Genetic analysis of human adenovirus type 108 circulating in China during 2014-2024.

Human adenovirus type 108 (HAdV-108) has been detected in multiple countries, including China, and is associated with severe acute respiratory infection (ARI) in children, with reported fatalities. However, studies on HAdV-108 remain limited. This study aimed to investigate the clinical and genetic characteristics of HAdV-108 in ARI children in China. From 2014 to 2024, 6720 respiratory samples were collected from hospitalized children with ARI at ten hospitals across northern and southern China, of which 505 (7.51%) tested positive for HAdV. The whole-genome and three major capsid protein gen

Long-read genome sequencing identifies cryptic structural variants in congenital aniridia cases.

BACKGROUND: Haploinsufficiency of the transcription factor PAX6 is the main cause of congenital aniridia, a genetic disorder characterized by iris and foveal hypoplasia. 11p13 microdeletions altering PAX6 or its downstream regulatory region (DRR) are present in about 25% of patients; however, only a few complex rearrangements have been described to date. Here, we performed nanopore-based whole-genome sequencing to assess the presence of cryptic structural variants (SVs) on the only two unsolved "PAX6-negative" cases from a cohort of 110 patients with congenital aniridia after unsuccessfully sh

Relationship Between Genetic Polymorphisms in Cell Cycle Regulatory Gene TP53 and Polycystic Ovarian Syndrome: A Case-Control Study and In Silico Analyses.

Polycystic ovarian syndrome (PCOS) is a complex endocrine and metabolic condition with several potential causes. Insulin resistance is a hallmark of PCOS that often coexists with hirsutism, hyperandrogenism, being overweight, and hormonal imbalances. The functioning of multiple replication and transcription factors is regulated by tumor suppressor genes (TSGs), which play a crucial role in maintaining genomic integrity and controlling the cell cycle of granulosa cells. In the present study, we examined how three single nucleotide polymorphisms (SNPs) in TP53, a cell cycle regulatory gene, affe

Congenital goiter with hypothyroidism caused by a 5' splice site mutation in the thyroglobulin gene.

In this work we have extended our initial molecular studies of a consanguineous family with two affected goitrous siblings (H.S.N. and Ac.S.N.) with defective thyroglobulin (Tg) synthesis and secretion because of a homozygotic deletion of a fragment of 138 nucleotides (nt) in the central region of the Tg mRNA, identified previously in H.S.N. In order to identify the intron/exon boundaries and to analyze the regions responsible for pre-mRNA processing corresponding to a 138 nt deletion, we performed a screening of a human genomic library. The intron/exon junction sequences were determined from

Mitochondrial genomic alterations in cholangiocarcinoma cell lines.

Cholangiocarcinoma (CCA) is a diverse collection of malignant tumors that originate in the bile ducts. Mitochondria, the energy converters in eukaryotic cells, contain circular mitochondrial DNA (mtDNA) which has a greater mutation rate than nuclear DNA. Heteroplasmic variations in mtDNA may suggest an increased risk of cancer-related mortality, serving as a potential prognostic marker. In this study, we investigated the mtDNA variations of five CCA cell lines, including KKU-023, KKU-055, KKU-100, KKU213A, and KKU-452 and compared them to the non-tumor cholangiocyte MMNK-1 cell line. We used O

Analysis of genetic variation in Theiler's virus during persistent infection in the mouse central nervous system.

The genetic changes occurring in the BeAn strain of Theiler's murine encephalomyelitis virus (TMEV) during persistent infection in the mouse central nervous system (CNS) were studied. RNase T1-oligonucleotide fingerprinting of the RNAs of 28 BeAn viruses isolated at various times postinfection (p.i.) demonstrated that mutation occurred throughout the infection. Although plaque-purified BeAn virus was used to inoculate mice intracerebrally, genetically different viruses were recovered from the CNS. One to three oligonucleotide changes were found up to Day 152 p.i., but all three viruses isolate

A Frosty Genetic Screen Unmasks a Major Regulatory Role for SHORT VEGETATIVE PHASE of Flowering in Response to a Cold Snap.

Flowering in plants is controlled by internal and environmental signals, with temperature playing a critical role. Accordingly, Arabidopsis thaliana plants show temperature-dependent variation in flowering time. Because unexpected cold periods can occur at any time, plants have evolved mechanisms to detect such cold snaps and delay flowering. Plants are more cold-tolerant in the vegetative stage, whereas flowers are sensitive and often show reduced reproductive success due to damage to floral structures and gametes. At the molecular level, delayed flowering can be caused by repression of the F

Allele frequency trajectories across age groups reveal ongoing natural selection shaping disease susceptibility.

Understanding how selection shapes disease risk remains challenging. Variants influencing complex traits, including common diseases, can also impact fitness and thus be constrained by purifying selection. Consequently, genetic variance underlying disease susceptibility may be attributed to low-frequency, population-specific variants. We analyzed 509,817 genome-wide variants from 72,635 Han Taiwanese individuals to identify loci showing age-dependent allele frequency shifts that signal ongoing selection. After adjusting for potential age-related population structure, we detected 168 variants de

Fine-Scale Population Genomics Reveals Genetic Differentiation in the Brooding Amphipod Cheirimedon femoratus Across the South Shetland Islands, Antarctica.

