Visible light induced changes in the immune response through an eye-brain mechanism (photoneuroimmunology).

The immune system is susceptible to a variety of stresses. Recent work in neuroimmunology has begun to define how mood alteration, stress, the seasons, and daily rhythms can have a profound effect on immune response through hormonal modifications. Central to these factors may be light through an eye-brain hormonal modulation. In adult primates, only visible light (400-700 nm) is received by the retina. This photic energy is then transduced and delivered to the visual cortex and by an alternative pathway to the suprachiasmatic nucleus (SCN). The SCN is a part of the hypothalamic region in the b

Rhythms of mood: Bidirectional regulation of molecular clocks and neurotransmission in affective disorders.

Mood disorders, primarily major depressive and bipolar disorder, are characterized by significant neurochemical dysregulation and disturbances in biological rhythms. Recent evidence suggests for a complex, bidirectional interplay between the molecular circadian clock and major neurotransmitter systems, including monoamines (dopamine, serotonin, and norepinephrine), glutamate and GABA. While the suprachiasmatic nucleus acts as the master pacemaker, local clocks in limbic regions-such as the prefrontal cortex, hippocampus, ventral tegmental area and nucleus accumbens-maintain semi-autonomous osc

Modified atmosphere packaging delays enzymatic browning and extends the shelf life of fresh-cut lotus root by maintaining membrane lipid homeostasis.

Fresh-cut lotus root (FCLR) is prone to rapid browning and microbial growth after processing. Although modified atmosphere packaging (MAP) can inhibit browning in FCLR, the optimal gas composition and the physiological basis of browning inhibition remain unclear. In this study, a principal component analysis-assisted multi-index evaluation was employed to identify the optimal gas composition (MAP1: 1% O2, 15% CO2, 84% N2), followed by analyses of membrane lipid metabolism and untargeted metabolomics. After 15 d, MAP1 reduced browning by 72% and lowered mold/yeast counts and total aerobic count

Abusive use of Zolpidem as a Result of COVID-19 and Perspectives of Continuity of the Problem in the Post-Pandemic Period.

Zolpidem is a non-benzodiazepine hypnotic drug that works as a positive modulator of Gamma-Amino Butyric Acid-A (GABA-A) receptors, with high selectivity for α1 subunits. Given this selective binding, the drug has a strong hypnotic activity. Social isolation during the SARS-CoV-2 pandemic has contributed to increased rates of anxiety, depression, and insomnia. As a result, studies have pointed to a possible increase in the indiscriminate use of drugs with sedative effects, such as Zolpidem, during the pandemic. The aim of this work was to present prospective evidence that warns of the possibil

Deciphering pain: molecular mechanisms and neurochemical pathways-challenges and future opportunities.

This review delves into the intricate biological underpinnings of pain perception. It encompasses nociceptive signaling pathways, the molecular mechanisms involved, and the subjective experience of discomfort in humans. The initial focus is on nociceptor transduction, where specialized neurons transform noxious stimuli into electrical impulses. Subsequently, the review explores the central nervous system, elucidating how these signals are processed and modulated by critical elements such as ion channels, receptors, and neurotransmitters (e.g., substance P, glutamate, GABA). Shifting gears towa

Neurometabolic and Brain Functional Alterations Associated with Cognitive Impairment in Patients with Myasthenia Gravis: A Combined 1H-MRS and fMRI Study.

Whether patients with myasthenia gravis (MG) exhibit cognitive impairment is controversial. Also the underlying mechanisms are unknown. We aimed to investigate alterations in cognitive function, neurometabolite levels, and brain function in patients with MG and to explore the associations between abnormal regional brain functional activity, neurometabolite concentrations in the MPFC and left thalamus, and cognitive activity in patients with MG. Neuropsychological tests, proton magnetic resonance spectroscopy, and resting-state functional magnetic resonance imaging were performed on 41 patients

Histone methyltransferase Ezh2 coordinates mammalian axon regeneration via regulation of key regenerative pathways.

