Parasitic nematodes pose a significant threat to human and animal health, as well as cause economic losses in the agricultural sector. The use of anthelmintic drugs, such as Ivermectin (IVM), to control these parasites has led to widespread drug resistance. Identifying genetic markers of resistance in parasitic nematodes can be challenging, but the free-living nematode Caenorhabditis elegans provides a suitable model. In this study, we aimed to analyze the transcriptomes of adult C. elegans worms of the N2 strain exposed to the anthelmintic drug Ivermectin (IVM), and compare them to those of t

BACKGROUND: Lyme disease is a zoonotic disease caused by infection with Borrelia burgdorferi (Bb), the involvement of the nervous system in Lyme disease is usually referred to as Lyme neuroborreliosis (LNB). LNB has diverse clinical manifestations, most commonly including meningitis, Bell's palsy, and encephalitis. However, the molecular pathogenesis of neuroborreliosis is still poorly understood. Comprehensive transcriptomic analysis following Bb infection could provide new insights into the pathogenesis of LNB and may identify novel biomarkers or therapeutic targets for LNB diagnosis and tre

The amino acid arginine (Arg) is a recognized secretagogue of growth hormone (GH), and has been shown to induce GH gene expression. Arg is the natural precursor of nitric oxide (NO), which is known to mediate many of the effects of Arg, such as GH secretion. Arg was also shown to increase calcium influx in pituitary cells, which might contribute to its effects on GH secretion. Although the mechanisms involved in the effects of Arg on GH secretion are well established, little is known about them regarding the control of GH gene expression. We investigated whether the NO pathway and/or calcium a

Colorectal cancer (CRC) is one of the most lethal malignancies worldwide, and the precise identification of biomarkers from colonic adenoma to cancer is of great significance for preventing the development of adenocarcinoma. Given that existing methods inadequately capture the topological network relationships among genes, this study proposes a graph neural network model based on multifeature learning, named ChebTs, to investigate the correlation between key genes involved in the colorectal "adenoma-cancer" transition. The GSE41657 and GSE31905 datasets from the GEO database were stratified in

dcAdoMet (decarboxylated S-adenosylmethionine) is an essential intermediate in the synthesis of polyamines. Its content is normally very low, amounting to less than 5% of that of S-adenosylmethionine itself. It was found that in mice lacking spermine synthase there was a large increase in dcAdoMet and that overexpression of spermine synthase reduced the amount of this nucleoside. There was also an increase in dcAdoMet in cells derived from patients with Snyder-Robinson syndrome, a rare X-linked recessive human disease caused by SMS gene mutations that greatly reduce the content of spermine syn

In the discussion article the basic rules of human microelementosis theory (A. P. Avtsyn et al., 1991) and the review of modern data devoted to molecular-genetic mechanisms of iron homeostasis are represented. The conception of iron microelementosis (siderosis) with appropriate nomenclature and classification has been formulated. The main types of siderosis have been singled out as well hyposiderosis (a deficiency of iron), hypersiderosis (redundant accumulation of iron) and dissiderosis (an imbalance, redistribution of iron between functional and storage pools). In addition each type of sider

We described earlier the isolation of lfl25 mutation reducing the frequency of lysogenization of mutant cells by phage lambda (LycA phenotype). The mutation is mapped at present in the rpoC gene coding for the beta' subunit of bacterial RNA polymerase and named rpoC90. It is dominant and causes decrease in pools of some branched amino acids, pyrimidines and thiamine (vitamin B1) in mutant cells. Combination of rpoC90 mutation with some Rif-r mutations strengthens both the LycA phenotype and deficiency in metabolites mentioned above. Such combination was shown to also cause a defect in antiterm

BACKGROUND: Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by deficits in communication, social interaction, and behavioral regulation. Its etiology arises from a combination of genetic vulnerabilities and environmental influences. Bisphenol A (BPA) is an endocrine-disrupting chemical found in plastic-containing materials, including micro- and nanoplastic pollutants. Recent studies have shown that prenatal BPA exposure can alter behavior and the expression of genes related to autism and neurodevelopment.
METHODS: This study integrated and reanalyzed published RN

BACKGROUND: The c-Myc transcription factor is a master regulator and integrates cell proliferation, cell growth and metabolism through activating thousands of target genes. Our identification of direct c-Myc target genes by chromatin immunoprecipitation (ChIP) coupled with pair-end ditag sequencing analysis (ChIP-PET) revealed that nucleotide metabolic genes are enriched among c-Myc targets, but the role of Myc in regulating nucleotide metabolic genes has not been comprehensively delineated.