Antarctic marine ecosystems are sensitive to environmental change, and impacts on processes such as population connectivity will play a fundamental role in future population dynamics and persistence, affecting short-term demography and long-term evolution. We investigated the population genomics of the common benthic brooding Antarctic amphipod Cheirimedon femoratus (Pfeffer, 1888), using 8837 high-quality single-nucleotide polymorphisms (SNPs) from 87 individuals collected at 4 sites in the South Shetland Islands, separated by up to 200 km: Deception Island, King George Island, Livingston Isl

A 2004-2025 Bibliometric Study of Genetic Variation and Multiomics Biomarkers in Sepsis Based on 940 Publications.

Sepsis is a highly heterogeneous syndrome, and conventional clinical indicators and single biomarkers often fail to capture its biological complexity or support precise risk stratification. To clarify the development of research in this area, this bibliometric study analyzed the literature on the clinical translation of genetic variation and multiomics biomarkers in sepsis. Publications were retrieved from PubMed, Web of Science Core Collection, and Scopus for the period from 2004 to 2025, and 940 eligible records were analyzed using Bibliometrix, VOSviewer, and CiteSpace. The results showed r

Strain-resolved biology of Lactobacillus johnsonii across host systems.

Lactobacillus johnsonii is widely described as a probiotic species, but current evidence supports a strain-resolved view. Its biological effects depend on strain background, host niche, diet, challenge context and delivery format, including live cells, cell-derived structures and postbiotic fractions. This review synthesizes comparative genomic, mechanistic and intervention studies on L. johnsonii, using weaned piglets as the main translational reference because barrier injury, inflammation, microbial disruption, diarrhea and impaired growth often coincide after weaning. Piglet studies show th

Evolutionary Characteristics of the Protamine-Like Protein (PLI) Gene Family in Syngnathidae Fishes and Its Potential Regulatory Role in Sperm Morphological Specialization.

Syngnathidae fishes possess an elongated sperm morphology that is adapted for fertilization within the brood pouch. Protamine-like proteins (PLs) present a class of DNA-binding proteins that replace histones and bind to DNA during the late stages of vertebrate spermatogenesis, promoting chromatin condensation into compact sperm nuclei. To verify the correlation between the specialized sperm morphology of Syngnathidae fishes and PLI (PL Type I) gene family evolution, this study analyzed sperm morphology in 61 teleost species and revealed that Syngnathidae fishes have relatively elongated sperm

Missense mutations in the SNCA gene: Molecular mechanisms and clinical implications.

The SNCA gene on chromosome 4 encodes the alpha-synuclein (αSyn) protein, which plays a central role in the pathogenesis of synucleinopathies, including Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA). While αSyn has established roles in synaptic vesicle dynamics and neuronal signaling, alterations in SNCA regulation and sequence contribute to protein misfolding, aggregation, and loss of function. Alterations in secondary and tertiary structure, as well as protein aggregation, affect biochemical interactions, ultimately leading to pathogenesis. This

Disruption and adaptation: infant gut microbiota's dynamic response to SARS-CoV-2 infection.

BACKGROUND: The responses of the infant gut microbiota to infection significantly disrupt the natural intrahost evolutionary processes of the microbiome. Here, we collected a 16-month longitudinal cohort of infant gut microbiomes affected by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Then, we developed a multicriteria approach to identify core interaction network driving community dynamics under environmental disturbances, which we termed the Conserved Variated Interaction Group (CVIgroup). RESULTS: The CVIgroup showed significant advantages on pinpointing a sparse set asso

Exploring potential mechanisms of an African protective locus for Alzheimer's disease in APOEε4 carriers.

INTRODUCTION: We recently described that the African-specific A allele of rs10423769, which lies in an area of segmental duplications, reduces Alzheimer's disease (AD) risk by ∼ 75% in apolipoprotein E (APOE) ε4 homozygotes. METHODS: Short and long-read sequencing were used to identify the haplotype harboring rs10423769_A and examine DNA methylation and structural variation (SV). RESULTS: A unique 21 kb haplotype is shared amongst all rs10423769_A carriers (r2 > 0.95) and is present in all African ancestry tested. We identified methylation differences between the non-protective and rs1042376

Mapping Cerebellar Morphology in 15q11.2 CNV Carriers Using Normative Modeling.

Copy number variations (CNVs) at the 15q11.2 locus of the human genome have been associated with altered brain structure and increased risk for neurodevelopmental and neuropsychiatric disorders. The cerebellum is increasingly seen as a crucial brain region for neurodevelopmental conditions, yet the effects of 15q11.2 CNVs on cerebellar morphology remain largely unclear. Importantly, 15q11.2 CNVs shows reduced or incomplete penetrance (meaning that not all CNV carriers are affected) and variable expressivity (meaning that symptoms may differ between individuals with the same genetic alteration)

Emerging roles of natural killer cell ligands-HLA-E, HLA-F, HLA-G, MICA, and MICB-in in vitro fertilization outcomes.

Infertility affects approximately one in six individuals globally and represents a complex public health concern influenced by a range of biological, environmental, and socioeconomic factors. In vitro fertilization (IVF) has emerged as a pivotal assisted reproductive technology (ART), yet its success is often hindered by recurrent implantation failure (RIF) and hypertensive complications such as preeclampsia (PE). Recent research highlights the critical role of the immune system particularly non-classical Human Leukocyte Antigen (HLA) class I molecules (HLA-G, HLA-E, HLA-F) and MHC class I cha