Current treatments for neurodegenerative diseases and neural injuries face major challenges, primarily due to the diminished regenerative capacity of neurons in the mammalian CNS as they mature. Here, we investigated the role of Ezh2, a histone methyltransferase, in regulating mammalian axon regeneration. We found that Ezh2 declined in the mouse nervous system during maturation but was upregulated in adult dorsal root ganglion neurons following peripheral nerve injury to facilitate spontaneous axon regeneration. In addition, overexpression of Ezh2 in retinal ganglion cells in the CNS promoted

Monobutyrin and monovalerin improve gut-blood-brain biomarkers and alter gut microbiota composition in high-fat fed apolipoprotein-E-knockout rats.

Monobutyrin (MB) and monovalerin (MV), glycerol esters of short-chain fatty acids (SCFAs), have been shown to positively influence lipid profile and biomarkers in the gut and brain. This study examined whether MB and MV in high-fat diets, affected microbiota composition and gut-blood-brain markers in apolipoprotein E deficient (ApoE-/-) rats, a model for studies of lipid-associated disorders, and neurodegenerative processes in Alzheimer's disease (AD). ApoE-/- rats fed MB and MV increased Tenericutes and the brain neurotransmitter γ-aminobutyric acid (GABA), while the blood stress hormone cort

Modulatory effects of cannabinoids on brain neurotransmission.

Recreational and chronic cannabis use has been associated with a range of acute and chronic effects including; anti-nociceptive actions, anxiety, depression, psychotic symptoms and neurocognitive impairments. The mechanisms underlying cannabinoid-based drugs effects are not fully known but given the neuro-modulatory functions of the endocannabinoid system, it seems likely that agonistic activity at the cannabinoid type-1 receptors (CB1 ) might modulate the functions of other neurotransmitter systems. The present review has summarized the currently available pre-clinical and clinical data on th

The Role of Neuropeptide Y (NPY) in Alcohol and Drug Abuse Disorders.

Neuropeptide Y (NPY) is a neuromodulator that is widely expressed throughout the central nervous system (CNS) and which is cosecreted with classic neurotransmitters including GABA and glutamate. There is a long history of research implicating a role for NPY in modulating neurobiological responses to alcohol (ethanol) as well as other drugs of abuse. Both ethanol exposure and withdrawal from chronic ethanol have been shown to produce changes in NPY and NPY receptor protein levels and mRNA expression in the CNS. Importantly, manipulations of NPY Y1 and Y2 receptor signaling have been shown to al

Zinc-containing yeast extract promotes nonrapid eye movement sleep in mice.

Zinc is an essential trace element for humans and animals, being located, among other places, in the synaptic vesicles of cortical glutamatergic neurons and hippocampal mossy fibers in the brain. Extracellular zinc has the potential to interact with and modulate many different synaptic targets, including glutamate and GABA receptors. Because of the central role of these neurotransmitters in brain activity, we examined in this study the sleep-promoting activity of zinc by monitoring locomotor activity and electroencephalogram after its administration to mice. Zinc-containing yeast extract (40 a

Modulation of firing activity by endogenous GABAA receptors in the globus pallidus of MPTP-treated parkinsonian mice.

The globus pallidus in rodents, equivalent to the external segment of the globus pallidus in primates, plays an important role in movement regulation. Previous studies have shown abundant γ-aminobutyric acid (GABA)ergic innervation and GABAA receptors in the globus pallidus. In this study, we investigated the effects of endogenous GABAA receptors on the spontaneous firing activity of pallidal neurons in both normal and MPTP-treated mice using multi-barrel electrodes extracellular recordings in vivo. We found that in normal mice, pressure ejection of 0.1 mmol/L gabazine, a specific GABAA recept

[Premenstrual syndrome].

