Manufacturing, characterization, and degradation of a poly(lactic acid) warp-knitted spacer fabric scaffold as a candidate for tissue engineering applications.

Three-dimensional bioabsorbable textiles represent a novel technology for the manufacturing of tissue engineering scaffolds. In the present study, 3D bioabsorbable poly(lactic acid) (PLA) spacer fabric scaffolds are fabricated by warp-knitting and their potential for tissue engineering is explored in vitro. Changes in physical properties and mechanical performance with different heat setting treatments are assessed. To characterize the microenvironment experienced by cells in the scaffolds, yarn properties are investigated prior to, and during, hydrolytic degradation. The differences in yarn m…

Esophagus stretch tests: Biomechanics for tissue engineering and possible implications on the outcome of esophageal atresia repairs performed under excessive tension.

BACKGROUND: Esophageal biomechanical studies are important to understand structural changes resulting from stretches during repair of esophageal atresias as well as to obtain values to compare with the biomechanics of tissue-engineered esophagus in the future. This study aimed to investigate light microscopic changes after uniaxial stretching of the ovine esophagus. METHODS: In vitro uniaxial stretching was performed on esophagi (n = 20) of 1-month-old lambs within 4-6 h post-mortem. Esophagi were divided into 5 groups: control and stretched (1.1, 1.2, 1.3 and 1.4). Force and lengthening were

Cultivation of auricular chondrocytes in poly(ethylene glycol)/poly(ε-caprolactone) hydrogel for tracheal cartilage tissue engineering in a rabbit model.

Tissue engineering has the potential to overcome the limitations of tracheal reconstruction. To tissue-engineer a tracheal cartilage, auricular chondrocytes were encapsulated in a photocurable poly(ethylene glycol)/poly(ε-caprolactone) (PEG/PCL) hydrogel. Chondrogenic genes, including Sox9, Acan and Col2a1, were up-regulated in auricular chondrocytes after 2 weeks of in vitro cultivation in the PEG/PCL hydrogel. Co-cultivation of 70 % auricular chondrocytes and 30 % bone marrow mesenchymal stem cells (BMSCs) accelerated the chondrogenic genes' expression in the PEG/PCL hydrogel. Cartilaginous …

Orthopaedic regenerative tissue engineering en route to the holy grail: disequilibrium between the demand and the supply in the operating room.

Orthopaedic disorders are very frequent, globally found and often partially unresolved despite the substantial advances in science and medicine. Their surgical intervention is multifarious and the most favourable treatment is chosen by the orthopaedic surgeon on a case-by-case basis depending on a number of factors related with the patient and the lesion. Numerous regenerative tissue engineering strategies have been developed and studied extensively in laboratory through in vitro experiments and preclinical in vivo trials with various established animal models, while a small proportion of them…

One-step method for the preparation of poly(methyl methacrylate) modified titanium-bioactive glass three-dimensional scaffolds for bone tissue engineering.

A novel three-dimensional (3D) titanium (Ti)-doping meso-macroporous bioactive glasses (BGs)/poly(methyl methacrylate) (PMMA) composite was synthesised using PMMA and EO20PO70EO20 (P123) as the macroporous and mesoporous templates, respectively. Unlike the usual calcination method, the acid steam technique was used to improve the polycondensation of Ti-BGs, and then PMMA was partially extracted via chloroform to induce the macroporous structure. Simultaneously, the residual PMMA which remained in the wall enhanced the compressive strength to 2.4 MPa (0.3 MPa for pure BGs). It is a simple and g

Comparative study of different seeding methods based on a multilayer SIS scaffold: Which is the optimal procedure for urethral tissue engineering?

Seeding cells efficiently and uniformly onto three-dimensional scaffolds is key for engineering urological tissue with an ideal histological structure in vitro. Using an optimized seeding technology allows cells to cooperate positively with biomaterials, resulting in successful reconstructive surgery. In this study, we used four different types of seeding methods in a scaffold of small intestinal submucosa (SIS). The efficiency of the sandwich co-culture, layered co-culture, static-agitation seeding, and centrifugation seeding methods were compared. It was demonstrated that dynamic seeding met

Phenotypic fitness of primary endothelial cells cultured from patients with high cardiovascular risk or chronic kidney disease for vascular tissue engineering.

