We have previously reported that a monoclonal antibody-based antigen detection assay (AD12) is sensitive and specific for Bancroftian filariasis in Egypt. The purpose of the present study was to demonstrate the use of this assay in a sentinel population as a means of efficiently screening for filariasis endemicity. Antigen testing was performed with finger-prick blood collected during the day from 743 schoolchildren (ages 11-16 years). The school draws students from 5 villages in Qalubia Governorate, 35 km north-east of Cairo, Egypt. The prevalence of filarial antigenaemia in the school was 17

A nine month entomological and parasitological study was conducted between December 1991 and August 1992, at Kanchrapara, a town of the district North 24-Parganas, West Bengal. The town was found to be endemic for bancroftian filariasis. Microfilaria rate among the human population was 1.9 per cent (35 of 1200 persons). Average man hour density of C. quinquefasciatus was considerably high (31.10) and a higher vector prevalence was seen in summer. The infection and infectivity rates of the natural population of the vector were 2.3 and 0.28 per cent respectively. 9.2 and 1.7 per cent of the huma

A form of unexplained arthritis, not attributable to known causes, seen in children (0-14 yrs) in this endemic zone of Bancroftian filariasis was investigated for its association with filariasis. Nineteen cases of undiagnosed arthritis were screened for filarial IgG antibodies to Wuchereria by Stick Enzyme Linked Immunosorbent Assay (ELISA). All had large joint involvement, the commonest joint affected being the knee joint. Involvement was monoarticular in 10 and binarticular in 9. Joint pain was present in 18 and effusion in 12. Five patients had recurrent episodes. Sixteen (84.2%) showed fil

The risk of infection (RI) with Wuchereria bancrofti by vector mosquitoes has been conventionally estimated from the biting population of all age classes. This estimate of RI had no correlation with microfilaria (mf) prevalence and intensity of infection in humans. A new method for estimating RI from a resting population of Culex quinquefasciatus is proposed based on a longitudinal study in seventeen sites in Pondicherry, India. Unlike previous estimates, RI calculated by the present method considering the density of parous mosquitoes collected per man-hour and the mean number of infective sta

In October 1989, 58 apparently healthy Polynesian Wuchereria bancrofti carriers in whom microfilarial (mf) density was greater than or equal to 100 mf/ml were randomly allocated to treatment groups receiving single doses of either ivermectin at 100 mcg/kg or diethylcarbamazine (DEC) at 3 and 6 mg/kg. Six months later, half of the carriers initially treated with ivermectin 100 mcg/kg or DEC 3 mg/kg were given a second similar dose while the rest were given a placebo. By day 360 (6 months after retreatment), comparison of adjusted geometric mean mf counts per group indicated that (i) among the 3

The effects of a well-spaced diethylcarbamazine (DEC) mass drug application in areas highly endemic for Wuchereria bancrofti in Papua New Guinea are not known. In 1986 a semi-annual single-dose 6 mg/kg body weight administration of DEC was initiated in the Ok Tedi area of Western Province, Papua New Guinea. The rate of bancroftian filariasis in the area was 39%. Within two years the rate of detectable microfilaraemia was reduced from 31% to 11% in the treated group. The mean blood density of the parasite was reduced from 79 to 19 microfilariae per 20 microliters. A survey of untreated villages

A monoclonal antibody, Og4C3, directed against antigens of Onchocerca gibsoni (but not phosphorylcholine) has been used in a sandwich ELISA to detect a circulating antigen of Wuchereria bancrofti in human serum. The interfering effect of host antibody was reduced by first boiling one part of serum for 5 min in the presence of three parts of 0.1 M Na2EDTA, pH 4.0. A total of 119 sera from individuals and 8 pooled sera from clinically and/or parasitologically defined cases of filariasis, plus 8 individual and 1 pooled endemic control sera, all from the filariasis serum bank of the World Health O

The filariasis of W. bancrofti with nocturnal periodicity is, together with malaria, the principal endemic disease in the Comoros Islands. Well studied on Mayotte island, which remained a French Territorial Collectivity, its prevalence and distribution are less known in the Grand Comoros, Anjouan and Moheli islands which form the Federal Islamic Republic of the Comoros. The study of the nocturnal microfilariae in the Grand Comoros and Moheli in 1987 is the opportunity to assess the situation of the disease on this archipelago.

