PADI4-mediated citrullination of histone H3 stimulates HIV-1 transcription.

HIV-1 infection establishes a reservoir of long-lived cells with integrated proviral DNA that can persist despite antiretroviral therapy (ART). Certain reservoir cells can be reactivated to reinitiate infection. The mechanisms governing proviral latency and transcriptional regulation of the provirus are complex. Here, we identify a role for histone H3 citrullination, a post-translational modification catalyzed by protein-arginine deiminase type-4 (PADI4), in HIV-1 transcription and latency. PADI4 inhibition by the small molecule inhibitor GSK484 reduces HIV-1 transcription after T cell activat

Palmitoylation in cardiometabolic diseases: From molecular mechanisms to clinical implications.

Cardiometabolic diseases (CMDs), encompassing both cardiovascular diseases and metabolic syndromes, are among the leading causes of global morbidity and mortality. Protein S-palmitoylation, a reversible lipid-based post-translational modification, has emerged as a crucial regulator of cellular function and homeostasis. Palmitoylation is catalysed by palmitoyl transferases (PATs), also known as ZDHHC (zinc finger Asp-His-His-Cys) proteins, while a series of deacylases mediate the process of depalmitoylation. By modulating protein hydrophobicity, stability, subcellular localization, enzymatic ac

ZDHHC9 and spermine metabolism: a palmitoylation-driven pathway to prostate carcinogenesis.

BACKGROUND: Spermine is a natural metabolite that has been shown to hinder the progression of prostate cancer. Protein palmitoylation, a recently identified post-translational lipid modification, is catalyzed by a family of 23 enzymes known as zinc finger DHHC-type palmitoyltransferases (ZDHHCs). This study primarily investigates the role of ZDHHC9 in spermine metabolism and its potential association with prostate cancer. METHODS: We employed immunohistochemistry, Western blotting, and RT-qPCR techniques to assess ZDHHC9 expression in prostate cancer tissues and cell lines. The connection betw

Replication-associated base excision repair/single-strand break repair regulates PARG inhibitor response via the PRMT1/PRMT5/ATR axis.

Poly(ADP-ribose) polymerases 1 and 2 (PARP1/PARP2), and poly(ADP-ribose) glycohydrolase (PARG), modulate the level of poly(ADP-ribose) (PAR), a post-translational protein modification, in response to DNA damage or replication stress. Here, we find that replication-dependent and PARP1/PARP2-mediated PARylation recruits the base excision repair (BER)/single-strand break repair (SSBR) scaffold protein XRCC1 and the associated factors DNA polymerase β (POLB), aprataxin (APTX), and DNA ligase isoform 3 (LIG3). Further, these BER/SSBR proteins promote resistance to inhibitors of PARP1/PARP2 and PARG

DDRGK1 preserves intervertebral disc development through ufmylation.

UFMylation, similar to ubiquitination, is a unique post-translational modification which is indispensable in hematopoiesis, neurogenesis and chondrogenesis. However, its role in intervertebral disc development remains unclear. In this study, we focused on DDRGK domain containing protein 1 (DDRGK1), a pivotal component involved in UFMylation, and generated Ddrgk1fl/fl; Acan-CreERT2 (Ddrgk1cKO) mice to explore DDRGK1's regulatory function in the nucleus pulposus and cartilage endplate. We found that Ddrgk1 conditional knockout led to severe retardation of spinal growth, disruption of disc cellul

The hypusine pathway in Ixodes ricinus: molecular cloning and validation of deoxyhypusine synthase as a novel target for drug discovery to treat and prevent vector borne diseases.