In swine and other livestock, the uterine endometrium exhibits dramatic morphological and secretory changes throughout the oestrous cycle and during pregnancy. Such physiological changes are a reflection of extremely complex interactions between gene products (RNA and protein). The recent development of genomics and proteomics methods, as well as associated bioinformatics tools, has provided the means to begin characterising such interactions. Indeed, the analysis of the transcriptome and proteome of cells and tissues now comprises a new field of study known as 'systems biology'. Currently, th

BACKGROUND: To date, variants in the GBA gene represent the most frequent large-effect genetic factor associated with Parkinson's disease (PD). However, the reason why individuals with the same GBA variant may or may not develop neurodegeneration and PD is still unclear.
OBJECTIVES: Therefore, we evaluated the contribution of rare variants in genes responsible for lysosomal storage disorders (LSDs) to GBA-PD risk, comparing the burden of deleterious variants in LSD genes in PD patients versus asymptomatic subjects, all carriers of deleterious variants in GBA.
METHODS: We used a custom next-g

BACKGROUND: Asthenozoospermia (AZS) is a leading cause of male-factor infertility, yet the contribution of the urinary microbiota to sperm motility remains poorly defined, because previous work has focused almost exclusively on the seminal and gut microbiota. Here we used 16S rRNA gene sequencing to characterize the composition and functional profiles of the urinary microbiota in men with AZS and in fertile controls.
RESULTS: Isolated AZS and age-matched fertile controls were enrolled, and urinary specimens from participants were sequenced successfully. Alpha diversity of the urinary microbio

Progressive retinal neurodegeneration remains a major therapeutic challenge in ophthalmology because structural and functional loss may continue despite control of the primary insult, including intraocular pressure reduction in glaucoma, metabolic management in diabetic retinopathy, or suppression of acute inflammatory activity. Ferroptosis, an iron-dependent form of regulated cell death driven by phospholipid peroxidation and insufficient antioxidant buffering, has emerged as a potential mechanism linking metabolic stress, ischemic injury, excitotoxicity, and neuroinflammatory signaling to su

Melatonin is a naturally occurring compound that regulates many aspects of plant growth and development. Recently, its stress-protective potential has been extensively studied. This study investigates the effect of exogenous melatonin on non-enzymatic antioxidants, gene expression, and activity of key antioxidant enzymes in young winter wheat plants subjected to 5 days of drought. Melatonin was root-supplemented 24 h before or after the stress. The parameters were analyzed in the leaves of two Bulgarian cultivars at the end of drought, and after recovery. Drought activated both enzymatic and n

Spinal muscular atrophy (SMA) is an oft-fatal infantile-onset neuromuscular disease caused by low SMN protein. Administration of SMN-inducing agents to SMA newborns prevents early mortality, but therapeutic outcomes vary considerably, and disease mechanisms remain poorly understood. Genetic modifiers can provide clues to disease mechanisms and serve as targets for novel treatments. Here, we describe how one such modifier suppresses SMA in model mice. We show that the modifier, an Hspa8 G470R synaptic chaperone variant we previously identified, functions beyond an already defined role as an SMN

Acetate is a major metabolic end-product of Haemophilus influenzae and is produced via the phosphotransacetylase-acetate kinase (Pta-AckA) pathway, which also generates adenosine triphosphate (ATP). Despite its importance, the contribution of this pathway to H. influenzae physiology and virulence remains poorly understood. Here, we have investigated the individual roles of AckA and Pta for these processes by generating nonpolar single-gene knockouts in a chronic obstructive pulmonary disease (COPD)-isolate strain (Hi2019) and characterized their metabolic and infection phenotypes. Both mutants
