The use of the PREP system for handling of body fluids.

The Du Pont PREP automated sample processor is a centrifugally based, microprocessor controlled instrument that was designed for extraction of samples from biological fluids. Extraction takes place in cartridges containing either organic resins or bonded silica packings as extraction sorbants. This paper will discuss the application of several lipophilic and ion exchange sorbants to the extraction of biological samples from body fluids. The advantages of these different types of sorbants will be compared and their performance with automated sample preparation will be shown. A variety of applic

Biosafety evaluations of well-dispersed mesoporous silica nanoparticles: towards in vivo-relevant conditions.

This study aimed to investigate how mesoporous silica nanoparticles (MSNs), especially focussing on their surface functional groups, interacted with Raw 264.7 macrophages, as well as with zebrafish embryos. Upon introducing nanoparticles into a biological milieu, adsorption of proteins and biomolecules onto the nanoparticle surface usually progresses rapidly. Nanoparticles bound with proteins can result in physiological and pathological changes, but the mechanisms remain to be elucidated. In order to evaluate how protein corona affected MSNs and the subsequent cellular immune responses, we exp

Biological Impact of Silicon Nitride for Orthopaedic Applications: Role of Particle Size, Surface Composition and Donor Variation.

The adverse biological impact of orthopaedic wear debris currently limits the long-term safety of human joint replacement devices. We investigated the role of particle size, surface composition and donor variation in influencing the biological impact of silicon nitride as a bioceramic for orthopaedic applications. Silicon nitride particles were compared to the other commonly used orthopaedic biomaterials (e.g. cobalt-chromium and Ti-6Al-4V alloys). A novel biological evaluation platform was developed to simultaneously evaluate cytotoxicity, inflammatory cytokine release, oxidative stress, and

In vivo biodistribution and clearance studies using multimodal organically modified silica nanoparticles.

Successful translation of the use of nanoparticles from laboratories to clinics requires exhaustive and elaborate studies involving the biodistribution, clearance, and biocompatibility of nanoparticles for in vivo biomedical applications. We report here the use of multimodal organically modified silica (ORMOSIL) nanoparticles for in vivo bioimaging, biodistribution, clearance, and toxicity studies. We have synthesized ORMOSIL nanoparticles with diameters of 20-25 nm, conjugated with near-infrared (NIR) fluorophores and radiolabeled them with (124)I, for optical and PET imaging in vivo. The bio

Assessment of natural and synthetic wollastonite as source for bioceramics preparation.

Pseudowollastonite ceramics (beta-CaSiO3) from synthetic and natural sources were assessed with regard to their properties relevant to biomedical applications. Synthetic and natural CaSiO3 powders, with average particle size of 1.6 and 13.2 microm, respectively, were first employed. Powders were pressed and sintered at 1400 degrees C for 2 h. Pseudowollastonite was the only crystalline phase in sintered materials. Glassy phase, eight times more abundant in sintered natural wollastonite (SNW) than in the synthetic one (SSW), was observed in grain boundaries and in triple points. Larger grains a

Evaluation of metal bond strength to dentin and enamel using different adhesives and surface treatments.

Because adherence of base metal alloys is important for the long-term clinical success of adhesive fixed partial dentures, it has been necessary to improve adhesion to metal substrate by using different surface treatments. This study used different surface conditioning methods and two different luting resins to evaluate the shear bond strength of base metal alloys to dentin and enamel. Sixty noncarious freshly extracted human teeth were mounted in a plastic holder filled with autopolymerized acrylic resin. After the roots were removed and 30 flat enamel and 30 flat dentin surfaces were exposed

Designing of the centers for adsorption of bile acids on a silica surface.

Silicas chemically modified with attached aminopropyl, imidazolyl, and trimethylsilyl groups, with adsorptive and coordinative grafted hemin were synthesized. Adsorption of some bile acids on the surface of hydroxylated silica, synthesized siliceous adsorbents and cholestyramine has been studied. It was found that the main contribution to the total adsorption is caused by electrostatic attraction between anions of bile acids and positively charged sites of the surface of modified silica and also by dispersion interactions between steroid skeleton of bile acids and functional groups of modified

Photoinduced phenomena in layer-by-layer films of poly(allylamine hydrochloride) and Brilliant Yellow azodye.

