Expression patterns of promoters for NPY Y(1) and Y(5) receptors in Y(5)RitTA and Y(1)RVenus BAC-transgenic mice.

In the rat brain, neuropeptide Y (NPY) Y(1) and Y(5) receptors are coexpressed in various forebrain regions where they mediate several NPY-activated functions, including feeding behaviour, anxiety, neuronal excitability and hormone secretion. We studied the distribution pattern and cellular colocalization of the Y(1) and the Y(5) receptor gene expression in the mouse brain by using transgenic mice with genomically integrated BAC clones, where the coding regions of the Y(1) and Y(5) receptor genes were replaced by Venus and the synthetic transcription factor itTA reporter genes, respectively (T

Loss of Brca2 and p53 synergistically promotes genomic instability and deregulation of T-cell apoptosis.

BRCA2 is a breast cancer susceptibility gene of which the product is thought to be involved in monitoring genome integrity and cell cycle progression. Brca2-null mice have a defect in embryonic cellular proliferation and die in utero. Here we report the generation of T-cell lineage-specific Brca2-deficient (tBrca2(-/-)) mice using the Cre-loxP system. Mice with a flanked by loxP allele of Brca2 were crossed to transgenic mice bearing Cre recombinase driven by the T cell-specific promoter Lck. Thymic cellularity and distribution of subset populations were normal in tBrca2(-/-) mutants. Thymocyt

Molecular analysis of T-cell receptor transcripts in a human T-cell leukemia bearing a t(1;14) and an inv(7); cell surface expression of a TCR-beta chain in the absence of alpha chain.

The polymerase chain reaction (PCR) was used to amplify cDNA in order to characterize normal and hybrid T-cell receptor (TCR) gene rearrangements derived from a T-cell acute lymphoblastic leukemia (T-ALL) bearing a chromosome 7 inversion. The nucleotide sequence analysis of the amplified product showed the presence of an out-of-frame V beta/J gamma/C gamma transcript and an in-frame V gamma/J gamma/C beta transcript which result from an interlocus recombination between the TCR-beta and gamma loci and the transcription of the reciprocal hybrid TCR gene. The sequence analysis of the reciprocal D

Human common acute lymphoblastic leukemia-derived cell lines are competent to recombine their T-cell receptor delta/alpha regions along a hierarchically ordered pathway.

Rearrangements of the T-cell receptor (TCR) delta locus are observed in the majority of human B-cell precursor acute lymphoblastic leukemias (ALL) with a striking predominance of V delta 2(D)D delta 3 recombinations in common ALL (cALL) patients. Recently, we and others showed that almost 20% of cALL cases are characterized by further recombination of V delta 2(D)D delta 3 segments to J alpha elements, thereby deleting the TCR delta locus in analogy to the delta Rec/psi J alpha pathway in differentiating alpha/beta-positive T cells. We report here that two human cALL-derived cell lines, REH an

CRISPR-Cas12a-based lateral flow detection of white spot syndrome virus: a dual-target approach for detection of early and latent infection.

White spot syndrome virus (WSSV; family Nimaviridae; taxon species White spot syndrome virus) is a major viral pathogen that poses a significant threat to the global shrimp industry, with early detection being the most effective strategy for disease control. We developed a CRISPR-Cas12a-based dual-target detection assay for WSSV, specifically targeting the VP28 gene (gene product is a major envelope protein) and WSSV366 (a latency-associated gene), optimized using Indian WSSV isolates. Our CRISPR RNAs for both targets had high efficiency, and we evaluated the assay using fluorescence-based and

A generalizable framework for genetic code expansion in bacterial systems beyond E. coli.

Genetic code expansion (GCE) enables the site-specific incorporation of noncanonical amino acids into proteins, providing a powerful platform to investigate and engineer biological functions with molecular precision. While GCE has facilitated transformative studies in Escherichia coli, its utility in bacteria other than E. coli has been hindered by the lack of universal, high-efficiency methods for its implementation and optimization. To address this limitation, we introduce a modular, extensible platform for the generalizable implementation and optimization of GCE in phylogenetically diverse

Development of a one-pot RT-RAA/CRISPR-Cas13a assay for rapid genotyping of Nipah virus in pigs.

INTRODUCTION: Nipah virus (NiV) is a highly pathogenic zoonotic virus transmitted from bats to humans through pigs as a key intermediate host. Given the existence of two distinct NiV genotypes, which differ in clinical manifestations and transmission patterns in both humans and pigs, rapid and sensitive method for detection and genotyping is crucial for effective disease control. Isothermal amplification combined with CRISPR/Cas-based assay provides a promising approach to meet this need. METHODS: Conserved regions were identified by aligning the N gene sequences from 67 NiV strains. Specific

A Novel Conditional Adra2a-Knockout Mouse Line Reveals Cell-specific Contributions to Specific Dimensions of Sedation.

INTRODUCTION: Here, we create a conditional Adra2a line and use it to show that sedative, hypnotic, and hypothermic effects of α 2 -agonists are neuronally mediated via the α 2A adrenergic receptor. METHODS: We generated mice with loxP sites flanking Adra2a using CRISPR/Cas9 gene targeting. This line was crossed with lines encoding Cre recombinase (Cre) under the control of the Vgat, Snap25, and Dbh promoters. Cell-specific knockout was confirmed using fluorescent in-situ hybridization demonstrating targeted reduction in Adra2a mRNA in the appropriate cell types. Mice were given intraperitone

The Branching Projection Pattern of Ventral Subicular Cells Revealing the Complex Integrative Role of Key Hippocampal Outputs in Driving Fear and Anxiety.