Vascular tissue engineering relies on the combination of patient-derived cells and biomaterials to create new vessels. For clinical application, data regarding the function and behavior of patient-derived cells are needed. We investigated cell growth and functional characteristics of human venous endothelial cells (HVECs) from coronary arterial bypass graft (CABG), chronic kidney disease (CKD), and control patients. HVECs were isolated from venous specimens that were obtained during elective surgical procedures by means of collagenase digestion. Gene expression, proliferation, migration, secre…

Fabrication of electrospun thermoplastic polyurethane blended poly (l-lactide-co-e-caprolactone) microyarn scaffolds for engineering of female pelvic-floor tissue.

Potential scaffolds for repair of the female pelvic floor require new materials and fabrication by novel methods to improve cellular infiltration. An 'ideal' engineered scaffold for pelvic-floor tissue should mimic the three-dimensional (3D) network of the extracellular matrix (ECM), which possesses intricate macro- and nano-architecture. In this study, a series of blended poly(l-lactide-co-ecaprolactone) P(LLA-CL)/thermoplastic polyurethane (TPU) microyarn/microfibrous scaffolds were produced with different weight ratios via dynamic liquid electrospinning and electrospinning. Both biopolymers

Evaluation of the Clinical Usefulness of a Bioartificial Urethral Substitute Generated by Tissue Engineering in Laboratory Rabbits: Protocol for an In Vivo Study.

BACKGROUND: Surgical management of diseases compromising the integrity of the human urethra remains a major clinical challenge, and congenital conditions, such as hypospadias and epispadias, and acquired diseases, such as trauma, tumors, and strictures, are difficult to treat, highlighting the need for novel regenerative approaches capable of restoring urethral architecture and function. Recent advances in tissue engineering have allowed the generation of bioartificial tissues, including several models of artificial urethra generated with different types of cells and biomaterials. Although pot…

Modular Tissue-Engineered Motor Units Featuring Spinal Motor Neurons Innervating Self-Assembled Myofiber Bundles.

A motor unit is the functional unit of muscle contraction, consisting of a population of skeletal muscle fibers innervated by axon terminals from a motor neuron. Tissue engineering strategies are being pursued to treat neuromuscular injuries by mimicking aspects of native myofascicular architecture; however, the critical role of innervation in myofiber development is often overlooked. Our group previously developed a pre-innervated tissue-engineered muscle on nanofiber sheets, demonstrating that innervation facilitated myofiber maturation and function in vitro. The current study builds on this…

Human iPSC-derived 3D cardiac models for cardiomyopathies: organoids, spheroids, and engineered heart tissues as translational platforms.

Cardiomyopathies represent a substantial global health burden, yet progress in developing effective therapies was long constrained by the absence of physiologically relevant human models to understand molecular mechanisms and evaluating interventions. The advent of induced pluripotent stem cell (iPSC) technology has driven major advances in cardiomyopathy research, although important limitations persist. Current iPSC-based systems exhibit incomplete cellular maturation, lack functional vasculature and chamber level architecture, and possess an immature extracellular matrix. These factors must

Optogenetic tachypacing facilitates burst-induced arrhythmia in stem cell-derived human atrial engineered heart tissue.

AIMS: Large animal models for atrial fibrillation (AF) research are ethically challenging and costly. Suitable humanized in vitro models could circumvent these problems. Here, we aimed to combine tissue engineering, human induced pluripotent stem cell-derived atrial cardiomyocytes (hiPSC-aCM), and optogenetic pacing to trigger an in vitro fibrillation-like state, which can be analysed by simple video recording. METHODS AND RESULTS: Atrial engineered heart tissue (aEHT) was created from hiPSC-aCM and transduced with adeno-associated virus vectors to express the channelrhodopsins CheRiff2.0, Ch…

Case Report: Lessons learned from large animal implantation of an all-natural tissue engineered vascular graft.

