1-Octen-3-ol suppresses Penicillium expansum development, patulin biosynthesis and pathogenicity via oxidative stress and G-protein signaling.

Penicillium expansum is widely recognized as an important cause of postharvest apple decay, owing to its ability to induce blue mold and to produce the mycotoxin patulin, leading to significant economic losses and food safety risks. 1-Octen-3-ol, a natural volatile organic substance, has diverse biological activity. This study investigated the effects of 1-octen-3-ol on the growth, pathogenicity, and patulin biosynthesis of P. expansum, and explored the underlying antifungal mechanisms. Our results demonstrated that 1-octen-3-ol markedly restrained mycelial growth, spore germination, and sporu

Necroptosis activates NLRP3 inflammasome to drive necroinflammation in T-2 toxin-induced cartilage damage.

As a highly toxic mycotoxin present in contaminated food sources, T-2 toxin disrupts skeletal development by inducing cartilage injury. However, the underlying molecular mechanisms remain unclear. Following four weeks of gavage administration of T-2 toxin (200 ng/g body weight/day) to three-week-old male C57BL/6 mice, chondrocyte death in articular cartilage was accompanied by increased levels of p-RIPK3, p-MLKL, NLRP3, cleaved caspase-1, and IL-1β. In C28/I2 chondrocytes, T-2 toxin decreased cell viability and increased LDH release, accompanied by enhanced NLRP3 inflammasome-related inflammat

Maternal mycoestrogen exposure in relation to timing of birth and placental corticotropin releasing hormone concentrations in a U.S. pregnancy cohort.

Zearalenone (ZEN) is a mycotoxin that widely contaminates human food supplies. In animal models, exposure to ZEN has extensive impacts on reproduction and pregnancy including contributing to early parturition. Here, we translate that research to humans by examining gestational exposure to ZEN in relation to the timing of birth and placental corticotropin releasing hormone (pCRH), a hormone that regulates the timing of parturition. Utilizing data from a pregnancy cohort (UPSIDE; Rochester, NY; n = 299), we investigated exposure to ZEN and its metabolites in relation to: 1) gestational age at bi

Construction of a Self-Enhanced Electrochemiluminescent Sensor Based on Tandem Signal Amplification and a Self-Luminescent Lanthanide Covalent-Organic Polymer for Ochratoxin A Assay.

Ochratoxin A (OTA) is a type of mycotoxin found in various contaminated foods, and it is highly toxic to the livers and kidneys of humans. Herein, we develop a self-enhanced electrochemiluminescent (ECL) sensor based on tandem signal amplification and a self-luminescent europium covalent-organic polymer (Eu-COPTMT-BPA) for OTA assay. Eu-COPTMT-BPA is a self-enhanced ECL emitter that is obtained by using 2,4,6-trimethyl-1,3,5-triazine (TMT) and 2,2'-bipyridine-5,5'-dicarbaldehyde (BPA) as the first and second ligands to form a highly conjugated structure for sensitizing the Eu3+ luminescence. Bio-preservative potential of marjoram and fennel essential oil nano-emulsions against toxigenic fungi in citrus: integrating in-vitro, in-vivo, and in-silico approaches.

Citrus fruits, known for their vibrant flavours and health benefits, are susceptible to fungal attacks, particularly from toxigenic Penicillium fungi, which pose a significant pre- and post-harvest hazard. However, aromatic oils and their nanoparticles may effectively address this issue. Marjoram and fennel oils, alongside their nanoparticles, were extracted, and their aromatic constituents and antimicrobial activities were evaluated. A simulated medium with fungal spores was used to assess anti-toxigenic activity, and a simulated infection experiment was conducted with orange and lemon fruits

Mitigating T-2 toxin toxicity in Quail: The protective power of sodium butyrate.

As one of the most common worldwide contaminants in agricultural production, the T-2 mycotoxin is commonly found in moldy feed and its raw materials. It can slow the growth and suppress the immune function of farm animals, resulting in reduced economic benefits. As a feed additive, sodium butyrate can enhance immune function. However, the toxicological effects of the T-2 toxin on the spleen, thymus, and bursa of Fabricius and the protective mechanism of sodium butyrate against the T-2 toxin in quails are not known. In this study, 1-day-old Korean quails were fed either with T-2 toxin (0.9 mg/k

2,4-Diacetylphloroglucinol from multi-stress tolerant Pseudomonas guariconensis VDA8: Seed-applied bioprotectant for groundnut (Arachis hypogaea L.) against Aspergillus flavus NCIM 524 for post-harvest preservation.

The fungal contaminated crops or post-harvest products are unfit for consumption, hazardous to human health and threat to food safety. Soil borne phytopathogenic microbes cause severe yield losses at various stages and contribute to 40-50% crop mortality. Post-harvest fungal deterioration during storage is frequently observed primarily because of high oil content. At present, chemical disinfectants and agrichemicals are extensively used to control seed contaminating fungal infection as seed priming agent and/or post-harvest storage protectants. However, application of synthetic fungicides requ

TSC2/mTORC1 integrates deoxynivalenol signals recognized by membrane receptors IR and EGFR to restrict intestinal stem cell function.

Deoxynivalenol (DON) is a chemically stable mycotoxin with a slow natural degradation rate. Consumption of DON-contaminated food and feed poses significant health risks to human and livestock, leading to reduced productivity and substantial economic losses. The functionality of intestinal stem cells (ISCs) are compromised following sustained intracellular deoxynivalenol (DON) stress. Yet, it remains unclear how membrane receptors integrate extracellular DON to impair orderly ISC fate commitments. Here, we found that mechanistic target of rapamycin complex 1 (mTORC1), as well as its upstream si

FpCBS Affects a Diverse Range of Functions of Fusarium pseudograminearum Impacting Its Virulence to Wheat.

A cystathionine β-synthase (CBS) gene, FpCBS, of the wheat crown rot pathogen, Fusarium pseudograminearum, was identified. FpCBS was highly expressed during conidiation and the early phases of infection. Compared to the wild-type and FpCBS complemented strains, the FpCBS deletion strain had a similar hyphal morphology and growth rate but reduced conidiation. In culture, the FpCBS deletion strain showed broad-scale changes with a decrease in H2S levels, changes in sulphur-containing amino acids and 1207 differentially expressed proteins. The deletion strain also showed reduced deoxynivalenol (D
