The molecular and biochemical insight view of lycopene in ameliorating tramadol-induced liver toxicity in a rat model: implication of oxidative stress, apoptosis, and MAPK signaling pathways.

The influence of tramadol (TD) on hepatic tissue and the potential efficiency of lycopene to mitigate TD-induced hepatotoxic impacts were determined. Forty male albino rats were allocated into four groups: group I, untreated (placebo); group II, injected with TD (15 mg kg-1) intraperitoneally (i.p.); group III, gastrogavaged with lycopene (10 mg kg-1) per os (p.o.); and group IV received TD with lycopene with the same mentioned doses for 15 days. The results demonstrated that TD induced augmentation in tissue lipid peroxidation biomarker and disturbance in the antioxidant homeostasis and eleva

Characterization of locomotor response to psychostimulants in the parthenogenetic marbled crayfish (Procambarus fallax forma virginalis): A promising model for studying the neural and molecular mechanisms of drug addiction.

Although scientific research using mammalian models has made great strides in uncovering the enigmatic neural and molecular mechanisms orchestrating the state of drug addiction, a complete understanding has thus far eluded researchers. The complexity of the task has led to the use of invertebrate model systems to complement the research of drug-induced reward in mammalian systems. Invertebrates, such as crayfish, offer excellent model systems to help reveal the underlying mechanisms of drug addiction as they retain the ancestral neural reward circuit that is evolutionarily conserved across tax

Idiosyncratic recognition of UUG/UUA codons by modified nucleoside 5-taurinomethyluridine, τm5U present at 'wobble' position in anticodon loop of tRNALeu: A molecular modeling approach.

Lack of naturally occurring modified nucleoside 5-taurinomethyluridine (τm5U) at the 'wobble' 34th position in tRNALeu causes mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes (MELAS). The τm5U34 specifically recognizes UUG and UUA codons. Structural consequences of τm5U34 to read cognate codons have not been studied so far in detail at the atomic level. Hence, 50ns multiple molecular dynamics (MD) simulations of various anticodon stem loop (ASL) models of tRNALeu in presence and absence of τm5U34 along with UUG and UUA codons were performed to explore the dynami

Understanding the Dual Inhibition of COX-2 and Carbonic Anhydrase-II by Celecoxib and CG100649 Using Density Functional Theory Calculations and other Molecular Modelling Approaches.

Recent developments in the dual inhibition studies of cyclooxygenase-2 (COX-2) and carbonic anhydrase (CA-II) imply a promising platform for the development of new generations of nonsteroidal anti-inflammatory drugs (NSAIDs). CG100649 is such a molecule that got recently approved by Korean Ministry of Food and Drug safety (MFDS) and is being marketed by the name polmacoxib for the treatment of osteoarthritis. CG100649 significantly inhibits CA-II in blood and COX-2 in inflammatory tissues. However, the mechanism of CG100649 dual inhibition of COX-2/CA-II is not well understood. In this study,

High molecular weight hydroxyethyl starch solutions are not more effective than a low wmolecular weight hydroxyethyl starch solution in a porcine model of septic shock.

BACKGROUND: There is evidence that suggests that early fluid resuscitation is beneficial in the treatment of sepsis. We previously demonstrated that hydroxyethyl starch (HES) 130/0.42 attenuated capillary leakage better than HES 200/0.5. Using a similar porcine fecal sepsis model, we tested the effects of two new synthetic high molecular weight (700 kDa) hydroxyethyl starches with the same molar substitution of 0.42 but with a different C2/C6 ratio compared to 6% HES 130/0.42 on plasma volume (PV), systemic and tissue oxygenation. METHODS: This was a prospective, randomized, controlled animal

Guinea pig skin, a model for epidermal cellular and molecular changes induced by UVR in vivo and in vitro: effects on Mycobacterium bovis Bacillus Calmette-Guérin vaccination.

Previously, we reported that ultraviolet B-radiation (UVR) suppressed Bacillus Calmette-Guérin (BCG) vaccine-induced resistance to Mycobacterium tuberculosis in guinea pigs (GP). Herein, we investigated the cellular and molecular changes within the irradiated GP epidermis and the in vivo effect of supernatants from UV-irradiated (200 J m(-2)) epidermal cells (UV-sup) on M. bovis BCG vaccination. UVR increased the number of nucleated keratinocytes in the skin, but caused a decrease in the proportions of CD25(+)T cells. In the spleen, UVR resulted in a decrease in the proportions of T-cell subse

One-pot Synthesis, Molecular Modeling and In Vitro Antibacterial Activity of Novel 3-(1,3,4-Oxadiazol-2-yl) Quinoxaline 1,4-Dioxide and Pyrazolyl Analogs.

