We report a family with inherited macrothrombocytopenia and characteristic large membrane complexes in the platelets. Two affected subjects had platelet counts of 40 and 65 x 10(9)/L respectively as assessed by contrast phase microscopy. Ultrastructural studies revealed giant spheroid platelets with characteristic large membrane complexes and/or giant vacuoles containing platelet organelles. Immunohistochemical studies of actin and tubulin showed a disorganization of the microtubule and actin systems. These abnormalities were absent in leukocytes, indicating a platelet-specific cytoskeleton di…

The trabecular meshwork is a specialized tissue in the anterior chamber of the eye that regulates the aqueous humor outflow and controls the intraocular pressure. Cells in the trabecular meshwork are believed to be essential for maintenance of the outflow system, and their malfunctioning may lead to elevation of intraocular pressure and development of glaucoma. These cells are avid phagocytes. Using an in vitro tissue culture system, we have previously shown that bovine trabecular meshwork cells exhibited a short-term loss of cell-matrix adhesiveness after exposure to latex microspheres. We report the case of a young white man with Charcot-Marie-Tooth disease type 1 that began at 4 years. At 15 years, he developed proteinuria, arterial hypertension, and renal insufficiency. Renal biopsy specimens studied by electron microscopy showed deposition of nonamyloidotic microfibrils. This is the first report of fibrillary glomerulopathy associated with this neurological disorder.

Modern immunohistochemical methods allow a functional characterization of the human endolymphatic sac (ES) and its associated cell populations. The currently available immunohistochemical data of the extraosseous part of the human ES support the assumption that the epithelium is metabolically active and capable of both secretion and adsorption. The reactivity of the epithelial cells with antibodies against neuroendocrine antigens imply a paracrine activity of the human ES. Further results provide evidence for a possible role of the human ES in inner ear immune defence and indicate a putative f

Several observations indicate that endothelial cells interact with fibrinogen, fibrin, and their derivative products. The main biological function of fibrinogen on endothelial cells so far identified is its ability to promote adhesion, motility, and cytoskeletal organization of these cells. In other words, when fibrinogen is bound to a substratum, it behaves as a matrix protein like fibronectin, collagen, or vitronectin. Fibrinogen specifically binds to endothelial cells with low affinity and with a high number of binding sites. In addition, the putative fibrinogen receptor in endothelial cell

Dominant dystrophic epidermolysis bullosa of Cockayne-Touraine occurred in a father and his son. Clinically, the child presented evident blistering, while the father, despite of his work, where minimal skin traumata frequently occur, was rarely affected. Absent or rudimentary anchoring fibrils were seen in the normal-appearing skin of the father; this suggests that other factors may play a role in the dermolytic separation.

The ciliary epithelia of human fetal and adult eyes were investigated for the presence of intermediate filament proteins. Coexpression of cytokeratin and vimentin was found. This resembles the intermediate filament pattern of choroid plexus epithelium, another neuroepithelial tissue with secretory function.

After fixation of bone marrow with glutaraldehyde, tannic acid and saponin, a delicate network of microfibrils (10 nm) was observed in the extracellular space. Masses of microfibrils were most frequently observed between the endothelial cells of the sinusoidal wall and adventitial reticular cells, but were also observed at other sites throughout the marrow stroma. Microfibrils are an important component of the extracellular material of bone marrow and appear to provide an anchoring substrate for the endothelium.

Cytokeratins of normal epithelia and of some neoplasms of the female genital tract were studied by immunofluorescence microscopy of frozen sections and by two-dimensional gel electrophoresis of cytoskeletal proteins from microdissected tissues. All normal epithelia were stained with the monoclonal cytokeratin antibody KG 8.13 whereas certain monoclonal antibodies stained only simple epithelia. As revealed by gel electrophoresis the normal epithelia of the ovarian surface, oviduct, endometrium and endocervix contained cytokeratin polypeptides Nos. 7, 8, 18 and 19. In contrast, stratified exocer

Twelve biopsies from condylomatous lesions of the human cervix were examined by transmission electron microscopy in order to study koilocytotic cells. The ultrastructure of koilocytes, revealed mainly cytoplasmic changes in the perinuclear area. These changes first appeared in cells of the intermediate layers and extended to the surface of the epithelium. The cells involved showed small perinuclear areas, with a fine granular material, which appeared as a lysis of the cytoplasmic matrix, forming microvacuolelike structures that gradually enlarged and fused together, ending with the formation o