The nanoscale interactions between adjacent layers of layer-by-layer (LBL) films from poly(allylamine hydrochloride) (PAH) and azodye Brilliant Yellow (BY) have been investigated, with the films employed for optical storage and the formation of surface-relief gratings. Using Fourier transform infrared spectroscopy, we identified interactions involving SO3- groups from BY and NH+ groups from PAH. These electrostatic interactions were responsible for the slow kinetics of writing in the optical storage experiments, due to a tendency to hinder photoisomerization and the subsequent reorientation of

Overview of pulmonary alveolar macrophage renewal in normal rats and during different pathological processes.

We report experimental results on pulmonary alveolar macrophage (PAM) renewal in healthy rats and in rats treated with particles introduced in the lungs. Morphometric studies showed that the lungs of normal rats of the strain used in our study contain 20 x 10(6) PAM, 50 x 10(6) monocytes in alveolar capillaries, and about 3 x 10(5) interstitial macrophages. Pulse labeling with a tritiated thymidine (3HT) gave a labeling index of 0.4% for the monocytes, of which a few could be observed in mitosis within alveolar capillaries. These monocytes are likely to be the PAM precursors. The daily input (

Nanosized silicon nitride promotes vascularized osteogenesis through regulating the endothelial adherens junctions via the FRMD6/VE-cadherin pathway.

Effective vascularization is critical for successful bone regeneration. Herein, silicon nitride nanoparticles (Si3N4 NPs) were demonstrated to enhance early vascular regeneration and promote new bone formation in vivo. We found that Si3N4 NPs induced autophagy and stimulated angiogenesis after they were uptaken by human umbilical vein endothelial cells (HUVECs). Transcriptome sequencing revealed that the angiogenic effect of Si3N4 NPs was closely associated with modulation of adherens junctions (AJs). Specifically, Si3N4 NPs significantly increased phosphorylation of the core AJs protein VE-ca

Nanoencapsulation enhances stability, release behavior, and antimicrobial properties of Sage and Thyme essential oils.

The increasing demand for natural bioactive compounds in agriculture, food preservation, and pharmaceuticals has highlighted the need for effective delivery systems to enhance their stability and bioavailability. In this study, we address this challenge by developing and characterizing silica hollow nanospheres (HNSs) and hollow polymer nanocapsules (HPNs) for the encapsulation of essential oils (EOs), specifically those derived from Thyme (Thymus vulgaris) and Sage (Salvia officinalis). The HNSs were synthesized using tetraethyl orthosilicate (TEOS) via a sol-gel process, while urea-formaldeh

Highly Stable Silica-Coated Bismuth Nanoparticles Deliver Tumor Microenvironment-Responsive Prodrugs to Enhance Tumor-Specific Photoradiotherapy.

Radiosensitizers are agents capable of amplifying injury to tumor tissues by enhancing DNA damage and fortifying production of radical oxygen species (ROS). The use of such radiosensitizers in the clinic, however, remains limited by an insufficient ability to differentiate between cancer and normal cells and by the presence of a reversible glutathione system that can diminish the amount of ROS generated. Here, to address these limitations, we design an H2O2-responsive prodrug which can be premixed with lauric acid (melting point ∼43 °C) and loaded around the surface of silica-coated bismuth na

Antioxidant properties of probucol released from mesoporous silica.

Antioxidants play a vital role in scavenging reactive oxygen species (ROS) produced by the reduction of molecular oxygen from various cellular mechanisms. Under oxidative stress, an increase in the levels of ROS overwhelms the antioxidant response, causing oxidative damage to biological molecules, and leading to the development of various diseases. Drug compounds with potent antioxidant properties are typically poorly water soluble and highly hydrophobic. An extreme case is Probucol (PB), a potent antioxidant with reported water solubility of 5 ng/ml, and oral bioavailiability of <10%. In this
