Synthesis of a novel carbocyclic analog of bredinin.

The natural nucleoside antibiotic, bredinin, exhibits antiviral and other biological activities. While various nucleosides related to bredinin have been synthesized, its carbocyclic analog has remained unknown. Synthesis of this heretofore unknown analog of bredinin is described. The key precursor, (3aS,4R,6R,6aR)-6-((methoxy-methoxy)methyl)-2,2-dimethyltetrahydro-3aH-cyclopenta[d][1,3]dioxol-4-amine (5), was prepared from the commercially available compound, (1R,4S)-2-azabicyclo[2.2.1] hept-5-en-3-one (4). Our initial approach used intermediate 6, derived in three transformations from 5, for

Molecular surveillance of arboviruses in Nigeria.

Arboviral infections are fast becoming a global public health concern as a result of its high fatality rate and sporadic spread. From the ou … t conducting a surveillance for Yellow fever virus, Zika virus, Chikungunya virus, Dengue virus and Rift Valley fever virus in four states in Nigeria using molecular techniques. Controlling mosquitoes with semiochemicals: a review.

The use of semiochemicals in odour-based traps for surveillance and control of vector mosquitoes is deemed a new and viable component for integrated vector management programmes. Over 114 semiochemicals have been identified, yet implementation of these for management of infectious diseases such as malaria, dengue, chikungunya and Rift Valley fever is still a major challenge. The difficulties arise due to variation in how different mosquito species respond to not only single chemical compounds but also complex chemical blends. Additionally, mosquitoes respond to different volatile blends when t…

Epidemiological situation of transboundary animal diseases in North African countries-proposition of a regional control strategy.

The Food and Agricultural Organization (FAO) defined transboundary animal diseases (TADs) as those that are of significant economic, trade, … mouth disease, peste des petites ruminants, bluetongue, sheep/goats pox, brucellosis, West Nile and Rift Valley fever, as the major TADs in this region, while the major constraints identified were illegal animal movement, communal clashes, unreported outbreaks, poor vaccination coverage, and other factors peculiar to each etiology. Therefore, the establishment of e …

Comprehensive Analysis of Interaction Networks of Telomerase Reverse Transcriptase with Multiple Bioinformatic Approaches: Deep Mining the Potential Functions of Telomere and Telomerase.

Telomerase reverse transcriptase (TERT) is the protein component of telomerase complex. Evidence has accumulated showing that the nontelomer … TERT might play nontelomeric roles in the generation or development of some rare diseases, such as Rift Valley fever and dyscalculia. Thus, our data will help to decipher the interaction networks of TERT and reveal the unknown functions of telomerase in cancer and aging-related diseases.

Crystal Structure of Glycoprotein C from a Hantavirus in the Post-fusion Conformation.

Hantaviruses are important emerging human pathogens and are the causative agents of serious diseases in humans with high mortality rates. Like other members in the Bunyaviridae family their M segment encodes two glycoproteins, GN and GC, which are responsible for the early events of infection. Hantaviruses deliver their tripartite genome into the cytoplasm by fusion of the viral and endosomal membranes in response to the reduced pH of the endosome. Unlike phleboviruses (e.g. Rift valley fever virus), that have an icosahedral glycoprotein envelope, hantaviruses display a pleomorphic virion morp…

Identification of phlebovirus and arenavirus RNA sequences that stall and repress the exoribonuclease XRN1.

Regulated mRNA decay plays a vital role in determining both the level and quality of cellular gene expression. Viral RNAs must successfully … s of biochemical assays to demonstrate that the 3'-terminal portion of the nucleocapsid (N) mRNA of Rift Valley fever virus, a phlebovirus of the Bunyaviridae family, also can effectively stall and repress XRN1. The region responsible for impeding XRN1 includes a G-rich portion that likely forms a G-quadruplex structure. The 3'-terminal portions of ambisense-derive …

Hantavirus Gn and Gc envelope glycoproteins: key structural units for virus cell entry and virus assembly.

In recent years, ultrastructural studies of viral surface spikes from three different genera within the Bunyaviridae family have revealed a … een predicted for Gc of hantaviruses, and novel crystallographic data confirmed such a fold for the Rift Valley fever virus (RVFV) Gc protein. During virus cell exit, the assembly of different viral components seems to be established by interaction of Gn and Gc cytoplasmic tails (CT) with internal viral ribonucleocapsids. Moreover, recent findings show that hantavi …

Capripoxvirus-vectored vaccines against livestock diseases in Africa.

Five different viral diseases of livestock, lumpy skin disease (LSD), sheep pox (SPP), goat pox (GTP), Rift Valley fever (RVF) and peste des petits ruminants (PPR), circulate in the same regions of Africa, imposing a major burden on economic activity and public health. While commercial vaccines against these viruses are available, the cost of implementing regular vaccination regimens against multiple diseases is prohibitive for most African farmers. A single, affordable multivalent vaccine that simultaneously protects against all 5 diseases would therefore be of significant benefit to the live…

Host-feeding patterns of Culex pipiens and Cx. antennatus (Diptera: Culicidae) from a village in Sharqiya Governorate, Egypt.

Host-selection patterns of mosquitoes were determined over a 1-yr period at Abu Heif, a village in Sharqiya Governorate, Egypt, that has had a history of Rift Valley fever virus (RVFV) transmission. Culex pipiens L. and Cx. antennatus Becker were the most common mosquito species collected, and 8,252 blood meals from both species were analyzed by a precipitin technique. Host availability was estimated by a monthly census of human and animal populations. Both mosquito species exhibited opportunistic endophagic behavior. In bedrooms, 79% of Cx. pipiens fed on humans, compared with 53% of the Cx. …

Climate-resilient livestock systems in Sub-Saharan Africa: a one health systems framework integrating pastoralism, animal health, and sustainable development.

Pastoral livestock systems in Sub-Saharan Africa face accelerating climate threats that compromise food security, animal health, and human well-being across millions of rural households. Rising temperatures, recurrent droughts, erratic rainfall, and floods degrade rangeland productivity, restrict pastoral mobility, and reshape disease ecology, driving outbreaks of trypanosomiasis, tick-borne infections, and Rift Valley fever. These biophysical pressures interact with governance failures, farmer-herder conflict, and weak veterinary infrastructure to generate compound vulnerabilities that extend…

Smartphone AI-Enabled Lateral Flow Immunoassay Platform Using Advanced Quantum Dots for Intelligent Quantitative Diagnostics.

Lateral flow immunoassays (LFIAs) are widely used in point-of-care diagnostics for their low cost and simplicity, but conventional formats rely on subjective visual readout and lack quantitative accuracy and sensitivity. Here, we present a smartphone-guided, artificial-intelligence-enhanced LFIA platform that unites a rationally engineered dual-modal nanoprobe with on-device machine learning, demonstrated for quantitative detection of Rift Valley fever virus (RVFV). The platform combines three synergistic principles: advanced magnetic quantum dot nanoparticles (AQDs) that integrate a magnetic …
