BACKGROUND: Major depressive disorder (MDD) has a significant impact on global burden of disease. Complications in clinical management can occur when response to pharmacological modalities is considered inadequate and symptoms persist (treatment-resistant depression (TRD)). We aim to investigate inflammation, proxied by C-reactive protein (CRP) levels, and body mass index (BMI) as putative causal risk factors for depression and subsequent treatment resistance, leveraging genetic information to avoid confounding via Mendelian randomisation (MR).
METHODS: We used the European UK Biobank subcoho

BACKGROUND: Allergic conjunctivitis is an ocular immune disease which affects the conjunctiva, eyelids, and cornea. Growing evidence implicates the gut microbiota in balancing and modulating immunity response, and in the pathogenesis of allergic disease. As a result, gut microbial imbalance could be a useful indicator for allergic conjunctivitis. From the perspective of predictive, preventive, and personalized medicine (PPPM), clarifying the role of gut microbial imbalance in the development of allergic conjunctivitis could provide a window of opportunity for primary prediction, targeted preve

BACKGROUND: Observational studies have consistently reported associations between serum uric acid (SUA) and post-stroke epilepsy (PSE) in acute ischemic stroke (AIS), including a recently published large cohort study identifying a U-shaped relationship. However, whether these associations reflect causality and whether SUA provides incremental predictive value beyond established clinical factors remains unknown.
METHODS: we conducted a two-sample Mendelian randomization (MR) analysis to test whether lifelong genetically predicted serum urate shares a genetic causal architecture with general ep

A large number of studies have shown a close association between dietary habits and low hand grip strength. However, the presence of a causal connection between dietary habits and low hand grip strength remains unclear. All genome-wide association studies of dietary habits summary statistics were run in BOLT-LMM software (Broad Institute) using linear mixed models with 10 genetic principal components and genotyping array as covariates, and all phenotypes were averaged over multiple Food Frequency Questionnaires, adjusted for age and sex, and inverse normal transformed. INTRODUCTION: Inflammatory bowel disease (IBD), including Crohn's disease (CD) and ulcerative colitis (UC), imposes a global health burden. Observational studies suggest links between IBD and sarcopenia as well as obesity, but establishing causality is challenging due to confounding factors.
METHODS: This study utilized two-sample Mendelian randomization (MR) analyses to explore bidirectional causality between obesity, sarcopenia, and IBD, using genetic instruments from summary-level data. The primary causal estimates were derived using the inverse-variance weighted method. To ensure robustne

BACKGROUND: Alzheimer’s disease is a progressive neurodegenerative disorders with profound impacts on cognitive function. There is a shortage of economic evidence relating to the impact that Alzheimer’s disease has on healthcare costs and quality-adjusted life-years (QALYs). METHODS: We estimated conventional multivariable models of the association between Alzheimer’s disease and these core economic outcomes. However, these types of model may be confounded by omitted variables. We therefore also explored a causal approach using germline genetic variation as instrumental variables in a two-samp

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder with few therapeutic options. Observational data suggest that type 2 diabetes mellitus (T2DM) might protect against ALS, yet the mechanisms are unclear. Clarifying whether glucose or lipid metabolism underpins this protective effect could guide targeted interventions.
OBJECTIVE: This study aims to investigate if T2DM reduces ALS risk through glycemic or lipid pathways using a two-step Mendelian Randomization (MR) approach.
METHODS: Summary-level genetic data were sourced from FinnGen (n = 440,735), MA

INTRODUCTION AND AIMS: Observational studies suggest an association between metabolic syndrome (MetS) and periodontitis. However, observational studies are susceptible to reverse causation and confounding factors, so the causality of this association is uncertain. Causal association between compounds of MetS and periodontitis has been well studied. Using Mendelian randomisation (MR), we aimed to comprehensively evaluate the bidirectional relationship between MetS as a whole and periodontitis and provide clinical insight.
METHODS: We used genetic instruments from the most comprehensive genome-

Since December 2019, COVID-19 has triggered a global pandemic. The association of COVID-19 with the long-term reproductive situation of women and males is not clear. Thus, our aim was to assess the causal association between COVID-19 and infertility using Mendelian randomization (MR) analysis based on the OpenGWAS database. Two-sample MR analysis was conducted using one genome-wide association study (GWAS) on COVID-19 and infertility in individuals of European ancestry. The summary data of genetic variation come from the GWAS in European populations. We applied several MR methods, including MR

Genetic variation at HLA-DRB1 is a risk factor for visceral leishmaniasis (VL) caused by Leishmania donovani. The single nucleotide polymorphism rs9271252 upstream of the DRB1 gene provides a perfect tag for protective versus risk HLA-DRB1 four-digit alleles. In addition to the traditional role of the membrane-distal region of HLA class II molecules in antigen presentation and CD4 T-cell activation, the membrane-proximal region mediates 'non-traditional' multi-functional activation, differentiation, or death signals, including in DR-expressing T cells. To understand how HLA-DR contributes to d