The new 1,3,4-oxadiazolylquinoxaline ring system was synthesized by the condensation of the quinoxaline 1,4-dioxide acid hydrazide 6 with acetyl chloride in one-pot synthesis. In which the hydrazide was refluxed in excess of acetyl chloride, acetic anhydride or acetic acid in the presence of phosphorus oxychloride forming the 1,3,4-oxadiazolylquinoxaline ring system. Molecular modeling studies have been performed to evaluate their recognition at the hDHFR binding-pocket as potential hDHFR inhibitors. The antibacterial properties of these compounds showed reasonable activities towards gram nega

Reactions of deprotonated tyrosine and tryptophan with electronically excited singlet molecular oxygen (a1Δ(g)): a guided-ion-beam scattering, statistical modeling, and trajectory study.

The reactions of deprotonated tyrosine ([Tyr-H](-)) and tryptophan ([Trp-H](-)) with the lowest electronically excited state of molecular oxygen O(2)[a(1)Δ(g)] have been studied in the gas phase, including the measurement of the effects of collision energy (E(col)) on reaction cross sections over a center-of-mass E(col) range from 0.05 to 1.0 eV. [Tyr-H](-) and [Trp-H](-) were generated using electrospray ionization, and both have a pure carboxylate anion structure in the gas phase. Density functional theory calculations and RRKM modeling were used to examine properties of various complexes, t

Molecular approaches for structural characterization of a new potassium channel blocker from Tityus stigmurus venom: cDNA cloning, homology modeling, dynamic simulations and docking.

Potassium channels are involved in the maintenance of resting membrane potential, control of cardiac and neuronal excitability, neurotransmitters release, muscle contractility and hormone secretion. The Tityus stigmurus scorpion is widely distributed in Northeastern Brazil and known to cause severe human envenomations, inducing pain, hypoesthesia, edema, erythema, paresthesia, headaches and vomiting. Most potassium channel blocking peptides that have been purified from scorpion venoms contain 30-40 amino acids with three or four disulfide bridges. These peptides belong to α-KTx subfamily. On t

Nucleophilic substitution reactions at liquid/liquid interfaces: molecular dynamics simulation of a model S(N)1 dissociation reaction at the water/carbon tetrachloride interface.

The ionic dissociation step of the nucleophilic substitution reaction t-BuCl --> t-Bu(+) + Cl(-) is studied at the water/carbon tetrachloride interface using molecular dynamics computer simulations. The empirical valence bond approach is used to couple two diabatic states, covalent and ionic, in the electronically adiabatic limit. The umbrella sampling technique is used to calculate the potential of mean force along the reaction coordinate (defined as the t-Bu to Cl distance) at several interface regions of varying distances from the Gibbs dividing surface. We find a significant increase of th

Morphological characteristics of total joint arthroplasty-derived ultra-high molecular weight polyethylene (UHMWPE) wear debris that provoke inflammation in a murine model of inflammation.

It is recognized that the chronic inflammation in peri-prosthetic tissue that contributes to implant failure frequently is provoked by the presence of wear debris. Some wear debris is inevitable because of the nature of the prosthesis, but not all patients develop severe inflammatory responses. The precise factors that mediate the severity of tissue inflammation to wear debris has yet to be fully defined. Because wear debris retrieved from peri-prosthetic tissue consists of a heterogeneous mixture of materials with various sizes and shapes, this study evaluated the influence of two major physi

Ameliorate impacts of scopoletin against vancomycin-induced intoxication in rat model through modulation of Keap1-Nrf2/HO-1 and IκBα-P65 NF-κB/P38 MAPK signaling pathways: Molecular study, molecular docking evidence and network pharmacology analysis.

Nephrotoxicity is an indication for the damage of kidney-specific detoxification and excretion mechanisms by exogenous or endogenous toxicants. Exposure to vancomycin predominantly results in renal damage and losing the control of body homeostasis. Vancomycin-treated rats (200 mg/kg/once daily, for seven consecutive days, i.p.) revealed significant increase in serum pivotal kidney function, oxidative stress, and inflammatory biomarkers. Histologically, vancomycin showed diffuse acute tubular necrosis, denudation of epithelium and infiltration of inflammatory cells in the lining tubular epithel

Design, synthesis, and molecular modeling of quinoline-linked diphenylpyrazoles as multitarget EGFR, VEGFR-2, and COX-2 inhibitors with anticancer, anti-inflammatory and apoptosis-inducing properties.