Ticks are a group of arthropod vectors transmitting a variety of human pathogens, like Borrelia and the tick-borne Encephalitis virus. In Europe, Ixodes is the most important tick due to its wide distribution. Since the 20th century, Ixodes has significantly spread due to changes in biodiversity. Thus, there is an urgent need to decrease tick ubiquity in the environment to control tick-borne diseases. Deoxyhypusine Synthase (DHS) catalyzes the first step in the post translational modification (PTM) of the amino acid hypusine in eukaryotic initiation factor (eIF5A). Modified eIF5A plays a cruci

An in-silico analysis to identify structural, functional and regulatory role of SNPs in hMRE11.

Meiotic recombination 11 (MRE11) is a component of the tri-molecular MRE11-RAD50-NBS1 (MRN) complex, which functions as an exonuclease and endonuclease which is involved in identifying, signalling, protecting and repairing double-strand breaks in DNA (DSBs). Ataxia-telangiectasia-like disorder (ATLD) 1 and Nijmegen breakage syndrome (NBS)-like disorder are MRE11 associated diseases. In the present study, we used an integrated computational approach to identify the most deleterious SNPs and their structural and functional impact on human MRE11. Five of the 68 observed non-synonymous SNP (nsSNPs

Identification of Novel Native Autoantigens in Rheumatoid Arthritis.

The majority of patients diagnosed with rheumatoid arthritis (RA) have developed autoantibodies against neoepitopes in proteins that have undergone post-translational modification, e.g., citrullination or carbamylation. There is growing evidence of their molecular relevance and their potential utility to improve diagnosis, patient stratification, and prognosis for precision medicine. Autoantibodies reacting to native proteins may also have a role in RA pathogenesis, however, their reactivity patterns remain much less studied. We hypothesized that a high-density protein array technology could s

Prion diseases and emerging prion diseases.

Transmissible spongiform encephalopathies (TSEs), also called prion diseases, are fatal neurodegenerative disorders. An abnormal isoform of the prion protein (PrP(Sc)) generated by post-translational modification of the cellular prion protein (PrP(C)) is believed to be the main component of this infectious agent. PrP(Sc) is relatively resistant to proteinase K (PK) digestion. This characteristic has been widely accepted as the physicochemical basis for distinguishing between PrP(C) and PrP(Sc). PrP(C) is a glycoprotein that contains 2 Asn-linked glycosylation sites; it is present in the cells

Direct detection of transient alpha-helical states in islet amyloid polypeptide.

The protein islet amyloid polypeptide (IAPP) is a glucose metabolism associated hormone cosecreted with insulin by the beta-cells of the pancreas. In humans with type 2 diabetes, IAPP deposits as amyloid fibers. The assembly intermediates of this process are associated with beta-cell death. Here, we examine the rat IAPP sequence variant under physiological solution conditions. Rat IAPP is mechanistically informative for fibrillogenesis, as it samples intermediate-like states but does not progress to form amyloid. A central challenge was the development of a bacterial expression system to gener

Regulation of cold-inducible RNA-binding protein (CIRBP) in response to cellular stresses.

Cold-inducible RNA-Binding Protein (CIRBP) is a general stress-response factor in vertebrates harboring two domains: an RNA-recognition motif and a regulatory domain rich in RG/RGG motifs. CIRBP has been described to bind mRNAs upon various stress conditions (cold, infections, UV, hypoxia …) and regulate their stability and translation. The proteins encoded by its targets are involved in key stress-responsive cellular pathways including apoptosis, inflammation, cell proliferation or translation, thus allowing their coordination. Due to its role in regulating central cellular functions, the exp

Multi-omics analysis delineates the distinct functions of sub-cellular acetyl-CoA pools in Toxoplasma gondii.

BACKGROUND: Acetyl-CoA is a key molecule in all organisms, implicated in several metabolic pathways as well as in transcriptional regulation and post-translational modification. The human pathogen Toxoplasma gondii possesses at least four enzymes which generate acetyl-CoA in the nucleo-cytosol (acetyl-CoA synthetase (ACS); ATP citrate lyase (ACL)), mitochondrion (branched-chain α-keto acid dehydrogenase-complex (BCKDH)) and apicoplast (pyruvate dehydrogenase complex (PDH)). Given the diverse functions of acetyl-CoA, we know very little about the role of sub-cellular acetyl-CoA pools in parasit

In-depth O-glycosylation characterization and comparison of commercially available etanercept biosimilars by using Electron Activated Dissociation.