The ventral regions of the hippocampus play a crucial role in emotional responses through projections to the hypothalamus. In this context, the ventral hippocampus sends extensive projections to components of the medial hypothalamic defensive circuit, where projections to the anterior hypothalamic nucleus (AHN) influence both anxiogenic and innate fear responses, as well as contextual fear reactions. This study aims to examine the branching projection pattern of hippocampal cells that directly project to the AHN and identify the complete range of brain regions affected by key hippocampal outpu

Complement Activation in Maternal and Placental Pathology of Preeclampsia.

BACKGROUND: Preeclampsia is a multifactorial, pregnancy-related disorder characterized by new-onset hypertension and proteinuria, with distinct early- and late-onset forms. Although complement dysregulation has been implicated in the pathogenesis of preeclampsia, its causal role remains unclear. In mice, Crry (complement receptor 1-related protein Y) functions as a critical complement regulator at the fetal-maternal interface. Because complete Crry deficiency is embryonically lethal, direct in vivo investigation of complement activation in pregnancy has been challenging. METHODS: We generated

Nucleic Acid Amplification Tests for Candida Species Identification: A Systematic Review of Diagnostic Performance.

Rapid and accurate identification of Candida species is critical for guiding antifungal therapy, especially with the emergence of intrinsically resistant pathogens. However, diagnostics using culture-based methods remain slow and labor-intensive, limiting timely treatment decisions. This systematic review evaluated the diagnostic performance and clinical applicability of nucleic acid amplification tests (NAATs) for Candida species identification using a PubMed search completed on 23 June 2025. A total of 888 records were screened, 333 full-text articles were assessed, and 158 studies were incl

CRISPR-AMPED: A CRISPR/Cas-based immunoassay with attomolar sensitivity enabled by magnetic proximity extension and detection.

Clustered regularly interspaced short palindromic repeats (CRISPR)/Cas-associated systems have emerged as powerful tools for next-generation molecular diagnostics, particularly for nucleic acid detection. However, ultrasensitive protein detection is equally critical across diverse applications in biology and medicine, especially for diagnosing and prognosing diseases such as cancer, traumatic brain injury (TBI), Alzheimer's disease, and cardiovascular diseases. Despite recent efforts to adapt CRISPR/Cas systems for protein detection, these methods have typically achieved sensitivity in the fem

A purification-free one-pot CRISPR/Cas13a assay for detection of porcine epidemic diarrhea virus.

Porcine epidemic diarrhea virus (PEDV) causes up to 80-100% mortality in neonatal piglets, yet field surveillance remains constrained by RT-qPCR's dependence on column-based nucleic acid purification and centralized laboratory infrastructure-a bottleneck especially severe in the inhibitor-rich matrices typical of swine clinical samples. Here we report an integrated sample-to-answer methodology coupling a purification-free thermal lysis step with one-pot RT-RPA-CRISPR/Cas13a chemistry, delivering PEDV detection within 35 min. Allogeneic hematopoietic cell transplantation for partial RAG deficiency in children and adults: Excellent outcomes with a reduced-intensity posttransplantation cyclophosphamide-based approach.

BACKGROUND: Partial recombinase activating gene deficiency (pRD) leads to combined immunodeficiency with immune dysregulation. It can be cured by allogeneic hematopoietic cell transplantation (HCT), but optimal referral criteria and approaches remain to be defined. OBJECTIVE: Our study evaluated low-toxicity approaches to HCT for pRD. METHODS: Thirteen children and adults with pRD received radiation-free, predominantly reduced-intensity conditioning (pentostatin/cyclophosphamide/busulfan) HCT with posttransplantation cyclophosphamide-based graft-versus-host disease (GVHD) prophylaxis at medi

Selective genetic targeting of the mouse efferent vestibular nucleus identifies monosynaptic inputs and indicates function as multimodal integrator.

The vestibular system is a critical sensory modality required for coordinated movement, balance, and our ability to interact with the surrounding environment. Vestibular sensory neurons provide the nervous system with information about head rotation and acceleration. However, the nervous system can also modify the activity of sensory neurons and hair cells via the actions of the efferent vestibular system (EVS). The function of the EVS has remained unknown partly because of an inability to target efferent vestibular neurons in a selective manner to understand their synaptic inputs and function

Development and preliminary evaluation of real-time fluorescence-based RPA and RPA-CRISPR/Cas12a assays for rapid detection of Coxiella burnetii.

Coxiella burnetii (C. burnetii) is a ubiquitous zoonotic pathogen that exists widely in nature and is an obligate intracellular parasite of eukaryotic cells, causing Q fever in both humans and animals. Therefore, the surveillance and prevention of Q fever are of great importance for public health and the livestock industry. In this study, specific RPA primers, probes, and crRNAs were designed and synthesized targeting the IS1111 gene of C. burnetii, a real-time fluorescence recombinase polymerase amplification (qRPA) assay and a RPA combined with CRISPR/Cas12a (RPA-CRISPR/Cas12a) assay for the

Field deployment of a mobile suitcase laboratory for Buruli ulcer diagnosis in Ghana.

Molecular diagnostics are the gold standard laboratory confirmation test for Buruli ulcer (BU), a severe necrotising skin disease caused by Mycobacterium ulcerans (M. ulcerans). However, current molecular tests are often performed outside endemic areas, which results in delayed diagnosis and increased patient management costs. To overcome these challenges and facilitate rapid diagnosis of clinically suspected BU lesions in affected communities, we developed a portable laboratory platform contained in two Pelican cases (each measuring 56 cm × 45.5 cm × 26.5 cm). We evaluated the feasibility of
