent molecular mass of around 20 kDa. Electrospray mass spectrometry revealed the presence of three main components, whose mass differences are not interpretable as result of any common post-translational modifications, indicating the presence of distinct polypeptide chains. N-terminal Edman degradation yielded a single sequence o

Identification of differential protein recognition pattern between Naegleria fowleri and Naegleria lovaniensis.

Many pathogenicity factors are involved in the development of primary amoebic meningoencephalitis (PAM) caused by N fowleri. However, most of them are not exclusive for N fowleri and they have not even been described in other nonpathogenic Naegleria species. Therefore, the objective of this work was to identify differential proteins and protein pattern recognition between Naegleria fowleri and Naegleria lovaniensis using antibodies anti-N fowleri as strategy to find vaccine candidates against meningoencephalitis. Electrophoresis and Western blots conventional and 2-DE were performed for the id

Label-Free Quantitative Proteomic Analysis of Nitrogen Starvation in Arabidopsis Root Reveals New Aspects of H2S Signaling by Protein Persulfidation.

Hydrogen sulfide (H2S)-mediated signaling pathways regulate many physiological and pathophysiological processes in mammalian and plant systems. The molecular mechanism by which hydrogen sulfide exerts its action involves the posttranslational modification of cysteine residues to form a persulfidated thiol motif. We developed a comparative and label-free quantitative proteomic analysis approach for the detection of endogenous persulfidated proteins in N-starved Arabidopsis thaliana roots by using the tag-switch method. In this work, we identified 5214 unique proteins from root tissue that were

Met and the microenvironment: new insights for ovarian cancer metastasis.

BACKGROUND: Thoracic aortic aneurysm and dissection (TAAD) are devastating cardiovascular diseases with a high rate of disability and mortality. Lysine succinylation, a newly found post-translational modification, has been reported to play an important role in cardiovascular diseases. However, how succinylation modification influences TAAD remains obscure. METHODS: Ascending aortic tissues were obtained from patients with thoracic aortic aneurysm (TAA, n = 6), thoracic aortic dissection (TAD) with pre-existing aortic aneurysm (n = 6), and healthy subjects (n = 6). Global lysine succinylation

Aptamer-Based Targeted Protein Degradation: Mechanistic Principles, Delivery Strategies, and Translational Opportunities.

Targeted elimination of pathogenic proteins remains a fundamental challenge in therapeutic intervention, particularly for proteins considered "undruggable" by conventional inhibition-based approaches. Aptamer-based targeted protein degradation (TPD) has emerged as a versatile strategy that leverages the high specificity, structural programmability, and chemical tunability of nucleic acid aptamers to redirect disease-relevant proteins toward endogenous degradation pathways. Intracellular platforms, exemplified by aptamer-conjugated proteolysis-targeting chimeras (PROTACs), induce ubiquitination

Sleep-Gated Adaptation: A Conceptual Framework Linking Exercise-Induced Adaptive Consolidation to Interindividual Variability in Training Response.

Exercise-induced molecular signaling is transient, resolving within hours, whereas the structural and functional adaptations to repeated training develop over weeks to months. The processes that convert signaling into durable adaptation, the conditions that best support this conversion, and the sources of interindividual variability in training outcomes have largely been studied separately. This review proposes a conceptual framework for integrating these approaches. Adaptive Consolidation is introduced as an organizing concept for this intermediate transition, synthesized as a multi-filter ca

Lysine acetylation-mediated regulation of ferredoxin and ferredoxin reductase redox-active proteins in Haloferax volcanii.

Lysine acetylation is an evolutionarily conserved, post-translational modification that regulates metabolism and protein function, yet its role in archaeal electron transfer systems remains poorly understood. Here, we investigated lysine acetylation of the 2Fe-2S ferredoxin Hv Fdx (HVO_2995) and its flavin-dependent oxidoreductase Hv FdR (HVO_2345) partner in the halophilic archaeon Haloferax volcanii . Genetic and biochemical analyses established Hv Fdx as an essential 2Fe-2S ferredoxin with a midpoint redox potential of -385 mV. Lysine acetylation of Hv Fdx was found to occur primarily at K1

ALG1-mediated PDL1 glycosylation drives macrophage m2 polarization to promote bladder cancer progression.

Asparagine-linked glycosylation protein 1 homolog (ALG1), a β-1,4-mannosyltransferase, catalyzes the initial committed step of the N-glycosylation pathway. While its role in congenital glycosylation disorders is well established, the contribution of ALG1 to bladder cancer (BC) progression, particularly its impact on the tumor immune microenvironment, remains poorly understood. In this multi-omics study, we delineated the pathological significance and molecular mechanisms of ALG1 in BC. We demonstrated that ALG1 directly interacted with programmed death-ligand 1 (PDL1) and catalyzed its glycosy

SIRT7-mediated desuccinylation of FOXO4 suppresses ferroptosis to alleviate LPS-induced acute lung injury.

Protein succinylation, an emerging post-translational modification (PTM), assumes a crucial role in the initiation and advancement of inflammatory diseases. Ferroptosis, propelled by lethal lipid peroxidation, is intricately associated with the pathogenesis of inflammation. Targeting ferroptosis has recently emerged as a promising therapeutic approach for acute lung injury (ALI). Nevertheless, the crosstalk between protein succinylation and ferroptosis in the regulation of ALI remains ambiguous. FOXO4, a key regulator of oxidative stress responses, is dynamically regulated by various PTMs. We

Molecular mechanisms and regulatory networks of salt stress tolerance in plants.

Soil salinity is a major threat to crop productivity, sustainable agriculture, and global food security, with more than 833 million hectares of land affected worldwide. Salt stress restricts plant growth through osmotic stress, ion toxicity, oxidative damage, membrane disruption, reduced photosynthesis, and yield loss. Plants respond through coordinated regulatory networks that connect early stress perception with ion balance, osmotic adjustment, hormone signaling, transcript regulation, and protein modification. Recent advances have identified several sensory and signaling modules involved in

SGIV and RGNNV regulate fish innate immune responses by mediating ubiquitinated modifications of proteasome and ribosomal proteins.

Singapore grouper iridovirus (SGIV) and red-spotted grouper nervous necrosis virus (RGNNV) have been identified as the predominant DNA and RNA viral pathogens, respectively, affecting grouper populations. Protein ubiquitination, a crucial post-translational modification, plays pivotal regulatory roles in both host innate immune responses and viral replication processes. In the present study, we employed quantitative proteomics approaches to systematically identify and characterize alterations in ubiquitination-modified proteins in grouper spleen (Epinephelus akaara, GS) cells following SGIV an