This study examines the state of mosquito-borne lymphatic filariasis in Madras, Tamil Nadu, in southern India during the 1970s and into the 1980s. In its various forms it remains a public health problem of considerable magnitude in many tropical countries and affects both rural and urban populations [1. World Health Organization (Third Report). Expert Committee on Filariasis. Tech. Rep. Ser. No. 542, p. 7, 1974]. Here problems of delimiting the spatial extent of morbidity in Madras are discussed and sample survey data are examined which suggest that the disease is prevalent throughout the city

Previous studies have demonstrated that the induction of immunoregulatory mechanisms in the spleens of Brugia pahangi-infected jirds is correlated with the onset of microfilaremia. This study investigated the relationship between production of a factor with IL-2-like activity and the regulation of T cell-mediated responses in jirds experimentally infected with B. pahangi. A factor present in culture supernatants of mitogen-stimulated jird lymphocytes supported the proliferation of murine CTLL cells and provided the basis for an IL-2 assay. Mitogen induced proliferative responses and IL-2 produ

Ingestion of Wuchereria bancrofti microfilariae (mf) from humans by 639 Anopheles gambiae, 557 An. arabiensis, 117 An. melas and 9 An. funestus was investigated. The mf densities in blood fell into 3 groups; 0-32mf/ml, 107-122 mf/ml and 421-1140 mf/ml. In An. gambiae and An. arabiensis percentage of mosquitoes ingesting mf was strongly associated with mf density in host blood; in An. melas the association was much weaker. Mean number of mf ingested per mosquito was also strongly correlated to mf density in An. gambiae and An. arabiensis but not in An. melas. At low mf densities both An. gambia

Patent infections with the lymphatic filariae, Wuchereria bancrofti and Brugia malayi, are associated with suppressed in vitro cellular responsiveness to filarial antigens. In studies of bancroftian filariasis in Haiti, a significant number of microfilaremic individuals can be characterized as "responders" to filarial antigens. Cells from 37/74 untreated microfilaremic subjects responded to B. pahangi antigen (stimulation ratio greater than 2) as detected by in vitro blastogenesis. A comparison of responders to nonresponders revealed a significant difference in mean B. pahangi reactivity (15,8

Infective larvae of subperiodic B. malayi from South Kalimantan (Borneo), Indonesia collected from laboratory-raised Ae. togoi mosquitoes after feeding on infected mongolian gerbils (Meriones unguiculatus) were inoculated subcutaneously into the groin areas of 15 SD and 36 LE rats. Blood was examined weekly by membrane filtration and thick smears starting 10 weeks post-infection. Microfilariae were found in 3 SD and 4 LE rats, the mf infection rate of 20% and 11% respectively. The prepatent period was significantly shorter in the SD rats (99-112 days) than those in the LE rats (110-153 days).

To be more definitive as a serodiagnostic method, an ideal immunoassay for detection of filaria-specific antibody and filarial antigens in body fluids should identify active filarial infection and differentiate between past and present infection. It should perform well in the areas of precision, reproducibility, parallelism, and sensitivity and should use reference and quality control reagents prepared with human body fluids that contain defined amounts of filarial antigen. Moreover, a sufficient quantity of reference serum in stable form should be made available to permit interlaboratory cros

In a study carried out in Jakarta, Indonesia between 1969--1977, patients with clinical manifestations of filariasis were examined for microfilaremia. Finger blood (3 X 20 microliter) were examined by conventional methods, venous blood by membrane filtration, urine and hydrocele fluid by concentration methods. A total of 1,422 patients were examined and 182 (12.8%) were positive by blood smear; however, examination by membrane filtration and concentration of urine and hydrocele fluid did not appreciably increase the association between clinical manifestations and Wuchereria bancrofti infection

Human filariasis and its vectors are little known in Ethiopia. Wuchereia bancrofti is confined to the western lowlands and cannot be the aetiological agent of elephantiasis in the highlands. Onchocerca volvulus is widespread in the south-western highlands. Loa loa and Dipetalonema perstans are reported here for the first time and appear to be uncommon. Studies were conducted in 1973, mostly near Jimma in Kaffa Province, on potential vectors of all but the first of these parasites. Among Simuliidae, the only blackflies of apparent medical importance were S. woodi ethiopiense and a possibly new …

The Death to Onchocerciasis and Lymphatic Filariasis (DOLF) project conducts clinical and translational research to accelerate the global elimination of lymphatic filariasis (LF) and onchocerciasis (river blindness). Research needs were identified during several meetings of experts and helped study design. Over the last 15 years the project has conducted mass drug administration (MDA) studies and clinical trials with outstanding research teams in 11 countries that have led to many important discoveries. For example, DOLF studies showed that annual MDA was as effective as semiannual MDA for LF