The Premenstrual Syndrome (PMS) is defined as the repeated occurrence of psychic disability accompanied by physical symptoms such as weight-gain, mastodynia and edema during the luteal phase. Irritability, depressive mood, lack of concentration and anxiety are the dominant psychiatric features. These symptoms culminate during the premenstrual period and disappear at the onset of menstrual bleeding. The symptomatology is of variable degree. About 2 to 3% of all women of reproductive age are severely impaired by these symptoms. The etiology of PMS is unknown. Disturbance of serotonin metabolism

Central neural control of the lower urinary tract.

The lower urinary tract has two main functions, the storage and periodic elimination of urine, which are regulated by a complex neural control system in the brain and spinal cord. This neural system exhibits switch-like patterns of activity that are generated by visceral reflex circuits, some of which are under voluntary control. Experimental studies in animals indicate that the micturition reflex is mediated by a spinobulbospinal pathway passing through a coordination centre (the pontine micturition centre) located in the rostral brainstem. This reflex pathway is, in turn, modulated by higher

Integrating network medicine and foundation models reveals cell-type-specific regulatory alterations in Alzheimer's disease.

Alzheimer's disease is a complex neurodegenerative disorder characterized by progressive cognitive decline and neuroinflammation. Although its molecular hallmarks are well documented, cell-type-specific mechanisms driving gene dysregulation remain elusive. While single-cell RNA sequencing resolves cellular states, most studies focus on individual genes rather than coordinated programs. Moreover, a gap persists between interpretable network-based models and artificial intelligence foundation models, which capture complex interactions but lack mechanistic transparency. Whether these approaches c

Advances in the Application of Amino Acids in the Preservation of Postharvest Fruits and Vegetables.

Postharvest loss of fruits and vegetables remains a significant global challenge, while concerns over chemical residues, energy consumption, and storage-induced physiological disorders have accelerated the search for safe and eco-friendly preservation strategies. As key plant metabolites and signaling molecules, amino acids are gaining more attention for their roles in postharvest biology. This article reviews the primary postharvest issues of fruits and vegetables, the functional roles of representative amino acids, including γ-aminobutyric acid (GABA), glutamate (Glu), arginine (Arg), cystei

Role of the tail of the ventral tegmental area in the regulation of maternal behaviour: a pharmacological study in postpartum rats.

Maternal behaviour is characterized by increased motivation for pups, which facilitates the expression of behaviours promoting offspring survival and development. Motivated behaviours are critically dependent on brain dopamine systems, thus, understanding its functioning is necessary to elucidate maternal reward brain processing. The activity of dopamine systems is controlled by the inhibitory GABA cells of the tail of the ventral tegmental area or rostromedial tegmental nucleus (tVTA/RMTg). This brain region is involved in reward and avoidance behaviour, among other functions, however, its ro

The neurobiological foundation of effective repetitive transcranial magnetic brain stimulation in Alzheimer's disease.

Noninvasive brain stimulation (NIBS) techniques, such as repetitive transcranial magnetic stimulation (rTMS), are promising candidate therapeutics for Alzheimer's disease (AD). We review the evidence supporting the fundamental mechanisms of action of rTMS treatments in AD. rTMS exerts profound effects at different neurobiological and systems neurophysiological levels. By engaging distinct pre- and postsynaptic structures within the stimulated neural network, it directly or indirectly influences various cellular and molecular components. In AD, rTMS influences synaptic plasticity, inducing last

Sleep-promoting effects and mechanisms of Bacillus coagulans IDCC 1201: evidence from pentobarbital-induced sleep and electroencephalography analysis in mice.

Recent studies have explored the potential of probiotics to modulate sleep, particularly via neurotransmitter pathways. Bacillus coagulans IDCC 1201 is a high γ-aminobutyric acid (GABA)-producing strain, second only to Lactobacillus johnsonii IDCC 9203. This study investigated the sleep-promoting efficacy of B. coagulans IDCC 1201 using a pentobarbital-induced sleep model and electroencephalography (EEG)-based sleep architecture analysis. In the pentobarbital-induced sleep test, B. coagulans IDCC 1201 at ≥1 × 108 CFU per day significantly reduced sleep latency and increased sleep duration, sho