Glycosylation is one of the important post-translational modifications in protein drugs expressed by eukaryotic cells. It is closely related to the conformation, stability, and solubility of therapeutic proteins and holds vital biological significance. Etanercept contains 26 O-glycosylation sites with high glycosylation heterogeneity, which makes the analysis of O-glycosylation highly challenging. In this paper, using liquid chromatography-mass spectrometry (LC-MS), 11 O-glycosylation on the single chain of etanercept were first identified at the intact protein level, with Core1 and Core2 O-gl

An expression framework for production of recombinant methionine aminopeptidase in Escherichia coli: a case study of upstream process.

Incomplete removal of the initiator methionine is a frequent bottleneck in Escherichia coli-based production of recombinant proteins, causing heterogeneity and increased immunogenicity of biopharmaceuticals. Methionine aminopeptidase (MAP) is the key enzyme responsible for this post-translational modification, yet its endogenous activity is rapidly saturated under high-level expression conditions. Here, we report a case-study evaluation of a pBR322-derived expression system, previously applied to therapeutic peptides and insulin analogs, in combination with fed-batch cultivation for recombinan

Mechanisms, Biomarkers and Therapeutic Implications of Neuroinflammation in Alzheimer's Disease.

Alzheimer's disease (AD) represents the most prevalent neurodegenerative disorder worldwide, affecting millions of individuals and imposing substantial socioeconomic burdens. While traditional research has focused on amyloid-β (Aβ) plaques and neurofibrillary tangles as primary pathological hallmarks, mounting evidence implicates neuroinflammation as a critical third pillar in AD pathogenesis. This review critically evaluates current understanding of neuroinflammatory mechanisms in AD, examining the complex interplay between cellular mediators, molecular pathways and environmental triggers acr

SUMOylation inhibition potentiates the glucocorticoid receptor to program growth arrest of acute lymphoblastic leukemia cells.

Glucocorticoids are a mainstay in the treatment of B-cell acute lymphoblastic leukemia (B-ALL). The glucocorticoid receptor (GR), a ligand-activated transcription factor (TF), mediates their actions. Chromatin occupancy, chromatin-protein networks (chromatomes) and gene programmes of GR are regulated by SUMOylation, a post-translational modification with therapeutic implications in other hematomalignancies. To unravel the GR-SUMOylation crosstalk in B-ALL, we induced hypoSUMOylation in NALM6 B-ALL cells with a SUMOylation inhibitor (SUMOi, ML-792). Genome-wide profiling of GR and SUMO chromati

A novel approach to explore metabolic diseases: Neddylation.

Protein post translational modification (PTM) is the main regulatory mechanism for eukaryotic cell function, among which ubiquitination is based on the reversible degradation of proteins by the ubiquitin proteasome system to regulate cell homeostasis. The neural precursor cell expressed developmental downregulated gene 8 (NEDD8) is a kind of ubiquitin like protein that shares 80 % homology and 60 % identity with ubiquitin. The PTM process by covalently binding NEDD8 to lysine residues in proteins is called neddylation. The neddylation reaction could be regulated by NEDD8, its precursors, subst

Proline Isomerization: From the Chemistry and Biology to Therapeutic Opportunities.

Proline isomerization, the process of interconversion between the cis- and trans-forms of proline, is an important and unique post-translational modification that can affect protein folding and conformations, and ultimately regulate protein functions and biological pathways. Although impactful, the importance and prevalence of proline isomerization as a regulation mechanism in biological systems have not been fully understood or recognized. Aiming to fill gaps and bring new awareness, we attempt to provide a wholistic review on proline isomerization that firstly covers what proline isomerizati