The development of multitarget-directed ligands (MTDLs) has emerged as a promising strategy for simultaneously modulating oncogenic, angiogenic, and inflammatory pathways implicated in cancer progression. In this study, a new series of quinoline-diphenylpyrazole hybrids were rationally designed, synthesized, and biologically evaluated as potential EGFR/VEGFR-2/COX-2 inhibitors. The synthesized compounds exhibited variable in vitro cytotoxic effect, with compounds 7f and 7k identified as the most potent derivatives. Compound 7f demonstrated IC50 values of 9.63, 12.17, 7.06, and 8.53 μM, whereas

Multi-omics integration of molecular genetics, cytogenetics and immunophenotyping: a novel prognostic model for immune landscape characterization and outcome prediction in Chinese patients with acute myeloid leukemia.

BACKGROUND: Current genetic risk stratification systems for acute myeloid leukemia (AML), including the 2017 European Leukemia Network (ELN), 2022 ELN, and 2023 China (CN) stratifications, inconsistently categorize key genetic variants such as CEBPA and FLT3-ITD. These systems also demonstrate limited applicability to Chinese patients, complicating clinical decision-making. This study optimized genetic stratification for Chinese AML patients and developed a multi-omics prognostic model by integrating immunophenotypic and clinical characteristics to enhance outcome prediction. METHODS: We retr

Compound Augmentation of Myocardial Injury in a Rat Model of Coronary Heart Disease Induced by Ischemia/Reperfusion, Rheumatoid Arthritis, and High-Fat Diet: A Molecular Mechanistic Study.

AIMS: Coronary heart disease (CHD) associated with rheumatoid arthritis (RA) is a primary driver of mortality in RA patients. In this study, we sought to establish a combined rat model of CHD and RA by integrating cardiac ischemia/reperfusion (I/R), high-fat diet (HFD), and intradermal administration of bovine type II collagen emulsified in complete Freund's adjuvant. The aim of constructing this model is to investigate and analyze the pathogenesis of RA-induced CHD under the modulation of HFD and cardiac I/R exposure. METHODS AND RESULTS: Sixty-four male Sprague-Dawley rats were randomly cat

Tyrosine phosphorylation and the inhibitory C-terminal SAM domain moderately affect transient interactions in a EphA2 cytoplasmic fragment in solution: A combined experimental and molecular modeling study.

UNLABELLED: Eph receptors, the largest subfamily of single-pass transmembrane receptor tyrosine kinases, play essential roles in development, including axon guidance and cell positioning, and in adult functions such as synaptogenesis. While their canonical signaling is typically repulsive and ligand-dependent, non-canonical activity promotes cell migration and proliferation in several cancers through ligand-independent mechanisms. Proteases associated with neurodegeneration can cleave Eph receptors, generating near full-length intracellular region (ICR) fragments, whose signaling potential rem

Label-Free Proteomic Profiling of the dvls2 (CL2006) Caenorhabditis elegans Alzheimer's Disease (AD) Model Reveals Conserved Molecular Signatures Shared With the Human AD Brain.

Alzheimer's disease (AD) is the most common form of dementia, posing significant challenges to cognitive, emotional, social, and financial well-being. The biochemical and molecular pathways associated with AD are complex, making it difficult to study and simulate in patients or through in vitro research. Thus, animal models play a crucial role in investigating the development and progression of AD. One widely used model in neuroscience studies is the free-living nematode Caenorhabditis elegans (C. elegans). The development of transgenic animals has allowed for the construction of the dvls2 (CL

Prediction of interactions between the SARS-CoV-2 ORF3a protein and small-molecule ligands using the ANDSystem cognitive platform, graph neural networks, and molecular modeling.

In recent years, artificial intelligence methods based on the analysis of heterogeneous graphs of biomedical networks have become widely used for predicting molecular interactions. In particular, graph neural networks (GNNs) effectively identify missing edges in gene networks - such as protein-protein interaction, gene-disease, drug-target, and other networks - thereby enabling the prediction of new biological relationships. To reconstruct gene networks, cognitive systems for automatic text mining of scientific publications and databases are often employed. One such AI-driven platform, ANDSyst
