The APOLLO trial: a proof-of-concept study for vitamin A nasal drops in COVID-19-related postinfectious olfactory dysfunction.

Postinfectious olfactory dysfunction (PIOD) is common in COVID-19 patients. This 2-arm double-blinded randomized controlled trial (RCT) aimed to establish proof-of-concept for vitamin A versus placebo as a treatment modality for patients with PIOD. This study compared 9,000 IU daily self-administered vitamin A intranasal drops versus peanut oil drops over 12 wk in COVID-19 patients with PIOD. Outcome measures included: olfactory bulb volume (OBV), olfactory sulcus depth, cerebral functional MRI blood oxygen level dependent (BOLD) signal, Sniffin' Sticks TDI score, SSParoT, olfactory disorder q…

Olfactory ensheathing cell membrane properties are shaped by connectivity.

Olfactory ensheathing cells (OECs) have been repeatedly implicated in mediating plasticity, particularly in situ in the olfactory nerve in which they support the extension of olfactory sensory neuron (OSN) axons from the olfactory epithelium to the olfactory bulb (OB). OECs are specialized glia whose processes surround OSN axon fascicles within the olfactory nerve and across the OB surface. Despite their purported importance in promoting axon extension, and following transplants, little is known about either morphology or biophysical properties of OECs in situ. In particular, cell-cell interac…

Generalization of fear learning is shaped by inhibitory sensory processing in mice.

When an organism learns that a sensory stimulus predicts a threat, the brain's neural representation of the stimulus somehow incorporates th … neurons in proportion to the generalized fear of each odor. Locally blocking GABAB receptors in the olfactory bulb caused mice to switch from generalizing fear in proportion to similarity to instead overgeneralizing fear to all odors equally. Learning-induced sensory plasticity thus plays a causal role in shaping fear generalization.

Apoptotic neurodegeneration induced by influenza A virus infection in the mouse brain.

Neurodegeneration in the brain induced by the WSN strain of influenza A virus was investigated after stereotaxic introduction into the olfactory bulb of C57BL/6 mice. Immunohistochemistry detected WSN virus-infected neurons in the anterior olfactory nucleus as early as day 3 postinfection. Thereafter, they became shrunken and showed loss of neurite-immunolabeling and chromatin condensation. Infected neurons died by day 12, degenerating into multiple small granular bodies. The terminal deoxynucleotidyltransferase-mediated dUTP nick end-labeling method demonstrated DNA fragmentation in infected …

Regulation of continuous but complex expression pattern of Six1 during early sensory development.

BACKGROUND: In vertebrates, cranial sensory placodes give rise to neurosensory and endocrine structures, such as the olfactory epithelium, inner ear, and anterior pituitary. We report here the establishment of a transgenic mouse line that expresses Cre recombinase under the control of Six1-21, a major placodal enhancer of the homeobox gene Six1. RESULTS: In the new Cre-expressing line, mSix1-21-NLSCre, the earliest Cre-mediated recombination was induced at embryonic day 8.5 in the region overlapping with the otic-epibranchial progenitor domain (OEPD), a transient, common precursor domain for

A screen for genes expressed in the olfactory organs of Drosophila melanogaster identifies genes involved in olfactory behaviour.

BACKGROUND: For insects the sense of smell and associated olfactory-driven behaviours are essential for survival. Insects detect odorants with families of olfactory receptor proteins that are very different to those of mammals, and there are likely to be other unique genes and genetic pathways involved in the function and development of the insect olfactory system. METHODOLOGY/PRINCIPAL FINDINGS: We have performed a genetic screen of a set of 505 Drosophila melanogaster gene trap insertion lines to identify novel genes expressed in the adult olfactory organs. We identified 16 lines with expre

Efficiency of laryngeal motor nerve repair is greater with bulbar than with mucosal olfactory ensheathing cells.

The real ability of OECs provided by olfactory mucosa cultures (OM-OECs) and those from olfactory bulb cultures (OB-OECs) must be better characterized in order to propose their future clinical application. Therefore, we used a lesion of the vagus nerve (VN), which constitutes a severe motor denervation due to long distance of the muscular targets (4.5 cm). We performed a section/anastomosis surgery of the VN, at the third tracheal ring. Then, OM-OECs and OB-OECs were injected in matrigel around the lesion site. Three months after surgery, laryngeal muscle activity, synkinesis phenomena and lat…

Children who can't smell the coffee: isolated congenital anosmia.

Two children with isolated congenital anosmia, a rare syndrome of deficient restricted neuronal migration, are presented with early diagnosis confirmed by standardized smell testing and detailed neuroimaging studies. Recognition of this disorder and its spectrum of presentations provides important insights into the molecular mechanisms underlying the development of the olfactory system.

Nerve growth factor receptor expression in the young and adult rat olfactory system.

Nerve growth factor (NGF) and its receptor (NGFR) are proteins that have a role in the normal development and survival of neurons in the peripheral and central nervous systems. During development, NGF is necessary for outgrowth of axons and establishment of synapses, and NGFR is the transmembrane protein that binds NGF and brings it into the cell. NGF and NGFR expression in the rat olfactory system have been studied previously, and age differences in NGFR are explored further in this study, using immunocytochemistry and immunoelectron microscopy to determine the changes in two different ages:

Persistence of the nervus terminalis in adult bats: a morphological and phylogenetical approach.

The presence of the terminalis system in adult bats is demonstrated by light microscopical investigation of several species of Microchiroptera. In late embryonic and fetal stages of the mouse-eared bat (Myotis myotis) the compact central terminalis ganglion gradually differentiates into a three-dimensional network of cord-like ganglia and fiber bundles. Rostrally the terminalis system is in immediate contact with the medial-most fila olfactoria; caudally terminalis rootlets attach near the border between the olfactory bulb and the septum of the brain. With respect to the findings presented her…

Regional changes in brain histamine levels following dietary-induced thiamine deficiency in rats.

Histamine levels in thiamine deficient rats were significantly lower in the hippocampus, amygdala, olfactory bulb, thalamus and pons-medulla oblongata than those of normal and pair-fed groups. In the case of the hypothalamus, thiamine deficiency produced a significant increase in histamine levels. These changes observed in the thiamine deficient group were reversed to the normal levels by supplying the normal diet. These data present a new finding that thiamine deficiency affects the central histaminergic neuron system as well as other monoaminergic systems.