Cardiovascular disease continues to be the number one cause of morbidity and mortality across the world. Coronary artery bypass graft (CABG) procedures are the most commonly performed major surgery in the U.S. Grafts are difficult to source as patients do not have many sites from which to harvest donor tissues as autografts. Plastic grafts have issues of infection and are only used as a last resort. Tissue engineered vascular grafts have potential to solve the need for all-natural vascular grafts in the clinic. In this study, we evaluate the feasibility of a completely biological engineered va

Long-term safety and efficacy outcomes of the Acellular Tissue Engineered Vessel (ATEV) in extremity arterial trauma repair.

BACKGROUND: In cases of arterial injury, use of autologous vein for arterial repair is sometimes not feasible. We previously reported the short-term (ie, 30-day) outcomes of the Acellular Tissue Engineered Vessel (Symvess) in treating extremity vascular injuries. This study evaluates the long-term safety, efficacy, and mechanical durability of Symvess in patients with arterial trauma. METHODS: This prospective analysis focused on patients from the CLN-PRO-V005 clinical study who sustained noniatrogenic vascular injuries to the extremities, and for whom saphenous vein was not feasible for use.

Tissue engineering as a tool in a novel approach to the comprehensive treatment and management of a deeply and extensively burned patient: case report.

Deep and extensive thermal burns with concurrent inhalation injuries can be associated with a high mortality rate, especially among elderly patients. Injuries of this type require treatment in highly. specialized centers. Early excision and autografting are the standard of care for extensive, deep burns. To achieve a functionally and aesthetically satisfactory burn scar, allogeneic acellular dermal matrices (ADMs) can be used as co-grafts alongside autologous split-thickness skin grafts (STSGs). Additionally, the application of in vitro-cultured autologous keratinocytes and fibroblasts has bee

Dynamic molecular landscape in dorsal root ganglion for peripheral nerve regeneration promoted by tissue engineered nerve graft.

INTRODUCTION: The limited regenerative capacity of the nervous system represents a significant clinical challenge in the context of peripheral nerve injuries. An innovative strategy for sciatic nerve repair has been developed using tissue-engineered nerve grafts (TENGs) composed of skin-derived precursor Schwann-like cells (SKP-SCs) and a silk fibroin-chitosan scaffold. However, the reason why SKP-SCs-TENG demonstrated superior enhanced nerve regeneration compared to the autograft and scaffold groups remains unclear. OBJECTIVES: The present work aims to elucidate the superiority and molecular

Construction of tissue engineered cornea with skin-derived corneal endothelial-like cell and mechanism research for the cell differentiation.

INTRODUCTION: Corneal endothelial transplantation accounts for most of corneal transplantation for treating corneal diseases, however severe shortage of corneal donors is the biggest obstacle. In our previous study, we differentiated human skin-derived precursors (SKPs) into corneal endothelial cell (CEC)-like cells with a co-culture system. In this study, we aimed to investigate cell differentiation molecular mechanism and evaluate the function of CEC-like cells by developing tissue-engineered corneas in order to improve cell production efficiency and provide basic research for clinical trans

CO2 as an engine for neurofluid flow: Exploring the coupling between vascular reactivity, brain clearance, and changes in tissue properties.

The brain relies on an effective clearance mechanism to remove metabolic waste products for the maintenance of homeostasis. Recent studies have focused on elucidating the forces that drive the motion of cerebrospinal fluid (CSF), responsible for removal of these waste products. We demonstrate that vascular responses evoked using controlled manipulations of partial pressure of carbon dioxide (PaCO2) levels, serve as an endogenous driver of CSF clearance from the brain. To demonstrate this, we retrospectively surveyed our database, which consists of brain metastases patients from whom blood oxyg

A knock down strategy for rapid, generic, and versatile modelling of muscular dystrophies in 3D-tissue-engineered-skeletal muscle.

BACKGROUND: Human iPSC-derived 3D-tissue-engineered-skeletal muscles (3D-TESMs) offer advanced technology for disease modelling. However, due to the inherent genetic heterogeneity among human individuals, it is often difficult to distinguish disease-related readouts from random variability. The generation of genetically matched isogenic controls using gene editing can reduce variability, but the generation of isogenic hiPSC-derived 3D-TESMs can take up to 6 months, thereby reducing throughput. METHODS: Here, by combining 3D-TESM and shRNA technologies, we developed a disease modelling strateg
