Changes in plasma gastrin, pancreatic polypeptide and pulse rate were examined in three adult female pigs in which venous cannulae had been placed and in two of which arterial cannulae allowed sampling of arterial blood or recording of pulse rate. Gastrin, pancreatic polypeptide and pulse rate increased when the pigs ate after fasting overnight. Pancreatic polypeptide was reduced to or below resting levels following administration of atropine (25, 50 or 100 micrograms/kg intravenously) after which gastrin remained at about its previous levels or rose and pulse rates rose. A case of fatal gas gangrene occurring after venous catheterization and the administration of sympathomimetic amines is reported. We administered [125I]thyroxine intravenously to adult male rats and measured uptake and subcellular distribution of the hormone and its metabolites in brain. A constant plasma drug concentration can be achieved and maintained by the intravenous administration of an initial bolus loading dose in conjunction with a constant rate and an exponential intravenous drug infusion. Studies on the in vivo degradation of 14C-labelled nitroprusside (NP) were conducted in rats and indicated that the primary product was cyanide and not thiocyanate. The concentrations of doxycycline were measured in 36 enucleated, enlarged prostate glands. After administration of 200 mg of doxycycline intravenously significant tissue concentrations (mean value 1.79 mug/g) were rapidly obtained. The mean concentration of the drug 14 hours after a repeated oral dose of 100 mg was 1.56 mug/g. The ratios of tissue and serum concentrations were 0.610 and 0.568, respectively. As there are only a few antibiotics which penetrate into the prostate, the importance of these data for the treatment of bacterial or mycoplasma prostatitis is obvious.
